Uncovering the multi-target pharmacological mechanism of Xuebijing injection against sepsis by a systems pharmacology approach

Sepsis is a life-threatening organ dysfunction that is associated with a high risk of ***(XBJ)injection,a Chinese herbal compound preparation,has been widely used for the treatment of sepsis in *** purpose of this research is to decipher the underlying multi-target pharmacological mechanism of XBJ in the treatment of sepsis using a systems pharmacology *** in XBJ were collected by literature *** corresponding putative targets of XBJ were screened from the Traditional Chinese Medicine Systems Pharmacology(TCMSP),Swiss target Prediction(STP),and Search Tool for Interacting Chemicals(STITCH)***-related targets were summarized using the Genecards,DrugBank,and Online Mendelian Inheritance in Man(OMIM)*** intersection targets were obtained with Venny ***,protein-protein interaction(PPI)networks were constructed with the STRING database and Cytoscape ***,degree,betweenness,and closeness were calculated to recognize the core targets in the PPI ***,the pharmacological mechanism of XBJ against sepsis was predicted via gene ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)*** the literature review,the 33 most commonly cited chemical ingredients,were screened out as major *** analysis revealed that the major compounds of XBJ modulated 218 common targets associated with *** PPI network analysis,41 genes,including IL-6,AKT1,STAT3,TP53,and MAPK1,were identified as core *** results of GO and KEGG enrichment revealed that the potential biological functions of XBJ against sepsis were mainly involved in cytokine receptor binding,cytokine activity,growth factor receptor binding,growth factor activity,and chemokine *** crucial pathways were closely associated with initial immune activation(CLR/TLR4-NF-κB/MAPK pathway),the acute inflammatory response(TNFMAPK/caspase and IL-MAPK/STAT pathways),and the late inflammation

The role of copper element has been an increasingly relevant topic in recent years in the fields of human and animal health, for both the study of new drugs and innovative food and feed supplements. This metal plays an important role in the central nervous system, where it is associated with glutamatergic signaling, and it is widely involved in inflammatory processes. Thus, diseases involving copper(Ⅱ) dyshomeostasis often have neurological symptoms, as exemplified by Alzheimer's and other diseases(such as Parkinson's and Wilson's diseases). Moreover, imbalanced copper ion concentrations have also been associated with diabetes and certain types of cancer, including glioma. In this paper, we propose a comprehensive overview of recent results that show the importance of these metal ions in several pathologies, mainly Alzheimer's disease, through the lens of the development and use of copper chelators as research compounds and potential therapeutics if included in multi-target hybrid drugs. Seeing how copper homeostasis is important for the well-being of animals as well as humans, we shortly describe the state of the art regarding the effects of copper and its chelators in agriculture, livestock rearing, and aquaculture, as ingredients for the formulation of feed supplements as well as to prevent the effects of pollution on animal productions.

The more information obtained about the driving environment,the more ensures driving *** to the complex driving environment of farmland roads,targets beside the road sometimes have an important impact on driving *** achieve this goal,a novel real-time detection and prediction algorithm of targets was *** whole image was divided into four parts by RCM:driving region,crossroad region,roadside region,and the other *** addition,a safety policy for every part was enforced by the algorithm,which was based mainly on the combination of the YOLACT and *** this basis,a self-collected data set of 5000 test samples is used for *** detection accuracy of the algorithm in the data set could reach up to 90%,and the processing speed to 30.4 *** addition,experiments were carried out on actual farmland roads,and the results showed that the proposed algorithm was able to detect,track,and predict targets on the farmland road,and alarm to driver in time before the targets rush into the *** study provides an important reference for the safe driving of agricultural vehicles.

