Over expression of Resistin in adipose tissue of the obese induces insulin resistance

AIM: To compare Resistin mRNA expression in subcutaneous adipose tissue (SAT) and its correlation with insulin resistance (IR) in postmenopausal obese women. METHODS: A total of 68 postmenopausal women (non obese = 34 and obese = 34) were enrolled for the study. The women of the two groups were age matched (49-70 years). Fasting blood samples were collected at admission and abdominal SAT was obtained during surgery for gall bladder stones or hysterectomy. Physical parameters [age, height, weight, body mass index (BMI)] were measured. Biochemical (plasma insulin and plasma glucose) parameters were estimated by enzymatic methods. RNA was isolated by the Trizol method.SAT Resistin mRNA expression was done by real time- reverse transcription polymerase chain reaction (RT-PCR) by using Quanti Tect SYBR Green RT-PCR master mix. Data was analyzed using independent Student's t test, correlation and simple linear regression analysis. RESULTS: The mean weight (52.81 ± 8.04 kg vs 79.56 ± 9.91 kg; P Resistin level was also significantly higher in postmeno-pausal obese women compared to postmenopausal non obese women (9.05 ± 5.15 vs 13.92 ± 6.32, P Resistin mRNA expression was also significantly (0.023 ± 0.008 vs 0.036 ± 0.009; P Resistin mRNA expression showed positive (direct) and significant correlation with BMI (r = 0.78, P Resistin (r = 0.76, P Resistin mRNA expression in pos

Expression of Resistin Protein in Normal Human Subcutaneous Adipose Tissue and Pregnant Women Subcutaneous Adipose Tissue and Placenta

The expression of Resistin protein in normal human abdominal, thigh, pregnant women abdominal, non-pregnant women abdominal subcutaneous adipose tissue and placenta and the relationship between obesity, type 2 diabetes mellitus （T2DM）, pregnant physiological insulin resistance （IR） and gestational diabetes mellitus （GDM） was investigated. The expression of Resistin protein in normal human abdominal, thigh, pregnant women abdominal, non-pregnant women abdominal subcutaneous adipose tissue and placenta was detected by using Western blotting method. Fasting serum glucose concentration was measured by glucose oxidase assay. Serum cholesterol （CHOL）, serum triglycerides （TG）, serum HDL cholesterol （HDL-C） and serum LDL cholesterol （LDL-C） were determined by full automatic biochemical instrument. Fasting insulin was measured by enzyme immunoassay to calculate insulin resistance index （IRI）. Height, weight, systolic blood pressure （SBP） and diastolic blood pressure （DBP） were measured to calculate body mass index （BMI） and body fat percentage （BF%）. Resistin protein expression in pregnant women placental tissue （67 905±8441）（arbitrary A values） was much higher than that in subcutaneous adipose tissue in pregnant women abdomen （40 718 ± 3818, P〈 0. 01 ）, non-pregnant women abdomen （38 288±2084, P〈0.01）, normal human abdomen （39 421±6087, P〈0.01） and thigh （14 942 ±6706, P〈0.001） respectively. The Resistin expression in abdominal subcutaneous adipose tissue showed no significant difference among pregnant, non-pregnant women and normal human, but much higher than that in thigh subcutaneous adipose tissue （P〈0. 001）. Pearson analysis revealed that Resistin protein was correlated with BMI （r=0.42）, fasting insulin concentration （r=0.38）, IRI （r=0.34）, BF% （r=0.43） andfasting glucose （r=0.39）, but not with blood pressure, CHOL, TG, HDL-C and LDL-C. 目的:探讨子痫前期患者胎盘组织中脂源性分泌因子瘦素(leptin)、抵抗素(Resistin)、脂联素(adiponectin)、内脂素(visfatin)mRNA表达水平及其意义。方法:采用RT-PCR方法测定36例正常足月妊娠妇女(A组)、24例轻度子痫前期患者(B组)和17例重度子痫前期患者(C组)胎盘组织中脂源性分泌因子leptin、Resistin、adiponectin、visfatin mRNA表达水平。结果:①C组leptin、Resistin mRNA表达水平分别为1.361±0.261、1.515±0.177,显著高于B组的0.898±0.201、1.055±0.113和A组的0.559±0.175、0.508±0.119(P0.05)。结论:脂源性分泌因子leptin、Resistin、adiponectin、visfatin在子痫前期的发病中有重要作用,可能参与了胰岛素抵抗的病理生理过程。

The objective of this study was to investigate the effects of Resistin on insulin signaling in human umbilical vein endothelial cells(HUVECs).HUVECs were incubated with recombinant human Resistin(0–100 ng/mL)for 24 h.Akt and endothelial nitric oxide synthase(eNOS)phosphorylation levels of endothelial cells under basal or insulin stimulated conditions were measured by Western blot.Nitric oxide(NO)production of HUVECs was also detected.The results showed that Resistin could signiﬁcantly inhibit Akt and eNOS phos-phorylation and NO production in endothelial cells under insulin stimulated conditions(PResistin could result in a decrease in eNOS phosphorylation(PResistin exerted an inhibitory effect on NO production by inhibiting insulin signaling and eNOS phosphorylation in endothelial cells.

