pomegranate leaf attenuates lipid absorption in the small intestine in hyperlipidemic mice by inhibiting lipase activity

pomegranate leaf(PGL)has a definite role in regulating lipid ***,pharmacokinetic results show the main active ingredient,ellagic acid,in PGL has lower oral bioavailability,suggesting that the lipid-lowering effect of PGL may act through inhibiting lipid absorption in the small *** results demonstrated that pomegranate leaf and its main active ingredients(i.e.,ellagic acid,gallic acid,pyrogallic acid and tannic acid)were capable of inhibiting pancreatic lipase activity in *** computational molecular docking,the four ingredients had good affinity for pancreatic *** lipid overload experiments showed that a large dosage of PGL significantly reduced serum total cholesterol(TG)and triglycerides(TC)levels in addition to inhibiting intestinal lipase activity,which demonstrated that PGL could inhibit lipase activity and reduce the absorption of *** also found that PGL could reverse the reduced tight-junction protein expression due to intestinal lipid overload,promote Occludin and Claudin4 expression in the small intestine,and enhance the intestinal mucosal *** conclusion,we demonstrated that PGL can inhibit lipid absorption and reduce blood TG and TC by targeting pancreatic lipase,promoting tight-junction protein expression and thereby preventing intestinal mucosa damage from an overload of lipids in the intestine.

Protective Effects of pomegranate and Its Bioactive Components on Hepatic Disease

By summarizing the pharmacological effects of pomegranate extract and its active components,such as punicalagin,punicalin,gallic acid,ellagic acid,caffeic acid,and chlorogenic acid,it is found that the extract exhibits therapeutic effects on liver injury,viral hepatitis,metabolic dysfunction-associated fatty liver disease,liver fibrosis,and liver *** evidence suggests that these natural products may alleviate liver diseases through multi-targeted therapeutic mechanisms,including anti-inflammation,anti-oxidative stress,immunoregulation,and *** underlying mechanisms by which pomegranate exerts hepatoprotective activities may be attributed to the regulation of multiple signaling pathways,including P62/Nrf2,TGF-β1/Smad7,Wnt/β-catenin,MAPK/Nrf2,Nrf2/Keap1,Akt/FOXO3a,MAPK/NF-κB,***,pomegranate can serve as a functional food,nutritional supplement,or adjuvant in the modern treatment of liver diseases.

Efficiency of combining pomegranate juice with low-doses of cisplatin and taxotere on A549 human lung adenocarcinoma cells

Objective: To test the coalescence effect of two chemotherapy drugs at low effective dose(cisplatin and taxotere) combined with pomegranate juice on A549 cancer cells. Methods: Infrared spectroscopy method is a qualitative test that was performed to ensure the existence of the phytochemicals providing the antioxidant activity through the presence of the hydroxyl group(-OH). The viability of A549 cell line and normal MCs was tested using the neutral red uptake, Clonogenic survival, XTT and Cell migration assays. Results: Our results showed that this combination firstly led to a greater decrease in the viability of cells comparing to those treated with chemotherapy drugs alone, and secondly led to a significant reduction in cell migration. Conclusions: These data suggest a synergistic effect between the pomegranate and cisplatin which makes probably this combination a powerful option for treating lung adenocarcinoma and in parallel minimizing the systemic side effects.

Exploring the Transcriptome Landscape of pomegranate Fruit Peel for Natural Product Biosynthetic Gene and SSR Marker Discovery

