A method based on UV-Vis spectrometry（UV） and vertex component analysis（VCA） was proposed to characterize the processing batch for preparation of prepared radix scutellariae using water steamingIn the proposed method, UV spectra of the radix scutellariae that were collected at different intervals of the processing procedure were measured, then the measured UV spectra of mixtures were resolved by VCA, and the estimated vertex components（VCs） and their variation of relative concentration were used to characterize the processing batchThe results show that estimated VCs have chemical significance with spectral characteristics of source signal when the source spectra are overlappedThe variation of the relative VCs’ concentrations can explain the physical and chemical changes of active components during the processing batchThe endpoint of the processing batch was determined as 35 min, which is more accurate and overcomes the shortcomings that different endpoints may be made by sensory analysis of different operatorsthe endpoint of the process batch that was determined by inspection of the variation of VCsUV-VCA method provides an alternative approach for characterization of the traditional Chinese medicines processing procedure.

Flavonoid profiling of a traditional Chinese medicine formula of Huangqin Tang using high performance liquid chromatography

The quality control processes for herbal medicines have been problematic. Flavonoids are the major active components of Huangqin Tang(HQT, a traditional Chinese medicine formula). In this study, we used a combinative method approach consisting of chromatographic fingerprinting(high performance liquid chromatography; HPLC), quantitative methods and a pharmacodynamic evaluation model to analyze the flavonoids of HQT obtained from different sources. Ten batches of HQT were analyzed by the HPLC fingerprinting method and 26 common peaks were detected, of which 23 peaks corresponded with the chemical profile of HQT. In addition, 11 major compounds were identified by LC–MS analysis(liquid chromatography–tandem mass spectrometer; LC–MS^n) and quantified by the HPLC quantitative method approach. The studied10 batches of HQT were found to be homogeneous in their composition with a similarity between 0.990 and1.000. The distribution of the 11 identified compounds was found to be very similar among the batches. Only slight pharmacodynamic differences were detected between the different batches, confirming the homogeneity of HQT. The results of this study prove that the combination of chromatographic fingerprinting and quantitative analysis can be readily used for comprehensive quality control of herbal medicines.

A synopsis on flavonoids from the roots of Scutellaria baicalensis with some insights on baicalein and its anti-cancer properties

Flavones are the most dominant type of flavonoids isolated from the roots of Scutellaria baicalensis(Radix scutellariae), which is a traditional medicinal plant in East Asian countries, including China, Japan and *** of the flavones are derivatives with methoxyl and hydroxyl groups, and they include baicalein, baicalin, chrysin, norwogonin, oroxylin A and *** possesses anti-cancer activities against a wide spectrum of human cancer cells by inducing apoptosis and cell cycle arrest, and by inhibiting angiogenesis, metastasis and *** examples of the effects of baicalein on apoptosis, cell cycle arrest and metastasis are presented with discussion on the molecular targets and *** on the structure-activity relationships of flavonoid cytotoxicity towards human cancer cells show that the potent cytotoxic activities of baicalein can be attributed to its-OH groupsat C5, C6 and C7(triple hydroxylation) of ring A, carbonyl group at C4 of ring C, and C2–C3 double bond of ring *** on structural modifications of baicalein have shown that the configurations at C6 of ring A are critical factors influencing its anti-proliferative *** the remarkable anti-cancer properties, the future prospects for developing baicalein into an anti-cancer drug are promising.
