In vitro antibacterial activity and major bioactive components of Cinnamomum verum essential oils against cariogenic bacteria,Streptococcus mutans and Streptococcus sobrinus

Bioprospecting for anti-Streptococcus mutans:The activity of 10% Sesbania grandiflora flower extract comparable to erythromycin

Objective:To search an herbal material,capable of inhibiting plaque producing bacteria Streptococcus ***:Twenty materials comprising 10 flowers and 10 rhizomes were extracted with70% *** activity was then examined at a concentration of 10%(w/v) against Streptococcus mutans in vitro on Mueller–Hinton ***(Oxoid,20 mg disc) was used as a positive ***,to establish a fingerprint guide for authentication or quality control,the most potent material was further analyzed regarding its chemical constituents by means of reversed phase-high performance liquid chromatography(HPLC) and thin-layer chromatography(TLC).Results:Of the tested samples,Sesbania grandiflora(***) flower and Costus speciosus rhizome extracts showed the most potent activity with inhibited zone diameters of 18.5 and 14.8 mm,*** the other hand,other extract plants showed a diameter zone in the range of 0.5–10.6 mm or being inactive(diameter=0 mm).The activity of *** was comparable to that of erythromycin(diameter=18.0 mm).The best separation was achieved on HPLC system with acetonitrile-water with a ratio of2:8,and a flow rate at 0.5 m L/***,meanwhile,was featured on chloroform–methanol(8.5:1.5) as a mobile ***:*** flower is a promising material to be developed as the active ingredient of anti-plaque toothpaste as well as mouthwash *** developed HPLC and TLC system can be used for a further standard in its material authentication as well as for a fingerprinting of quality control during the manufacturing process.

Efficacy of xylitol and fluoride mouthrinses on salivary mutans streptococci

Objective:To evaluate the level of salivary Mutatis streptococci(MS) after rinsing with xylitol, fluoride,and a combination of xylitol and fluoride solutions,compared with distilled water. Evaluation of Surface Roughness and <i>Streptococcus mutans</i>Adhesion to Bulk-Fill Resin Composites Polished with Different Systems

Purpose: Bacterial adhesion represents the initial step in biofilm formation, dental caries and decay. This study aimed to evaluate and compare surface roughness and bacterial adhesion to bulk fill resin composites polished with different systems. Methods: Filtek Z350 XT (Incremental-fill resin composite), Filtek Bulk-fill Posterior (Bulk-fill resin composite), and Tetric N Ceram (Bulk-fill resin composite) were used as resin composites. The polishing systems used in this study were Sof-Lex multi-step, PoGo one step, and Mylar strip. Scanning electron microscope (SEM) was used to examine the surface roughness and adhesion of Streptococcus mutans ATCC 25175 standard strain to bulk-fill resin composites. Results: The type of restorative materials did not affect the surface roughness or bacterial adhesion (p > 0.05) but the polishing systems were significant (p R = 0.943) between surface roughness and bacterial adhesion to the tested surfaces. Conclusion: Regardless of the restorative material, Mylar polishing system revealed the smoothest surface and the lowest adhesion of S. mutans as compared to Pogo one step and Sof-Lex multi-step polishing systems.

Caries is a disease of human dentition characterized by the loss of mineralized surfaces of the tooth;it is an infectious disease of the oral cavity in which biofilms play a causative role. Control of biofilms has traditionally relied on non-specific removal of plaque by mechanical means such as brushing, although it is difficult to remove biofilms by this method. Caries is also a widespread infection in children. Streptococcus mutans and S. sobrinus are important causative agents of caries. They produce a homologous exocellular polysaccharide called glucan, which strongly adheres to the enamel surface. This is a review of oral microbial biofilm formation by S. mutans and other related bacteria.

The prevalence of virulent clonal strains of mutans streptococci in vivo and co-culture succession of the strains in vitro—Virulence potential of mutans streptococci

Purpose:To examine selected putative virulent prop-erties of mutans streptococci in two groups with dif-ferent caries activity and to examine co-culture hy-bridization of the strains in vitro. Methods: A set of strains from caries-free subjects (115) and another set from caries-active subjects (165) were isolated. Each strain was characterized for three virulence determi-nants. The clinical bacteria were then cocultured by three strains exhibiting the highest levels of virulence. Isolate colonies of last filial generation bacteria were enrichment-incubated and estimated for virulence again. RAPD-PCR and MLEE analyses were processed for parental bacteria and last filial genera-tion one. Results: No difference associated with caries activity of the subjects from whom the isolates origi-nated. Virulent properties of a filial generation strains was not different in the same generation, but was very different from their parent strains. Conclu-sion: The coexist properties of virulent polyclonal strain of MS may hold in a very general conditional sense in a dental plaque ecosystem in vivo, however, one of the co-culture strains may became dominant and displa- ced the others as the result of continuous ecological succession in vitro.

Ozone action on Streptococcus mutans and Lactobacillus fermentum : A pilot study

AIM: To study the effectiveness of ozone in the elimination of cariogenic bacteria, followed with fluoride ***: Sixty extracted teeth free of caries were used, and five groups were constituted. In Group Ⅰ,the teeth were immersed in artificial saliva. In Group Ⅱ,the teeth were inoculated with Streptococcus mutans(S. mutans) and immersed in artificial saliva. In GroupⅢ the teeth were inoculated with Lactobaccilus fermentum(L. fermentum) and immersed in artificial *** Group Ⅳ the teeth were inoculated with S. mutans and L. fermentum and immersed in artificial saliva and the teeth in Group Ⅴ were inoculated with S. mutans and L. fermentum, and were subjected to the application of ozone and to the action of a fluoride mineralizing gel. DIAGNOdent was used to evaluate the caries of the teeth 3 wk after inoculation of bacteria and after that the teeth of Group Ⅴ were subjected to the application of ozone during 60 s, by HealO zone. After the application of ozone, products of the remineralization kit supplied by the manufacturer were applied daily, during 30 d. At the end samples were collected for analysis and evaluation of bacterial activity by polymerase chain reaction. RESULTS: Regarding the value of caries, obtained via DIAGNOdent, in the initial measurement the groups are homogeneous(P = 0.730). Synthesis and evaluation of a novel antibacterial dental resin composite with quaternary ammonium salts

The novel quaternary ammonium bromide (QAB)-containing oligomers were synthesized and applied for developing an antibacterial resin composite. Compressive strength (CS) and S. mutans (an oral bacteria strain) viability were used to evaluate the mechanical strength and antibacterial activity of the formed composites. All the QAB-modified resin composites showed significant antibacterial activity and mechanical strength reduction. Increasing chain length and loading significantly enhanced the antibacterial activity but dramatically reduced the CS as well. The 30-day aging study showed that the incorporation of the QAB accelerated the degradation of the composite, suggesting that the QAB may not be well suitable for development of antibacterial dental resin composites or at least the QAB loading should be well controlled, unlike its use in dental glass-ionomer cements. The work in this study is beneficial and valuable to those who are interested in studying antibacterial dental resin composites.