目的检测溃疡性结肠炎患者的血清抵抗素（Resistin）与几丁质酶-3样蛋白-1（YKL-40）水平,探讨其在溃疡性结肠炎发生发展中的作用。方法收集符合入选标准的溃疡性结肠炎患者52例及性别、年龄相匹配的健康体检者52例，采用酶联免疫吸附试验法（ELISA法）测定血清Resistin及YKL-40水平，比较它们在溃疡性结肠炎患者与健康对照者的差异及治疗前后两者水平的变化，分析其与疾病活动程度、病变范围的关系及其与超敏C反应蛋白的相关性。结果溃疡性结肠炎患者血清Resistin水平、YKL-40水平及hs-CRP水平分别为19.56±4.13ng/m1,29.38±6.30ng/m1,12.42±3.70mg/L,均显著高于正常对照组（13.89±3.15ng/m1,20.0±4.83ng/m1,4.76±1.16mg/L），差异有统计学意义（P＜0.01）。溃疡性结肠炎患者经氨基水杨酸制剂或糖皮质激素治疗1周后血清Resistin水平、YKL-40水平明显降低（分别为14.56±4.14ng/m1，22.35±4.77ng/m1）,差异有统计学意义（p＜0.05）。重度溃疡性结肠炎患者血清Resistin、YKL-40水平明显高于轻、中度患者，且中度患者血清Resistin水平高于轻度者，但轻、中度患者中血清YKL-40水平无明显差异（P＞0.05）。病变累及全结肠的溃疡性结肠炎患者血清Resistin、YKL-40水平明显高于病变累及左半结肠、直肠及直肠乙状结肠患者，左半结肠组患者血清YKL-40水平高于直肠及直肠乙状结肠组患者，但血清Resistin水平在直肠及直肠乙状结肠组、左半结肠组患者中无明显差异（P＞0.05）。溃疡性结肠炎患者血清Resistin水平与hs-CRP水平相关（r=0.457，P ＜0.05）;血清YKL-40水平与hs-CRP水平呈正相关（r=0.557, P ＜0.01）。结论 1.溃疡性结肠炎患者血清抵抗素及YKL-40水平增高，且与病变范围、临床分度相关，提示它们可作为疾病活动的指标;2.溃疡性结肠炎患者治疗后血清Resistin水平、YKL-40水平较治疗前明显降低，提示可作为其疗效判断的指标;3.溃疡性结肠炎患者血清Resistin水平、YKL-40水平与hs-CRP存在相关性，提示联合检测血清抵抗素、YKL-40及hs-CRP对溃疡性结肠炎的诊断、病情监测、疗效评估等方面有一定的作用。

目的研究左旋甲状腺素(L-T4)替代治疗对亚临床甲状腺功能减退(SCH)孕妇外周血外周血抵抗素(Resistin)、异前列腺素(8-iso-PGF2α)水平的影响,为临床治疗提供参考依据。方法选择2014年6月-2016年9月在该院产科进行产检的孕妇100例为研究对象,根据是否自愿接受治疗分为研究组和对照组,每组各50例。研究组给予L-T4替代治疗,对照组不予干预。比较两组患者甲状腺素、促甲状腺素、血脂水平、Resistin、8-iso-PGF2α水平、并发症及胎儿窘迫、胎儿受限情况。结果治疗后,研究组FT3、TSH水平显著降低于对照组(PResistin水平显著升高于对照组(P0.05)。结论 L-T4替代治疗SCH,通过提高孕妇外周血Resistin水平,降低8-iso-PGF2α水平,调节血脂,疗效安全可靠。

Expression of Resistin mRNA in Adipose Tissue of Rat Model with Polycystic Ovarian Syndrome and Its Implication

Summary: To investigate the relationship between the insulin resistance (IR) of polycystic ovarian syndrome (PCOS) rat model induced by dehydroeplandrosterone (DHEA) and hormonal changes in the ovarium and the Resistin mRNA levels in adipose tissue, 21-day-old female SD rats were divided into two groups in pairs. The rats in group 1 were injected daily (s.c) with DHEA for up to 20 days and the rats in group 2 injected with oil at the same time. Ovarian weight, serum insulin levels and sex hormone levels in rat blood of both groups were determined. Oral glucose tolerance tests, light microscopic and electronic microscopic examination were performed. The levels of Resistin mRNA in adipose tissue were measured by reverse transcription-polymerase chain reaction (RT-PCR). Our results showed that the ovarian weight in group 1 was greater than that in group 2 (PResistin mRNA level of white adipose tissue in the DHEA-induced group was significantly higher than that in the control rats (PResistin secreted by adipose tissue may mediate IR in PCOS, and it is likely involved in the pathogenesis and development of PCOS.