pomegranate fruit peel is rich in bioactive plant natural products, such as hydrolyzable tannins and anthocyanins. Despite their documented roles in human nutrition and fruit quality, genes involved in natural product biosynthesis have not been cloned from pomegranate and very little sequence information is available on pomegranate in the public domain. Shotgun transcriptome sequencing of pomegranate fruit peel cDNA was performed using RNA-Seq on the Illumina Genome Analyzer platform. Over 100 million raw sequence reads were obtained and assembled into 9,839 transcriptome assemblies （TAs）（〉200 bp）. Candidate genes for hydrolyzable tannin, anthocyanin, flavonoid, terpenoid and fatty acid biosynthesis and/or regulation were identified. Three lipid transfer proteins were obtained that may contribute to the previously reported IgE reactivity of pomegranate fruit extracts. In addition, 115 SSR markers were identified from the pomegranate fruit peel transcriptome and primers were designed for 77 SSR markers. The pomegranate fruit peel transcriptome set provides a valuable platform for natural product biosynthetic gene and SSR marker discovery in pomegranate. This work also demonstrates that next-generation transcriptome sequencing is an economical and effective approach for investigating natural product biosynthesis, identifying genes controlling important agronomic traits, and discovering molecular markers in non-model specialty crop species.

Fe Nanoparticles Synthesized by pomegranate Leaves for Treatment of Malachite Green

A green and convenient pathway of preparing iron nanoparticles(FeNPs)with pomegranate leaf(PG)extract for highly effective removal of malachite green(MG)was proposed under ambient *** materials were characterized by scanning electron microscope(SEM),X-ray energy-dispersive spectrometer(EDS),Fourier transform infrared spectroscopy(FTIR),X-ray diffraction(XRD)and X-ray photoelectron spectroscopy(XPS)*** results show that their surfaces are capped and stabilized by PG extract with amorphous nature and without any detection of zero-valent *** size and surface valence state of FeNPs are the key factors that affect the MG removal *** the reagent volume ratio of PG extract to FeCl_(3) increases greater than 1,the cross-linked FeNPs become more obvious,having a homogeneous distribution with the size range from 30 to 40 nm,and show an increasing ratio ofFe(Ⅱ)/Fe(Ⅲ),which is in proportion to the degradation efficiency of MG,reaching higher than 95%in only 2 min by using 50 mg Fe/L FeNPs and 200 mg/L MG.

Colorectal cancer is the second leading cause of cancer-related deaths in the United *** studies prove that though chemotherapeutic agents are being used for the treatment of colon cancer,they become non-effective when the cancer progresses to an invasive *** consumption of certain dietary agents has been linked with various cancers,fruit juices have been investigated for their consistently protective effect against colon *** unique biochemical composition of fruit juices is responsible for their anticancer *** this review,the chemo-preventive effect of fruit juices such as pomegranate and citrus juices against colon cancer are *** this purpose,the bioavailability,in vitro and in vivo effects of these fruit juices on colorectal cancer are ***,there is a scarcity of studies involving human trials to estimate the preventive nature of these juices against colon *** review will support the need for more preclinical tests with these crude juices and their constituents in different colorectal cancer cell lines and also some epidemiological studies in order to have a better understanding and promote pomegranate and citrus juices as crusaders against colon cancer.

Fermentation of pomegranate juice by lactic acid bacteria and its biological effect on mice fed a high-fat diet

This study investigated the ability of Lactiplantibacillus paraplantarum CRL2051 and Lactiplantibacillus plantarum CRL2030,two lactic acid bacteria isolated from fruits,to ferment pomegranate juice to produce beverages with extended shelf life and functional *** acid bacteria consumed glucose and fructose as carbon sources,producing mainly lactic *** strains were initially grown in juice at 30℃ for 48 h,then refrigerated at 4℃ for 28 *** fermentation and cold storage,the total phenolic content and antioxidant capacity were conserved in fermented pomegranate *** functionality studies of the fermented beverages administered to C57BL/6 mice fed a high-fat diet were evaluated over 6 *** juice supplementation led to a marked improvement in glucose and triglycerides serum profile,which decreased around 40%and up to 48%,respectively,and counteracted fat deposition and body weight *** high-fat diet treatment significantly incremented the aspartate aminotransferase activity in the liver,whereas fermented pomegranate juice consumption slightly reduced *** histological changes associated with a high-fat diet were significantly reverted in juice-treated *** findings evidenced the biological benefits of a novel functional beverage obtained by lactic fermentation of pomegranate juice;this functional drink may protect against weight gain,liver damage,and dyslipidemia induced by a high-fat diet consumption.
