Objective:To study the effects of three-dimensional(3D)collagen scaffold on bone marrow cells to differentiate into ***:We cultured BMCs on the 3D collagen scaffolds and two-dimensional(2D)culture *** morphological characteristics of the differentiated DCs cultured on 2D and 3D collagen scaffold were imaged under optical microscope and Scanning Electron Microscopy(SEM).Next,we analyzed the expressions of CD11b,MHC-II and the co-stimulatory molecules CD40,CD80,CD86 and CD83 in i DCs and m DCs using flow cytometry on CD11c+*** then the phagocytosis ability of generated DCs,i DC-2D and i DC-3D were co-incubated with FITC–dextran and then the intensity of FITC taken up by the DCs was measured using flow cytometry on CD11c+*** addition,the T-cell proliferation was assessed using flow ***,the cytokines profiles of generated DCs were collected to measure IL-10,IL-12p70,and TNF-αby ELISA ***,we evaluated the suppressive effects of the generated DCs in vivo delay type hypersensitivity(DTH)in C57BL/6 ***:Our results showed that differentiated DCs derived from 3D collagen scaffold presented with high expression of CD11b and low expressions of CD11c,co-stimulator and MHC-II molecules compared with 2D ***,these cells possessed a weak antigen uptake ability and suppression of T-cell proliferation in ***,they displayed a potent immunoregulatory function to alleviate allo-delay type hypersensitivity(allo-DTH)in ***:The 3D culture provides a suitable niche in specialized DCs *** researches demonstrated that 3D collagen scaffold culture promotes the differentiation of BMCs into regulatory DCs,CD11bhi MHClo *** differentiated CD11bhi MHClo DCs possessed the capacity to negatively regulate immune response in vitro and in vivo,whereas a high level of IL-10 production.

Objective:We investigated the effect of regulatory dendritic cells(DCreg) on acute graft-versus-host disease(GVHD) and graft-versus-leukemia(GVL) in leukemia mouse models after allogeneic bone marrow transplantation(allo-BMT).Method:A SHI-1 leukemia allo-BMT model of C57BL/6 to BABL/c mice was *** with donor bone marrow cells and splenocytes were subsequently transplanted into myeloablatively irradiated *** animals were divided into the TBI(total body irradiation),leukemia,allo-BMT,and immature DC(im DC),and DCreg ***:Recipient survival time in the DCreg and im DC groups was significantly prolonged compared with that in alloBMT group(P < 0.05).Moreover,mild histological changes of GVHD or leukemia were observed in recipients in DCreg group,and clinical GVHD scores in this group were significantly decreased compared with those in transplantation group and im DC ***,the allogeneic chimerism rate was still 95-100% in DCreg group,suggesting complete donor type *** interferon-γ level decreased in the im DC and DCreg groups compared with that in allo-BMT group and even more significantly reduced in DCreg(P < 0.01).Additionally,Serum interleukin-10 level gradually increased in the im DC and DCreg groups(P< 0.01) whereas decreased in the allo-BMT ***:DCreg cell infusion successfully improved incidence and severity of GVHD in allo-BMT mouse models,while could also preserve the effect of GVL

Cellular strategies to induce immune tolerance after liver transplantation:Clinical perspectives

Liver transplantation(LT)has become the most efficient treatment for pediatric and adult end-stage liver disease and the survival time after transplantation is becoming longer due to the development of surgical techniques and perioperative ***,long-term side-effects of immunosuppressants,like infection,metabolic disorders and malignant tumor are gaining more *** tolerance is the status in which LT recipients no longer need to take any immunosuppressants,but the liver function and intrahepatic histology maintain *** approaches to achieve immune tolerance after transplantation include spontaneous,operational and induced *** first two means require no specific intervention but withdrawing immunosuppressant gradually during *** clinical factors or biomarkers so far could accurately predict who are suitable for immunosuppressant withdraw after *** the understanding to the underlying mechanisms of immune tolerance,many strategies have been developed to induce tolerance in LT *** strategy is one of the most promising methods for immune tolerance induction,including chimerism induced by hematopoietic stem cells and adoptive transfer of regulatory immune *** safety and efficacy of various cell products have been evaluated by prospective preclinical and clinical trials,while obstacles still exist before translating into clinical ***,we will summarize the latest perspectives and concerns on the clinical application of cellular strategies in LT recipients.
