Trypanosoma cruzi is the causative agent of Chagas *** parasite requires the intracellular niche in order to proliferate and disseminate the *** invasion,*** resides temporarily in an acidic vacuole which is lysed by a not well-understood *** electron microscopy was used to describe the process of *** escape from the parasitophorous vacuole over the *** HeLa(non-professional phagocytic cells)as host cell,we observed that recently internalized parasites reside in a membrane-bounded vacuole.A few hours later,the first sign of vacuole disruption appeared as membrane *** observation was followed by a progressive vacuole swelling as evidenced by an electron-lucent halo between the parasite and the vacuole ***,the vacuole membrane remnants reorganized as small vesicles that eventually disappeared from the vicinity of the ***,parasites reach the host cell cytosol where replication takes *** thorough ultrastructural description of this process set the base for a comprehensive understanding of the parasite-host cell interaction and,thus open the possibility of new therapeutic intervention strategies.

Toxoplasma gondii is an intracellular protozoa which can infect almost all kinds of nucleated *** is estimated that approximately 1/3 of the world’s population to be chronically infected with tissue cysts of this *** dwells inside a parasitophorous vacuole(PV)for development and multiplication after *** of the PV membrane(PVM)components come from the host membrane with only a small part from the parasite and host *** small GTPases Rho A,Rac1 and Rac2 were found recruited in the formation of PVM upon *** RH and Pru tachyzoites invasion to COS 7 cells and this involvement is dependant on the normal GTPase *** phenomena were testified by using over expression of Cyan Fluorescent Protein(CFP)tagged wild type Rho A,Rac1 and Rac2;Dominant Negative mutants of Rho N19,Rac1 N17 and Rac2N17;and constitutively active mutants of Rho A L63,Rac1 L61 and Rac2 L61 in COS7cells,and then infected with *** RH *** the wild type Rho A,Rac1and Rac2 were found involved in the *** 10 amino acids sequentially truncated Rho A mutants were used for the identification of the motif(s)of Rho A to decide the efficient recruitment into the *** enzymatic activity site deletion mutants Rho A D11-20 and Rho A D61-70 were found loosing the ability of being involved in *** chemotractant Epithelial Growth Factor(EGF)was used to activate Rho A WT transfected COS7 *** translocation of CFP tagged Rho A WT towards EGF in COS7 cells was not found for the protein recruited in the *** the contrary,the translocation was found for the protein in the host cell membrane and *** result implicated the Rho A recruited in PVM was isolated form the actomyosin *** infection rate and infetioncity of *** to host cells were compared among different group of COS7 cells:(A)mock;(B)Rho A WT over expression;(C)Rho N19over expression;(D)Rho A L63 over expression;(F)Rho A knocked-down with *** A WT over expre
