ALL-ARTIFICIAL SPORE-COLLECTING CULTIVATION OF TIAOBAN-ZICAI (porphyra yezoensis ueda)

An account of the all-artificial spore-collecting cultivation of Tiaoban-zicai (porphyra yezoensis ueda) in North China, carried out mainly after the Great Proletarian Cultural Revolution, is presented here. Scientific and technical workers integrating themselves with porphyra-cultivating peasants and learning the experiences of the masses, have eonducted investigations and thereby improved the cultivation techniques principally along three directions: (1) By employing comprehensive techniques in the culture of the conchocelis stage of tile porphyra, we have made at appropriate time the conchocelis discharge large numbers of conchospores, each shell (30—40 cm~2 in area) from 5 millions or more spores to as many as 80 millions in best cases. (2) Taking the discharge of large numbers of conchospores as the basis, and sufficient light intensity and vigorous agitation of the cultures as the conditions for materializing all-artifical spore-collectlng, we have developed a simple but efficient technique by em

＜正＞porphyra yezoensis ueda grows intertidal zone and is one of the most important *** intertidal zone is one of the harshest environments on earth and the intertidal zone algae are frequently exposed to high temperature,high light and osmotic pressure between tidal up and *** the long evolution,P. yezoensis have evolved mechanisms for photo-acclimation that favor survival in high light（HL）.And high light-inducible polypeptide（HLIP）is one of the light resistant *** this paper,we reported the HLIP *** and expression analysis from *** analysis indicates that it contains an ORF of 387 bp without intron encoding a polypeptide of 128 amino acids with similarity to HliC from Synechocystis *** of *** has a molecular mass of 13 kDa and have a theoretical pI of *** PHDhtm program clearly predicted a membrane-spanning-helix between 54-83 sites which is the conservation motif of light-harvesting chlorophyll a/b-binding（LHC）***-PCR analysis shows the high light induces expression of HLIP and under low high light no *** addition,the lengtj of exposed to high light affected the expression of HLIP.

＜正＞porphyra yezoensis ueda,as one of the most economically important marine crops,is own species in west of Pacific Ocean and has been predominately cultivated in China,Japan and Korea *** is now considered one of the best model organisms for molecular biology research and for genomic research of ***,molecular biology research in porphyra is far behind that in land *** of molecular markers will provide useful information for genomic study of *** this work,microsatellite markers were developed and applied to genetic analysis.A genomic library from ***,highly enriched for dinucleotide（GA）n/（CA）n motifs,was *** the clones with repeat sequence probes revealed positive clones account for about 75% of the total *** about 300 positive clones have been obtained and 147 clones have been *** these,30 primer pairs have been designed and 24（80%）can produce a single or double band.30 individual of the same *** line were used to analyze the characterization of the SSR *** ten population from different districts were used to study the population genetics of *** in *** works are still in *** microsatcllite markers developed in this study will be useful for genetic diversity analysis,linkage map construction as well as for genetic breeding ***.
