Goals: The aim of this study was to determine the antibiotic resistance profile of serotypes of Streptococcus pneumoniae strains circulating in Bangui. Methodology: A prospective and analytical analysis was carried out at the National Laboratory of Clinical Biology and Public Health from 2017 to 2022. The strains came from our study on the contribution to the study of antibiotic sensitivity of Streptococcus pneumoniae strains. The multiplex PCR test was used for its cost-effectiveness in terms of amplifiers which can be purified in order to be sequenced. It also makes it possible to detect several germs as well as their serotypes. For a PCR reaction, several elements are involved in the reaction medium or Master Mix. These are the desoxyribonucleotides (dNTPs), the magnesium ions (MgCl2) and the primers. A set of 14 primers divided into 3 classes were used. Class 1 primers served as an internal control by targeting the cpsA gene. It is a highly conserved gene found in capsular loci characterized to date. The primers of the second class were used to target specific serotypes by specific reactions (out of six possibilities). The group reaction was carried out using the primers of the third class in order to carry out an initial screening of the samples and to classify the pneumococcal isolates. Related serotypes were grouped based on the amplification of common genes. Using the technique of electrophoresis on agarose gel and an ultraviolet radiation device, the migration bands are then visualized and analyzed. The data collected had been entered into Excel 2010 and analyzed with Epi info 7. The exact Fischer chi2 test at the 5% threshold, the relative risk and its 95% confidence interval were used to compare the proportions and determine the associations. Results: 187 antibiotic-resistant strains of Streptococcus pneumoniae were collected. The average frequency of serotypes 1, 9A, 4 and untypeable identified were 43.59%, 18.18%, 18.27% and 39.57% respectively. The frequency of serotype 1 was predominant for the age

Background: Since 2021, high-risk Human Papilloma Virus (HR-HPV) testing has been the recommended screening test for cervical cancer for all settings;either used alone in a “test and treat” strategy, or with a triage test, with or without biopsy, before treatment. Cameroon has rolled out immunization against HPV 16 and 18, but studies show a higher prevalence of non-16/18 HR-HPV types. Objectives: Determine the prevalence of precancerous lesions, in women with HR-HPV infection and evaluate association of digital cervicography (DC) VIA/VILI positivity with HPV serotype, as a measure of their contribution to precancer and cancer incidence. Methodology: The study was cross-sectional, descriptive, and analytic. It took place at the Etoug-Ebe and Ekoudoum Baptist Hospitals in Yaoundé, during the period April-September 2022. We reviewed the records of women screened for cervical cancer between February 2020 and December 2021 and evaluated the prevalence of lesions on digital cervicography (DC) with VIA/VILI for women positive for HR-HPV serotypes. The data were analyzed using SPSS version 20.0 for Windows. P values Results: We identified 315 cases with a positive HR-HPV deoxyribonucleic acid (DNA) test, 224 (71.1%) had a DC VIA/VILI triage test done. Of these, 30 (13.4%) women had a positive DC VIA/VILI, with five women (2.2%) having lesions suggestive of cancer. Out of 11 cases positive for HPV 16 alone, 05 (45.5%) had a positive DC VIA/VILI test. Of the 14 cases positive for HPV 18 alone, 03 (21.4%) had a positive VIA/VILI, meanwhile only 19 (10.7%) of the 177 cases positive for non-16/18 HPV had a positive VIA/VILI test. Conclusion: A high proportion of women (13.4%) with HR HPV had a positive DC VIA/VILI, with a significant proportion (2.2%) having lesions suggestive of invasive cervical cancer HR-HPV serotype was associated with DC VIA/VILI positivity;HPV 16 had the strongest association (45.5%), followed by HPV 18 (21.4%), and non-16/18 HR-HPV (10.7%), suggesting a decreasing order of oncogenicity.

A rapid, simple, and sensitive immunoagglutination assay with silica nanoparticles for serotype identification of Pseudomonas aeruginosa

An agglutination test based on colored silica nanoparticles(colored SiNps) was established to detect serotypes of Pseudomonas aeruginosa. Monodisperse colored SiNps were used as agglutination test carriers. The colored SiNps were prepared through reverse microemulsion with reactive dyes, sensitized with 11 kinds of mono-specific antibodies against P. aeruginosa, and denoted as IgG-colored SiNps. Eleven kinds of IgG-colored SiNps were individually mixed with P. aeruginosa on a glass slide.Different serotypes of P. aeruginosa could be identified by agglutination test with evident agglutination. The P. aeruginosa could be detected in a range from 3.6×10^5 to 3.6×10^12 cfu mL^–1. This new agglutination test was confirmed to be a specific,sensitive, fast, easy-to-perform, and cost-efficient tool for the routine diagnosis of P. aeruginosa.

