摘      要:Objective:To study the effects of three-dimensional(3D)collagen scaffold on bone marrow cells to differentiate into ***:We cultured BMCs on the 3D collagen scaffolds and two-dimensional(2D)culture *** morphological characteristics of the differentiated DCs cultured on 2D and 3D collagen scaffold were imaged under optical microscope and Scanning Electron Microscopy(SEM).Next,we analyzed the expressions of CD11b,MHC-II and the co-stimulatory molecules CD40,CD80,CD86 and CD83 in i DCs and m DCs using flow cytometry on CD11c+*** then the phagocytosis ability of generated DCs,i DC-2D and i DC-3D were co-incubated with FITC–dextran and then the intensity of FITC taken up by the DCs was measured using flow cytometry on CD11c+*** addition,the T-cell proliferation was assessed using flow ***,the cytokines profiles of generated DCs were collected to measure IL-10,IL-12p70,and TNF-αby ELISA ***,we evaluated the suppressive effects of the generated DCs in vivo delay type hypersensitivity(DTH)in C57BL/6 ***:Our results showed that differentiated DCs derived from 3D collagen scaffold presented with high expression of CD11b and low expressions of CD11c,co-stimulator and MHC-II molecules compared with 2D ***,these cells possessed a weak antigen uptake ability and suppression of T-cell proliferation in ***,they displayed a potent immunoregulatory function to alleviate allo-delay type hypersensitivity(allo-DTH)in ***:The 3D culture provides a suitable niche in specialized DCs *** researches demonstrated that 3D collagen scaffold culture promotes the differentiation of BMCs into regulatory DCs,CD11bhi MHClo *** differentiated CD11bhi MHClo DCs possessed the capacity to negatively regulate immune response in vitro and in vivo,whereas a high level of IL-10 production.
