Background Nontuberculous mycobacterium （NTM） had been reported to cause cutaneous infections which are difficult to interpret due to the variability of the clinical manifestations. Among NTM infections, Mycobacterium marinum （M. marinum） are mostly seen to cause skin infection. It is therefore important to establish a rapid approach for detection and identification of M. marinum from lesions of patients with suspected M. marinum infections. Methods Specimens were obtained from 5 patients with swimming pool granuloma. DNA was extracted and polymerase chain reaction （PCR） was performed. PCR products were digested with Hae III and BstE II, then analysed by pattern restriction analysis to detect heat shock protein （hsp） 65 kD gene. Results The 65 kD hsp gene was found in all specimens from patients with swimming pool granuloma. PCR restriction analysis （PRA） identified all 5 samples to be M. marinum infections, and the result was consistent with that of routine bacteriological identification. The lesions subsided or markedly improved upon treatment. Conclusions PRA is a sensitive, specific and rapid method in identification of mycobacteria. Application of this method will be helpful for early diagnosis of mycobacterial skin infections.

Background: An experiment was conducted to evaluate the effects of feed restriction(FR) and sex on the quantitative and qualitative carcass traits of Morada Nova lambs. Thirty-five animals with an initial body weight of 14.5 ± 0.89 kg and age of 120 d were used in a completely randomized study with a 3 × 3 factorial scheme consisting of three sexes(11 entire males, 12 castrated males and 12 females) and three levels of feeding(ad libitum – AL and 30% and 60% FR).Results: Entire males presented greater hot and cold carcass weights(P 0.05) concentrations, and the concentrations of palmitic acid, palmitoleic acid, stearic acid, oleic acid and conjugated linoleic acid were higher(P restriction.

A new heteromorphismof C banded region was discovered in chromosome 4. PstⅠinduced G-like banding pattern.The duration of enzymatic digestion the concentration of glycerol in the reaction bufferas well as the ageing and heating of chromosome preparations all had a significant influ-ence on the banding effects of the REs.

Jeep problem is a kind of model of logistics in extreme situation, which has application in exploration and aircraft problems. The optimal distance and driving strategy of multiple jeeps problem are known. We consider multiple jeeps problem with container restriction, which is more complicated in the proof of feasibility and optimality of a driving strategy. We investigate when it can achieve the same optimal distance as without restriction.Based on the non-restricted optimal distance, a new driving strategy is proposed. We provide the necessary and sufficient condition to ensure the feasibility of the strategy, and obtain the maximal feasible distance.

In 1991, we found that methylation outside the PvuⅡ recognition sequence could partially inhibit its cleavage activity. To clarify the molecular mechanism, three plasmids with different methylation states were constructed. Then, together with the original one, four plasmids were digested with different amounts of PvuⅡ. Results show that methylation on both sites results in 90% inhibition; moving the methylated site one base further away decreases the inhibitory effect to about 30%; with the adjacent dam methylation site eliminated, the inhibitory effect disappears. The data suggest that the inhibition of cleavage activity caused by outside methylation is not "all or none", and the degree of inhibition is dependent on the position and the number of methylated bases.

Numerical study on the restriction speed of train passing curved rail in cross wind

The results of numerical investigations of aerodynamic forces and moment coefficients of flow passing a simplified train geometry under different wind speeds are summarized. To compute numerically the different coefficients, the three-dimensional Reynolds-Averaged Navier-Stokes (RANS) equations, combined with the k-ε turbulence model, were solved using finite volume technique. The pres-sure-velocity fields were coupled using the SIMPLE algorithm. At each iteration the pressure correction was obtained by solving a velocity divergence-derived Poisson-like equation. With the computed aerodynamic forces, the formula of the restriction speed at which the train passed curved rail in cross wind was deduced to analyse the influences of aerodynamic forces on the restriction speed. Results of numerical investigations showed that aerodynamic lift and overturn moment increased more and more rapidly with train speed and wind speed. The enhancement trends showed nonlinear phenomena and enhanced risk in the course of train movement. When the train travels at a high speed and encounters a huge cross wind, the influence involved by nonlinear risk increment will extremely impair safety of train. The following conclusion can also be drawn: The effect of aerodynamic lift makes restriction speed reduce, however, the influences of aerodynamic drag to the limit train speed rest on the direction of wind flow. When the wind blows from inner rail to outer rail, aerodynamic forces shall reduce the restriction speed, by contraries, when the wind blows from outer rail to inner rail, aerodynamic forces shall increase the restriction speed.

Background: Food is an important environmental factor that affects animals' energy metabolism and food shortage has significant effects on animals' behavior, physiology and biochemistry. However, to date few studies have focused on the thermogenesis and its effects on the body condition of birds. In this study, we examined the effects of food restriction on the body mass, basal metabolic rate(BMR) and body composition, and several physiological, biochemical and molecular markers potentially related to thermogenesis, in the Chinese Bulbul(Pycnonotus sinensis).Methods: Birds in the control group were provided with food ad libitum whereas those in the food restriction group were provided with one-half of the usual quantity of food for 12 days. Oxygen consumption was measured using an open-circuit respirometry system. Mitochondrial state 4 respiration and cytochrome c oxidase(COX) activity in the liver and pectoral muscle were measured with a Clark electrode. Avian uncoupling protein(avUCP) mRNA expression was determined in pectorals muscle with quantitative Real-time PCR.Results:Chinese Bulbuls in food restriction group decreased in body mass,BMR and internal organ(heart,kidneys,small intestine and total digestive tract)mass compared with the control group over the 12-day period of food restriction.Bulbuls in the food restriction group also had lower levels of state-4 respiration,COX activity in the liver and muscle,and mitochondrial avUCP gene expression in muscle compared to the control group.BMR was positively correlated with body mass,state 4 respiration in the liver and COX activity in the muscle.Conclusions:Our data indicate that Chinese Bulbuls not only sustain food shortage through simple passive mechanisms,such as reducing body and organ mass and energy expenditure,but also by reducing energetic metabolism in the liver and muscle.

The restriction width of carcass by the belts( RWCB) as an important parameter of radial tire design has been neglected for a long time. In order to improve the accuracy and efficiency of tire profile design,the calculating method of RWCB is proposed. The equilibrium profile is calculated by geometric model and variational approach,based on it,the predicted model of RWCB is developed for tire design. Finally,four different designs of 12R22.5 tires are investigated by experiment and finite element method,which is used to validate the accuracy of the theoretical method. Results indicate that experimental and finite element analysis results are found to be in good agreement with theoretical results; linear relationships are existed between the cord length and RWCB,and also existed between the position of belt and RWCB; tires designed by the methods have smaller and more uniform displacement,so the method can be used for tire optimized design.
