The hepatopancreatic necrosis disease(HPND)is a common shrimp *** of the black tiger shrimp penaeus monodon has suffered with the frequent outbreak of *** the molecular mechanisms of host response to HPND is of pivotal importance for improving its farming *** this study,the RNA-Seq platform was utilized to investigate transcriptomic changes of the hepatopancreas in pond-cultured *** with HPND symptoms.A total of 62071 genes with an average length of 953 bp were obtained,and a lot of simple sequence repeat(SSR)loci related to these genes were ***,5204 differentially expressed genes(DEGs)were detected between healthy shrimp and HPND shrimp,with 3399 genes upregulated and 1,805 genes *** genes had a wide range of biological functions,and several well-known immunerelated genes including integrin alpha 5,integrin beta 1,C type lectin and catalase were among the *** signaling pathways including the extracellular matrix(ECM)receptor interaction,phagosome and lysosome pathways were significantly upregulated in HPND-infected shrimps,indicating their involvement in the immune responses of shrimp against *** data obtained in this study offers new insights into the molecular mechanisms of shrimp host response to HPND disease,and provides a resource for molecular marker development and genetic breeding studies of ***.

Objective:To examine the structure of the oviduct of the shrimp penaeus ***:The oviducts of *** with three different major groups of ovarian development(Group(Gr.) 1:Stages I & V;Gr.2:Stages II & III;and Gr.3:Stage IV) were examined by light,transmission electron,and scanning electron microscopies,***:The epithelium of the oviduct in Gr.1 was composed of tall simple columnar cells with their basal nuclei located on the basement membrane and its thick collagen *** Gr.2,the oviduct seemed to produce some substances and their epithelial cells became transitional with centrally located nuclei and formed some ***,the epithelial cells in Gr.3(at Stage IV) were disorganized,disrupted,and shed accumulated spherical secretory substances including some cellular contents into the ***:The structural changes of the *** oviduct were related to ovarian maturation stages(Grs.1-3).Prior to spawning,only the oviduct epithelium at ovary Stage IV produced and secreted a number of spherical secretion substances into the *** substances may act as the oviductal lubricants to facilitate the spawning process.

Dredging influenced sediment toxicity of endosulfan and lindane on black tiger shrimp (penaeus monodon Fabricius) in Chantaburi River estuary in Thailand

Series of experiments, including background sediment elutriate, spiked-sediment elutriate and spiked- water toxicity tests, were carried out to study the effect of dredging on survivability and ecological competence of commercially important shrimp postlarvae of penaeus monodon Fabricius. The test sediment represented the most contaminated from endosulfan and lindane in the Chantaburi river mouth estuary. The maximum sediment contamination levels were 49.4 μg/kg of endosulfan and 58.1 μg/kg of lindane in dry weight basis. Dose-response studies in static, non-renewal spiked-sediment elutriate toxicity tests revealed that LC 50 (96 h) values for survival effect based on measured concentrations were 0.50 (0.26-0.95) μg/L for endosulfan and 0.92 (0.63-1.34) μg/L for lindane. The LOECs for endosulfan and lindane were 0.141 and 0.429 μg/L, respectively. The NOECs were estimated to be the dissoluted endosulfan and lindane concentrations from the Chantaburi estuary sediment. The effects on phenoloxidase enzyme activity in shrimp postlarvae exposed for sub-lethal to lethal doses in sediment elutriate were not significant (p>0.05) at all tested dose levels. The worst case dissolution concentrations of endosulfan and lindane in the water column upon simulatory dredging in the Chantaburi estuary were found to be toxic to shrimp postlarvae on lethal and sub-lethal effects. The NOECs of respective chemicals (i.e., 0.034 and 0.16 μg/L) shared the same orders of magnitude for corresponding NOECs established for whole salt water aquatic life by the USEPA and "safe" levels calculated by 0.01 safety factor approach. In addition, the significance of multiple pollutant assessments for ecological toxicity testing is highlighted.

mRNA was isolated from the hepatopancrease of shrimp penaeus monodon with a PolyAT-tract System 1000 Kit. By using mRNA as template, double - strand cDNA with EcoR I/Xho I ends was synthesized by using a ZAP Express cDNA Synthesis Kit. The cDNA was inserted into the lambda ZAP Express vector predigested with EcoR l/Xho Ⅰ, and the recombinant DNA was in vitro packaged into lambda phage with GigapackⅢ Gold packaging extracts. These recombinant phages were then used to transfect E. coli XL1 - Blue MRF', and finally a cDNA expression library was constructed. The library is 7.2 × 10~5pfu in capacity and its recombination ratio is higher than 99 % . The size of the inserted cDNAs was determined by EcoR l/Xho I digestion of 9 phagemids prepared by in vivo excision of plaques selected randomly from amplified cDNA library . The longest inserted cDNA is about 1.6 kb in length. The complete sequence (about 1.2 kb) of actin cDNA was amplified from the library by PCR reveals that this library contains full-length cDNAs of penaeus monodon hepatopancreas and is available for screening and expression of shrimp genes.

Distribution of injected Vibrio anguillarum in body of penaeus monodon was studied with immunohistochemical method. Bacteria could be detected throughout the experiment in some individuals; however in lymphoid tissue, gill, heart and haemolymph of all vibrio injected shrimp, the bacteria could be observed only 5 min after injection. The bacteria density in haemolymph, haemolymph of the hepatopancreas and gills decreased with time. In the lymphoid organ and heart, the bacteria density was the highest 48 h after injection, then decreased. Nodules could be formed in the heart, lymphoid organ and injection site.

＜正＞In present study,a M-phase phosphoprotein 6（Mpp6） gene was obtained from the ovary and neurosecretory organ in eyestalk cDNA library of black tiger shrimp（penaeus monodon）.By PCR testing,comparisons of nucleotides showed 95%homologies between the PCR product and the partial *** full length black tiger shrimp Mpp6（PmMpp6） cDNA have obtained,homology analysis of the deduced amino acid sequence of the PmMpp6 with other known Mpp6 sequences by MatGAT software revealed that the PmMpp6 was high homology with other invertebrates.A conserved signature sequence of the Mpp6 family was found in the PmMpp6 deduced amino acid *** of the tissue expression pattern of the PmMpp6 showed that the PmMpp6 mRNA was expressed in all tissues tested,with highest levels in lymphoid tissue and ***,the Mpp6 expression was found to be different in six stages of ovarian development.