Dengue outbreaks in Taiwan, 1998-2017: Importation, serotype and temporal pattern

Objective: To ascertain the role of imported cases and serotypes on dengue outbreaks in Taiwan which have been sporadic yet highly volatile during the past two decades, exhibiting record-breaking magnitude in recent years. Methods: Confirmed case and serotype data from Taiwan Centers for Disease Control during 1998-2017 were fully examined, with fitting of weekly and daily case data of each city/county to a mathematical model to pinpoint the waves of cases and their locations. Moreover, we quantify the timing of turning point and transmission potential of each wave and determine its circulating serotype, to ascertain any pattern or connection between the variations in circulating serotypes and the magnitude/transmissibility of outbreak. Results: While the number of imported case increased steadily during past two decades, the yearly number of indigenous cases fluctuated wildly. Moreover, while yearly percentages of serotypes for imported cases remains steady, that of indigenous cases does not exhibit any clear pattern. There was at least one wave of reported cases somewhere in Taiwan every year from 1998 to 2015, except in 2016-2017. The effective reproduction number R for all waves in all locations ranged from 1.14 to 2.87, with the exception of two Tainan waves, in 2010(3.95) and 2015(6.84). Four major outbreaks of over 2000 cases reveal circulation of one dominant serotype. Conclusions: Correlation between imported cases and indigenous outbreak prove to be difficult to ascertain, even with the availability of serotype data. However, although there had been occasional co-circulation of serotypes in one location, and for some years with different serotypes circulating in different locations, all major outbreaks of over 2 000 cases during the past two decades are due to circulation of mainly a single serotype, perhaps indicating greater transmission potential with one dominating serotype.

Acinetobacter baumannii has three clusters of genes for polysaccharide synthesis.Capsular polysaccharides and lipopolysaccharides are major virulence determinants in many Gram-negative bacteria,and play key roles in the overall fitness and virulence in A.baumannii,meanwhile poly-beta-1-6-N-acetylglucosamine was critical for biofilm formation.A variety of studies have been conducted which focused on the surveillance of antimicrobial resistance genes and clones in A.baumannii.However,a tool to perform serotype prediction in A.baumannii is still lacking.Here we present Bautype,a tool for whole genome sequencingbased K and O locus typing in A.baumannii.Using Bautype,users can upload and analyze the genome assembled in a web browser,with the results being displayed in a tabular or graphical format.We demonstrated the high accuracy of predicting K and O locus diversity using Bautype when analyzing more than 3000 A.baumannii genomes.Using our software,we were able to analyze K and O locus diversity among 593 genomes belonging to ST208(3 K and 8 O).These diverse loci can serve as epidemiological markers for Multi-drug Resistance(MDR)strain subtyping.We also identified 45 novel O locus types.Our findings highlight the importance in monitoring the capsule type and LOS of A.baumannii strains for which Bautype is a tailored clinical software.Bautype is available at http://***.

Genetic diversity of Listeria monocytogenes isolates from farm products in Shanghai,China

Farm products are considered important vehicles for the transmission of Listeria monocytogenes(L.monocytogenes).The typing of L.monocytogenes from farm products contributes to the surveillance and source tracing of the pathogen.In this study,77 L.monocytogenes strains from seven farm product categories in Shanghai were analyzed by serological typing,multilocus sequence typing(MLST),pulsed-field gel electrophoresis(PFGE),and whole-genome nucleotide polymorphism(wgSNP).The results showed that the 77 isolates were classified into four serovars(1/2a,1/2c,1/2b,and 4b),and of these,1/2a(n=47)was the most prevalent.Seventeen sequence types(STs)were generated by MLST with two novel STs(ST1402 and ST1403),and 20 of 77 L.monocytogenes isolates had high genetic identity with previously documented outbreak isolates according to the minimum spanning tree from the MLST results.Moreover,34 PFGE patterns(PF1-PF34)were differentiated,and based on a similarity value higher than 80%by the unweighted pair group method dendrogram,the discriminatory index was relatively low(equal to 0.775).Furthermore,14 isolates were chosen and further analyzed by wgsNP based on the previous typing results,which demonstrated that wgSNP and MLST yielded mostly consistent typing results but higher resolution than PFGE.In conclusion,77 L.monocytogenes isolates from farm products collected in nine districts in Shanghai were highly genetically diverse,and 20 of these isolates had high relatedness with previously documented outbreak strains worldwide.The results indicate a possible cross-contamination risk of L.monocytogenes and a potential public health concern resulting from farm products during the supply chain in Shanghai,China.