This paper presents augmented input estimation(AIE)for multiple maneuvering target ***-target tracking(MTT)is based on two main parts,data association and *** data association(DA),the best observations are assigned to the considered *** real conditions,the number of observations is more than targets and also locations of observations are often so scattered that the association between targets and observations cannot be done *** this case,for general MTT problems with unknown numbers of targets,we present a Markov chain Monte-Carlo DA(MCMCDA)algorithm that approximates the optimal Bayesian filter with low complexity in *** DA,estimation and tracking should be *** in general cases,many targets can have maneuvering motions,then AIE is proposed to cover both the non-maneuvering and maneuvering parts of motion and the maneuver detection procedure is *** model with an input estimation(IE)approach is a special augmentation in the state space model which considers both the state vector and the unknown input vector as a new augmented state *** comparisons based on the Monte-Carlo simulations are also made to evaluate the performances of the proposed method and other older methods in MTT.

Radio-frequency(RF) tomography is an emerging technology which derives targets location information by analyzing the changes of received signal strength(RSS) in wireless links. This paper presents and evaluates a novel RF tomography system which is capable of detecting and tracking a time-varying number of targets in a cluttered indoor environment. The system incorporates an observation model based on RSS attenuation histogram and a multi-target tracking-by-detection filtering approach based on probability hypothesis density(PHD) filter. In addition, the sequential Monte Carlo method is applied to implement the multi-target filtering. To evaluate the tracking system, the experiments involving up to 3 targets were performed within an obstructed indoor area of 70 m2. The experimental results indicate that the proposed tracking system is capable of tracking a time-varying number of targets.

Background:Tumor microenvironment plays an essential role in the growth of *** how tumor cells co-evolve with tumor-associated immune cells and stromal cells is important for tumor ***:In this paper,we propose a logistic population dynamics model for quantifying the intercellular signaling nebvork in non-small-cell lung cancer (NSCLC).The model describes the evolutionary dynamics of cells and signaling proteins and was used to predict effective receptor targets through combination strategy ***,we optimized a multi-target strategy analysis algorithm that was verified by applying it to virtual patients with heterogeneous ***,to deal with acquired resistance which was commonly observed in patients with NSCLC,we proposed a novel targeting strategy-tracking targeted therapy,to optimize the treatment by improving the therapeutic strategy ***:The synergistic effect when inhibiting multiple signaling pathways may help significantly retard carcinogenic processes associated with disease progression,compared with suppression of a single signaling *** traditional treatment (surgery,radiotherapy and chemotherapy) tends to attack tumor cells directly,the multi-target therapy we suggested here is aimed to inhibit the development of tumor by emasculating the relative competitive advantages of tumor cells and promoting that of normal ***:The combination of traditional and targeted therapy,as an interesting experiment,was significantly more effective in treatment of virtual patients due to a clear complementary relationship between the two therapeutic schemes.

The basic unit in life is cell.?It contains many protein molecules located at its different organelles. The growth and reproduction of a cell as well as most of its other biological functions are performed via these proteins. But proteins in different organelles or subcellular locations have different functions. Facing?the avalanche of protein sequences generated in the postgenomic age, we are challenged to develop high throughput tools for identifying the subcellular localization of proteins based on their sequence information alone. Although considerable efforts have been made in this regard, the problem is far apart from being solved yet. Most existing methods can be used to deal with single-location proteins only. Actually, proteins with multi-locations may have some special biological functions that are particularly important for drug targets. Using the ML-GKR (multi-Label Gaussian Kernel Regression) method,?we developed a new predictor called “pLoc-mGpos” by in-depth extracting the key information from GO (Gene Ontology) into the Chou’s general PseAAC (Pseudo Amino Acid Composition)?for predicting the subcellular localization of Gram-positive bacterial proteins with both single and multiple location sites. Rigorous cross-validation on a same stringent benchmark dataset indicated that the proposed pLoc-mGpos predictor is remarkably superior to “iLoc-Gpos”, the state-of-the-art predictor for the same purpose.?To maximize the convenience of most experimental scientists, a user-friendly web-server for the new powerful predictor has been established at http://***/pLoc-mGpos/, by which users can easily get their desired results without the need to go through the complicated mathematics involved.