Baseline Resistin predicts the incidence of cardiovascular events in acute coronary syndrome patients after stent placement

Previous studies have implicated a strong link between circulating plasma Resistin and coronary artery disease （CAD）. The aim of this study was to investigate whether Resistin elevation predictsworse outcome ofstent-placement in acute coronary syndrome （ACS） patients. Methods Totally 126 ACS participants who were eligible for stent-placement were enrolled for 2 years＇ follow-up by monitoring clinical end points occur- ring via outpatient visits or/and telephone call. Baseline characteristics were collected. All participants were divid- ed into low Resistin group （〈 3.464 μg/mL） and high Resistin group （〉 3.464 μg/mL）. Clinical end points in terms of major adverse cardiovascular events （MACE）, cardiovascular death, non-fatal myocardial infarction, ischemic stroke, and coronary revascularization （CR） were compared. Results Patients with stenosis in three- vessels presented more frequently in high Resistingroup than in low Resistingroup （35.6% vs. 48.3%, P = 0.001）. Incidences of MACE and CR weresignificantlyhigher in high Resistin group than those in low Resistingroup （31.6% vs. 18.8%, P = 0.018）; 15% vs. 7.58%, P = 0.006）. Conclusions In patients with baseline Resistin eleva- tion, the incidence of MACEis significantly increased after stent-placement.

Resistin is not an appropriate biochemical marker to predict severity of acute pancreatitis:A case-controlled study

AIM:To assess levels of serum Resistin upon hospital admission as a predictor of acute pancreatitis(AP) severity.METHODS:AP is both a common and serious disease,with severe cases resulting in a high mortality rate.Several predictive inflammatory markers have been used clinically to assess severity.This prospective study collected data from 102 patients who were diagnosed with an initial acute biliary pancreatitis between March 2010 and February 2013.Measurements of body mass index(BMI) and waist circumference(WC) were obtained and serum Resistin levels were analyzed at the time of hospital admission using enzyme-linked immunosorbent assay.Additionally,Resistin levels were measured from a control group after matching gender,BMI and age.RESULTS:A total of 102 patients(60 females and 42 males) were diagnosed with acute gallstone-induced pancreatitis.The mean age was 45 years,and mean BMI value was 30.5 kg/m2(Obese,class?Ⅰ).Twentytwo patients(21.6%) had severe AP,while eighty-eight patients had mild pancreatitis(78.4%).Our results showed that BMI significantly correlated with pancreatitis severity(P = 0.007).Serum Resistin did not correlate with BMI,weight or WC.Furthermore,serum Resistin was significantly higher in patients with AP compared to control subjects(P Resistin values upon admission were 17.5 ng/mL in the severe acute biliary pancreatitis group and 16.82 ng/mL in the mild AP group(P = 0.188),indicating that Resistin is not an appropriate predictive marker of clinical severity.CONCLUSION:We demonstrate that obesity is a risk factor for developing severe AP.Further,although there is a correlation between serum Resistin levels and AP at the time of hospital admission,Resistin does not adequately serve as a predictive marker of clinical severity.

Effect of Tumor Necrosis Factor-αon Resistin Expression in 3T3-L1 Adipocytes and Its Mechanism

Summary: In order to investigate the effect of tumor necrosis factor-α (TNFα) on Resistin expression in 3T3-L1 adipocytes, and further explore its mechanisms, the differentiated 3T3-L1 adipocytes were incubated with 0, 1, 10, 100 ng/mL TNFα respectively for 24 h, and then the expression of Resistin was determined. The differentiated 3T3-L1 adipocytes were incubated with 100 ng/mL TNFα for 3, 6, 24 h respectively, and then the expression of Resistin mRNA was analyzed. 3T3-L1 adipocytes were induced to differentiate into mature adipocytes. The cells were randomly divided into 4 groups for culture. In the control group, no drugs were added. Cells of TNFα group were treated with 100 ng/mL TNFα. In Ro-31-8220 group, 5 μmol/L protein kinase C inhibitor Ro-31-8220 was added. With TNFα+Ro-31-8220 group, 100 ng/mL TNFα were added 1 h after the addition of 5 μmol/L Ro-31-8220. All adipocytes were cultured for 24 h. Reverse transcription-polymerase chain reaction (RT-PCR) and Western blotting were employed to detect the expression of Resistin gene. Our results showed that Resistin protein and mRNA in 3T3-L1 adipocytes were inhibited by TNFα at different concentrations (P0.05). It was concluded that TNFα could inhibit the expression of Resistin in 3T3-L1 adipocytes. The mechanism may be that the expression of Resistin is partly controlled by protein kinase C signal conduction pathway.