and pili genes are also investigated.Methods:This multicentre,prospective,observational study is conducted in seven major tertiary hospitals in Malaysia among non-pregnant adults.Simultaneously,a retrospective study is conducted in the selected hospitals with similar approaches.GBS isolates are subjected to phenotyping,serotyping by multiplex PCR,antimicrobial susceptibility testing and PCR-detection of GBS virulence and pilus genes.Seven housekeeping genes are amplified and sequenced for multi-locus sequence typing.Discussion:Findings from the study may contribute to the management of clinical practice to diagnose and prevent GBS related diseases in a timely manner.Prudent use of antibiotics is encouraged by monitoring antimicrobial resistance.

Distribution and Prevalence of serotypes of Group B Streptococcus Isolated from Pregnant Women in 30 Countries:A Systematic Review

Objective:This review aimed to compile scientific data on the distribution and prevalence of group BStreptococcus (GBS) serotypes isolated from pregnant women across 30 countries from 2010 to 2019.Methods:This was a systematic review that addresses the distribution and prevalence of GBS in pregnant women.The search included studies published between January 2010 and December 2019 in PubMed,Virtual Health Library (BVS),ScienceDirect,Scientific Electronic Library Online (SciELO),and LILACS databases.We also surveyed relevant articles published in English,Spanish,and Portuguese between February and April 2020.Original articles,communication,short report,theses,and dissertations were included.The prevalence of GBS colonization,method for capsular serotyping,antimicrobial resistance,distribution and prevalence of serotypes were extracted from each study.Results:In all,795 publications were identified.After applying the eligibility criteria,48 articles were included for the final systematic analysis;most articles were from Asia and were published during the years 2014 to 2017.For the identification of serotypes,most studies used the polymerase chain reaction technique.There were records of all 10 GBS serotypes,namely,Ia,Ib,and II–IX,among the countries analyzed.GBS susceptibility and resistance to antibiotics were addressed in 37.5% of the publications analysed.Conclusion:This review showed that GBS serotypes are distributed differently in the 30 analyzed countries,with serotypes Ia,Ib,and II to V being the most prevalent.Furthermore,our results highlighted the relationship of GBS with maternal colonization,implications for neonates,and antibiotic resistance.

Molecular Profiles and Antimicrobial Susceptibility of First Isolates of <i>Streptococcus agalactiae</i>serotype IX in Argentina

Streptococcus agalactiae (GBS) is considered as the most important cause of invasive bacterial disease in infants. There are few current data from serotype IX of GBS worldwide. The present work has been done in order to make a contribution to the knowledge of serotype IX through phenotypic and genotypic study of virulence and resistance. Of 200 strains tested, 5.5% were serotype IX and all were colonizing. In 63.6% of the strains the presence of the bac, bca and hylB genes was determined, and in 54.5% of lmb and rib. All strains were susceptible to Clindamycin and Erythromycin however five isolates showed the resistance genes: ermB, ermTR and/or mefA. The presence of serotype IX in Misiones, a province situated in the northeaster of Argentina, which limits with Paraguay and Brazil in South America, gives the region a particular situation. Currently, public health efforts are aimed at prevention and treatment, study of the virulence mechanisms, surveillance of resistance to antibiotics and the development of effective vaccines to prevent GBS infection.

A Case of Carbapenem Resistant Non-K1/K2 serotype <i>Klebsiella pneumoniae</i>Liver Abscess

Klebsiella pneumoniae liver abscess (KPLA) has been described as an invasive syndrome with extrahepatic complications. The majority of KPLA is caused by capsular serotype K1 and K2 isolates. We report a case of carbapenem resistant Klebsiella pneumoniae liver abscess. The patient initially presented with infected right above-the-knee amputation and was later found with a large liver abscess. Initial antimicrobial susceptibility showed carbapenem resistant K. pneumoniae (CRKP). Further molecular workup revealed that the isolate was a less virulent non-K1/K2 serotype, and both rmpA and kfu genes were negative. The lack of outer membrane porins likely contributed to the carbapenem resistance. To our knowledge, this is a first reported case of carbapenem resistant, non-K1/K2 serotype K. pneumoniae liver abscess in the United States.
