More substances leaked from a higher-vigor seed sample than from a lower-vigor sample. This indicates that, in some cases, electric conductivity does not represent seed vigor level very well, especially for high-vigor seeds. Results from germination, germination index, leachate conductivity, and the ratio of K^＋/Na^＋ from three-seed lots of Chinese cabbage （Brassica pekinensis （Louv.） Rupr） showed that K^＋/Na^＋ correlated well with germination and germination index. The ability of K^＋/Na^＋ to indicate well changes in vigor was further supported by investigation in soybean （Glycine max （L.） Merr.） seeds and another cultivar of Chinese cabbage seeds. Thus, seed leakage of K^＋/Na^＋ can accurately indicate seed vigor, whereas the conductivity test failed to do so. Furthermore, K^＋/Na^＋ showed up bigger quantitative differences in vigor level than did the conductivity test. This findings provide a more sensitive and accurate index for the assessment of seed vigor. The mechanisms of Na^＋ and K^＋ ion transport are also discussed.

seed vigor is an important characteristic of seed quality,and rice cultivars with strong seed vigor are desirable in direct-sowing rice production for optimum stand *** the present study,the quantitative trait loci (QTLs) of three traits for rice seed vigor during the germination stage,including germination rate,final germination percentage,and germination index,were investigated using one recombinant inbred line (RIL) population derived from a cross between japonica Daguandao and indica IR28,and using the multiple interval mapping (MIM) *** results show that indica rice presented stronger seed vigor during the germination stage than japonica rice.A total of ten QTLs,and at least five novel alleles,were detected to control rice seed vigor,and the amount of variation (R2) explained by an individual QTL ranged from 7.5% to 68.5%,with three major QTLs with R2>20%.Most of the QTLs detected here are likely to coincide with QTLs for seed weight,seed size,or seed dormancy,suggesting that the rice seed vigor might be correlated with seed weight,seed size,and seed *** least five QTLs are novel alleles with no previous reports of seed vigor genes in rice,and those major or minor QTLs could be used to significantly improve the seed vigor by marker-assisted selection (MAS) in rice.

Raffinose family oligosaccharides （RFOs） accumulate in seeds during maturation desiccation in many plant species. However, it remains unclear whether RFOs have a role in establishing seed vigor. GALACTINOL SYNTHASE （GOLS）, RAFFINOSE SYNTHASE （RS）, and STACHYOSE SYNTHASE （STS） are the enzymes responsible for RFO biosynthesis in plants. Interestingly, only raffinose is detected in maize seeds, and a unique maize RS gene （ZmRS） was identified. In this study, we found that two independent mutator （Mu）-interrupted zmrs lines, containing no raffinose but hyperaccumulating galactinol, have significantly reduced seed vigor, compared with null segregant controls. Unlike maize, Arabidopsis thaliana seeds contain several RFOs （raffinose, stachyose, and verbascose）. Manipulation of A. thaliana RFO content by overexpressing ZmGOLS2, ZmRS, or AtSTS demonstrated that co-overexpression of ZmGOLS2 and ZmRS, or overexpression of ZmGOLS2 alone, significantly increased the total content of RFOs and enhanced Arabidopsis seed vigor. Surprisingly, while overexpression of ZmRS increased seed raffinose content, its overexpression dramatically decreased seed vigor and reduced the seed amounts of galactinol, stachyose, and verbascose. In contrast, the atrs5 mutant seeds are similar to those of the wild type with regard to seed vigor and RFO content, except for stachyose, which accumulated in atrs5 seeds. Total RFOs, RFO/sucrose ratio, but not absolute individual RFO amounts, positively correlated with A. thaliana seed vigor, to which stachyose and verbascose contribute more than raffinose. Taken together, these re- sults provide new insights into regulatory mechanisms of seed vigor and reveal distinct requirement for RFOs in modulating seed vigor in a monocot and a dicot.

