Cardiovascular metabolic diseases are the leading cause of death worldwide, and their pathogenesis has attracted much attention. non-targeted metabolomics serves as a more important approach for exploring their pathogenesis in recent years. The use of coenzyme Q10(CoQ10) as an adjuvant treatment with routine clinical therapy against the risks of cardiovascular disease has shown benefits. However, there is still no study to explore the potential biomarkers related to the risks of cardiovascular metabolic diseases after coenzyme Q10 intervention via the application of non-targeted metabolomics. The aim of the present work is to determine the biological effects of Coenzyme Q10 on new biomarkers related to the risks of cardiovascular metabolic diseases via the nontargeted metabolomics approach. Sources of Support: This work was supported by the National Natural Science Foundation of China(No. 82030098, 81872617 and 81730090), and Shenzhen Science, Technology, and Innovation Commission(No. JCYJ20180307153228190). Methods In this randomized, double-blinded, placebo-controlled trial, 95 dyslipidemic subjects without taking any hypoglycemic or hypolipidemic drugs were administrated to 120 mg CoQ10 or placebo daily for 24 weeks. Ultra-performance liquid chromatography coupled with Triple TOF 5600 plus(UPLC-MS/MS) was used to analyze the serum metabolites of subjects with 24-week coenzyme Q10 supplementation or 24-week placebo. The treatments of acquired MS data and LC-MS data were performed using XCMS software and R software. The online KEGG, HMDB database was used to annotate the metabolites by matching the exact molecular mass data(m/z) of samples with those from the database. Results The non-targeted approach shows that after 24-weeks consumption of coenzyme Q10 significantly changes several metabolites related to cardiovascular metabolic diseases. Objective To investigate changes in the urinary metabolite profiles of children exposed to polycyclic aromatic hydrocarbons(PAHs)during critical brain development and explore their potential link with the intestinal *** Liquid chromatography-tandem mass spectrometry was used to determine ten hydroxyl metabolites of PAHs(OH-PAHs)in 36-month-old ***,37 children were categorized into low-and high-exposure groups based on the sum of the ten ***-high-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry was used to identify non-targeted metabolites in the urine ***,fecal flora abundance was assessed by 16S rRNA gene sequencing using Illumina *** The concentrations of 21 metabolites were significantly higher in the high exposure group than in the low exposure group(variable importance for projection>1,P<0.05).Most of these metabolites were positively correlated with the hydroxyl metabolites of naphthalene,fluorine,and phenanthrene(r=0.336–0.531).The identified differential metabolites primarily belonged to pathways associated with inflammation or proinflammatory states,including amino acid,lipid,and nucleotide ***,these distinct metabolites were significantly associated with specific intestinal flora abundances(r=0.34–0.55),which were mainly involved in *** Higher PAH exposure in young children affected metabolic homeostasis,particularly that of certain gut microbiota-derived *** investigation is needed to explore the potential influence of PAHs on the gut microbiota and their possible association with neurodevelopmental outcomes.

Trimester-specific urinary metabolome alterations associated with gestational diabetes mellitus:A study in different pregnancy stages

Gestational diabetes mellitus(GDM),a frequently-occurring disease during pregnancy,may cause some adverse healthy outcome of both mother and ***,the knowledge about metabolite alterations during the pathogenesis and development process is ***,a large longitudinal nontargeted metabolomics study of 195 pregnant women(64 women with subsequently developed GDM and131 healthy controls)was *** participant provided urine samples at three timepoints during early,middle and late pregnancy,*** metabolic profiles of 585 urine samples(195×3)were measured by using ultra-high performance liquid chromatography coupled with Orbitrap high-resolution mass *** the 56 identified metabolites,the levels of eight metabolites increased and three ones decreased in the first trimester,the concentration of one metabolite increased and those of 20 decreased in the second trimester,as well as the levels of five metabolites increased and two decreased in the third *** false discovery rate correction,the levels of valine and 5-acetamidovalerate in GDM group significantly increased in the first trimester,the levels of 1-methylguanine and 1,3-dihydro-(2 H)-indol-2-one significantly decreased in the second trimester and three metabolites(threonine,OH-octanedioyl-carnitine and pimelylcarnitine)increased and N-acetyltryptophan decreased in the third trimester,*** metabolites,such as pantothenic acid and threonine,had significant interaction effects between gestational stage(different trimester)and group(GDM or control).The differential metabolites were involved in“tryptophan metabolism”,“purine metabolism”,“valine,leucine and isoleucine degradation”and other *** findings may provide insights into further pathogenesis study of GDM.
