＜正＞Soil born disease including root knot nematodes（Meloidogyne spp.） caused severe economic loss in Plastic *** experiment was carried out to control these disease using High Temperature *** temperature steam（HTS） was pumped up into the under-ground（depth 50cm） through pored plastic pipe（diameter 10cm） in plastic *** temperature was reached over 80t after one hour treatment with *** density of microorganism in the soil including fungi,actinomycetes and bacteria was significantly decreased by HTS treatment compared to non-treated *** HTS treatment,the average population of root knot nematodes was significantly reduced from 1 570 heads to 0（Head/300g soil）.The withering rates of oriental melon by root knot nematodes was 5.2%in HTS and 10.4%in chemical fumigation（dazomet） treatment but 60.2%was withered in control in middle stage of growth at 150 days after *** effect of HTS treatment was as effective as dazomet fumigation ***,we concluded that HTS can be used as an environment-friendly control method,alternate to chemical fumigation for the control of Soil born disease in plastic house.

The cereal cyst nematode(CCN) is worldwide and caused the yield losses of wheat ranging from 10%to 40%.For the short of resistance resource in wheat,it’s in urgent need of exploring and breeding new resistance variety from sibling *** the Transcriptome sequence to the Aegilops varabilis NO.1 infected by cereal cyst nematode,the transcriptome data indicates Phenylalanine metabolism and Iso-quinoline alkaloid metabolism plays an important role in the defense responses to the early nematode infectioa The candidate TDC(tryptophan decarboxylase) gene is the key gene of these two metabolic *** VIGS technology to silence TDC1 gene of the Aegilops varabilis NO.1,second stage juveniles(CCN) were used to infect silenced ***-time PCR data indicated that the expression of this gene already reduce 60%,is significantly lower than control group;CCN infection is more than the control ***,expression vectors containing AeVTDC1 was constructed and used for transformation of tobacco(Nicotiana tabacum) by *** positive plants were selected by PCR with specific primers and southern *** order to test resistance to root-knot nematode,the transgenic and wild-type tobaccos were cultivated in soil with root-knot *** analysis was used to measure the level of tryptamine in roots of AeVTDC1 transgenic *** in 6 AeVTDC1 transgenic tobaccos have been obtained through PCR and southern blot *** containing AeVTDC1 showed more resistant to root knot nematodes than wild ***-MS analysis of tryptamine was operated in transgenic and wild-type tobaccos,and the results showed that the level of tryptamine in transgenic tobaccos was much higher than that of wild-type *** preliminary results indicated that TDC plays a positive role in resistance to cereal cyst nematode and root knot nematodes in Aegilops varabilis NO.1.