seed vigor is an index of seed quality that is used to describe the rapid and uniform germination and the establish- ment of strong seedlings in any environmental conditions. Strong seed vigor in low-temperature germination conditions is particularly important in direct-sowing rice production systems. However, seed vigor has not been selected as an important breeding trait in traditional breeding programs due to its quantitative inherence. In this study, we identified and mapped eight quantitative trait loci （QTLs） for seed vigor by using a recombinant inbred population from a cross between rice （Oryza sativa L. ssp. indica） cultivars ZS97 and MH63. Conditional QTL analysis identified qSV-1, qSV-Sb, qSV-6a, qSV- 6b, and qSV-11 influenced seedling establishment and that qSV- 5a, qSV-Sc, and qSV-8 influenced only germination. Of these, qSV-1, qSV-Sb, qSV-6a, qSV-6b, and qSV-8 were low-tempera- ture-specific QTLs. Two major-effective QTLs, qSV-1, and qSV-5cwere narrowed down to 1.13-Mbp and 4oo-kbp genomic regions, respectively. The results provide tightly linked DNA markers for the marker-assistant pyramiding of multiple positive alleles for increased low-temperature germination seed vigor in both normal and environments.

Oxygen sensing technology was employed to study the rapid methods for seed vigor assessment of Chinese fir （Cunninghamia lanceolata） and Masson pine （Pinus massoniana）. Firstly, seeds of five lots were performed using accelerated aging （AA） into three vigor levels. Then, four oxygen sensing indices, including increased metabolism time （IMT）, oxygen metabolism rate （OMR）, critical oxygen pressure （COP）, relative germination time （RGT） and the control indiees such as labora- tory germination indices, dehydrogenase activity （DA）, and electrical conductivity （EC） were analyzed by the tests of 15 samples. The results of correlation analysis between these indices and field emergence per- formances based on two-year and two-spot data showed that RGT and OMR should be indicated as the optimal oxygen sensing indices to rap- idly and automatically evaluate seed vigor of Chinese fir and Masson pine, respectively. On the basis, one-variable linear regression equations were built to forecast their field emergence performances by the two oxygen sensing indices.

seed vigor is a more promising seed quality character reflecting potential seed germination, field emergence and seed storage ability under different conditions than standard germination. Standard germination is influenced by genetic background and environmental effects during seed development and storage conditions. The latest research on physiological mechanism in seed vigor showed that at the late stage of seed development, the development of seed vigor involves some stress-resistant substances including late embryogenesis abundant （LEA） protein, oligosaccharides and abscisic acid （ABA）. Whereas the loss of seed vigor, or seed aging and deterioration, could be attributed to lipid peroxidation, chromosome deformation and genes aberrance, and embryo protein degradation, etc. seed vigor is a quantitative character controlled by multi-genes. Genetic and quantitative trait locus （QTL） analyses on seed-vigor trait in model plants, such as Arabidopsis and rice, are mostly concentrated on related morphology traits, whereas few physiological traits have been researched. It was concluded that, all of the QTL genetic characteristics of seed vigorincluding QTL quantities, its locus on chromosome, genetic effects, and interaction effects between genetic and environment, differed with plant species and used seed-vigor traits.

By assay of accelerated aging germination, germination index, vigor index, seedling length, seedling weight, electric conductivity, dehydrogenase activity, ATP content and acid phosphoesterase activity during seed development of turf-type tall fescue, the seed vigor of tall fescue were studied. Combining with seed moisture content and yield, the optimal harvesting time of tall fescue was determined. The results indicated that the seed vigor increased continuously along with seed maturity, and the higher seed vigor was achieved at 19th day after perk anthesis and maintained continuously until 31st day after peak anthesis. At 25th day after peak anthesis, the highest yield of 3 533 kg ha-1 and the good quality of seeds of tall fescue were harvested with 32.19% moisture content. From 22nd to 31st day after peak anthesis, the seed yield of 3 300 kg ha-1 and the good quality seeds of tall fescue were harvested with 40 - 12. 43% moisture content, and the span was the optimal harvesting time.
