Stable-isotope probing of bacterial community for dissolved inorganic carbon utilization in Microcystis aeruginosa-dominated eutrophic water

Dissolved inorganic carbon(DIC) is an important source of carbon in aquatic ecosystems,especially under conditions of increased frequency of cyanobacterial bloom. However, the importance of bacteria in direct or indirect utilization of DIC has been widely overlooked in eutrophic freshwater. To identify the functional bacteria that can actively utilize DIC in eutrophic freshwater during cyanobacterial bloom, stable-isotope probing(SIP) experiments were conducted on eutrophic river water with or without inoculation with cyanobacteria(Microcystis aeruginosa). Our 16 S rRNA sequencing results revealed the significance of Betaproteobacteria, with similar relative abundance as Alphaproteobacteria, in the active assimilation of H^(13)CO^(3-) into their DNA directly or indirectly, which include autotrophic genera Betaproteobacterial ammonia-oxidizing bacteria. Other bacterial groups containing autotrophic members, e.g. Planctomycetes and Nitrospira, also presented higher abundance among free-living bacteria in water without cyanobacteria. Microcystis aggregates showed a preference for some specific bacterial members that may utilize H^(13)CO^(3-) metabolized by Microcystis as organic matter, e.g. Bacteroidetes(Cytophagales, Sphingobacteriales), and microcystindegrading bacteria Betaproteobacteria(Paucibacter/Burkholderiaceae). This study provides some valuable information regarding the functional bacteria that can actively utilize DIC in eutrophic freshwater.

Evaluating the topological charge density with the symmetricmulti-probing method

We evaluate the topological charge density of SU(3) gauge fields on a lattice by calculating the trace of the overlap Dirac matrix employing the symmetric multi-probing(SMP) method in 3 modes. Since the topological charge Q for a given lattice configuration must be an integer number, it is easy to estimate the systematic error(the deviation of Q to the nearest integer). The results demonstrate a high efficiency and accuracy in calculating the trace of the inverse of a large sparse matrix with locality by using the SMP sources when compared to using point *** also show the correlation between the errors and probing scheme parameter r_(min), as well as lattice volume N_L and lattice spacing a. It is found that the computational time for calculating the trace by employing the SMP sources is less dependent on N_L than by using point sources. Therefore, the SMP method is very suitable for calculations on large lattices.

Proton probing of laser-driven EM pulses travelling inhelical coils

The ultrafast charge dynamics following the interaction of an ultra-intense laser pulse with a foil target leads to the launch of an ultra-short, intense electromagnetic(EM) pulse along a wire connected to the target. Due to the strong electric field(of the order of GV m^(-1)) associated to such laser-driven EM pulses, these can be exploited in a travelling-wave helical geometry for controlling and optimizing the parameters of laser accelerated proton beams. The propagation of the EM pulse along a helical path was studied by employing a proton probing technique. The pulse-carrying coil was probed along two orthogonal directions, transverse and parallel to the coil axis. The temporal profile of the pulse obtained from the transverse probing of the coil is in agreement with the previous measurements obtained in a planar geometry. The data obtained from the longitudinal probing of the coil shows a clear evidence of an energy dependent reduction of the proton beam divergence, which underpins the mechanism behind selective guiding of laser-driven ions by the helical coil targets.

Numerical Simulation of Near-Field Seismoacoustic probing of a Layer Inclusion in a Homogeneous Infinite Medium

Spatial distribution of acoustic and elastic waves generated by an elementary vibration source at seismic profiling frequencies in an infinite medium close to a layer inclusion, i.e., an extended layer, is numerically simulated. Point dipole radiation in a homogeneous infinite medium separated by a liquid layer of different medium density or acoustic wave velocity is considered. Transverse elastic SH-waves excited by an oscillating power source in a solid medium also located close to the layer of different propagation velocity than the velocity of the vicinity are analyzed. Formulae for the spatial distribution of the wave field amplitude are derived and computer graphics of field distribution images is presented. Wave reflection, penetration deep into the layer inclusion, and transmittance through it are examined. Results of the analysis can be applied to seismoacoustic probing of geologic environment by the near field of a harmonic vibration source.

Long-term nitrogen(N)fertilization imposes strong selection on nitrifying communities in agricultural soil,but how a progressively changing niche affects potentially active nitrifiers in the field remains poorly *** a 44-year grassland fertilization experiment,we investigated community shifts of active nitrifiers by DNA-based stable isotope probing(SIP)of field soils that received no fertilization(CK),high levels of organic cattle manure(HC),and chemical N fertilization(CF).Incubation of DNA-SIP microcosms showed significant nitrification activities in CF and HC soils,whereas no activity occurred in CK *** 44 years of inorganic N fertilization selected only 13C-ammonia-oxidizing bacteria(AOB),whereas cattle slurry applications created a niche in which both ammonia-oxidizing archaea(AOA)and AOB could be *** analysis indicated that Nitrosospira sp.62-like AOB dominated inorganically fertilized CF soils,while Nitrosospira sp.41-like AOB were abundant in organically fertilized HC *** 13C-AOA in HC soils were affiliated with the 29i4 *** 13C-nitrite-oxidizing bacteria(NOB)were dominated by both Nitrospira-and Nitrobacter-like communities in CF soils,and the latter was overwhelmingly abundant in HC *** 13C-labeled nitrifying communities in SIP microcosms of CF and HC soils were largely similar to those predominant under field *** results provide direct evidence for a strong selection of distinctly active nitrifiers after 44 years of different fertilization regimes in the *** findings imply that niche differentiation of nitrifying communities could be assessed as a net result of microbial adaption over 44 years to inorganic and organic N fertilization in the field,where distinct nitrifiers have been shaped by intensified anthropogenic N input.

Chronic Nitrogen Fertilization Modulates Competitive Interactions Among Microbial Ammonia Oxidizers in a Loess Soil

Nitrogen(N) application may lead to niche segregation of soil ammonia-oxidizing archaea(AOA) and bacteria(AOB), thereby reducing the competitive interactions between AOA and AOB due to higher ammonium substrate availability. However, the adaptive mechanisms of AOA and AOB under N enrichment remain poorly understood. Stable isotope probing(SIP) microcosm incubation was employed to reveal community changes of active AOA and AOB in a loess soil from a field experiment growing potatoes that received no N(control, CK), low N(LN, 75 kg N ha^(-1)), and high N(HN, 375 kg N ha^(-1)). The results showed that the soil potential nitrification rate(PNR) was measured by culturing of the soil samples from the field experiment. Soil PNR was significantly increased in HN by87.5% and 67.5% compared with CK and LN, respectively. Compared with CK, the^(13)C-amoA genes of soil AOA and AOB in HN had 2.58 × 10~4 and 1.55 × 10~6 copies, representing 1.6-and 16.2-fold increase respectively. It was indicated that AOB dominated soil ammonia oxidation. A phylogenetic analysis of the^(13)C-amoA gene showed that N application significantly increased the proportion of54 d9-like AOA up to 90% in HN, while the Nitrososphaera gargensis-like and Nitrososphaera viennensis-like AOA were inhibited and completely disappeared. Nitrogen application also resulted in the community shift of active AOB-dominant group from Nitrosospira briensis-like to Nitrosospira sp. TCH711-like. Our study provides compelling evidence for the emergence and maintenance of active nitrifying communities under the intensified N input to an agricultural ecosystem.
