Genetic Diversity of Main Inbred Indica Rice Varieties Applied in Guangdong Province as Revealed by molecular marker

Genetic diversity of 299 inbred indica rice varieties, including 33 introduced varieties, applied in Guangdong Province of China were assessed using 20 ILP （intron length polymorphism） and 34 SSR （simple sequence repeat） markers. Totally, 154 loci were screened for the 299 varieties, with the average number of alleles （Na）, rare alleles （Nr）, and polymorphism information content （PIC） scored at 3.4, 0.7 and 0.32, respectively. The Nei＇s genetic distance （GD） was estimated ranging from 0 to 0.7529 with an average of 0.4797. There was no significant difference of Na, Nr, PIC or GDs between the introduced and local varieties. Neighbor-joining （N J） analysis showed that the 299 varieties failed into three main distinct groups, and the 33 introduced varieties were distributed over all the groups or subgroups. Model-based cluster analysis demonstrated that only 73 （24.4%） of the 299 varieties and 7 （21.2%） of the 33 introduced varieties could be distinctly classified into the three groups. Analysis of molecular variance showed that within the groups divided by NJ analysis, the genetic variations revealed by ILP, SSR and these two combined were 7.7%, 5.6% and 6.6%, and within the groups divided by region （Guangdong local and the introduced varieties）, the genetic variables were 2.1%, 4.6%, 5.4%, respectively. These results suggested that the genetic diversity of the 299 inbred rice varieties in Guangdong Province was low, simultaneously relationship among varieties was poor and close in all kind of groups. Hence, it is very necessary to extend the genetic diversity during the breeding and selection practical procedure.

Morphological comparison and molecular marker screening of three Skeletonema species found in Changjiang(Yangtze)River Basin

In our recent investigations of diatom diversity,we studied three species,namely,Skeletonema costatum,Skeletonema subsalsum,and Skeletonema *** they have been found frequently in Changjiang(Yangtze)River Basin,their morphological and molecular identification is difficult in ***,to integrate morphological and molecular biological approaches,we compared systematically their morphological characters and performed phylogenetic *** strains of Skeletonema were collected and isolated from Shanghai and Jiangsu,China,and their morphological characteristics were examined by light microscopy(LM)and the scanning electron microscopy(SEM).Based on morphological comparison,we determined that *** is easy to distinguish from the other two *** heavily silicified areolae,undulated or cleft distal ends of terminal fultoportula processes(TFPPs),absence of basal pores of fultoportula processes(FPPs),the rootlike protrusions of FPPs,and no interlocking connection are the stable characteristics that can be used to identify ***,there are only two features that can distinguish *** from ***,namely the location of terminal rimoportulae(TRPs)and the distal shape of *** addition,we amplified and sequenced nine common genetic markers from the strains,from which 101 sequences were obtained,constructed phylogenetic trees based on the nine genes and evaluated that seven genes can be used to identify ***,and revealed that *** is the closest known relative of ***,and only ATP synthetase beta-subunit gene(atp B)is able to distinguish them from each other,which strongly support that it is an effective molecular marker for *** work provided a theoretical basis for the taxonomic study of Skeletonema.

The sea potato Acaudina *** a species of sea cucumber,belonging to the class Holothuroidea under Phylum *** becomes blooming recently in the East Sea of China,causing serious ecological ***,until now there are no molecular data for its larval identification and population genetic *** this study,we firstly screened a mtDNA fragment and demonstrated that it was the species-specific molecular marker for the identification of Acaudina *** then developed a quantitative polymerase chain reaction(PCR)method to evaluate the larval density of Acaudina *** on this molecular *** this method,we examined 116 plankton samples collected in four seasons from 13 stations along the coastal region in Fu-jian province,*** results showed that the high larval density was presented at stations 1,2,and 3,which were near a quay in the *** larval density increased from April and reached the highest value in June and July,suggesting temperature might be the main environmental factor on the effects of its population distribution and *** work provides an important molecular tool for species identification and risk evaluation of a potentially invasive species.

Development of molecular marker Linked with Cercospora Leaf Spot (CLS) Disease Resistance in Vigna radiata, Cloning, and Expression for Evaluating Antifungal Activity against Cercospora canescens

We developed a molecular marker for MAS of mungbean resistant varieties against CLS from the consensus sequence(MB-CLsRG)of identified RGAs(MB-ClsRCaG1 and MB-ClsRCaG2).The MB-CLsRG sequence-specific primer pair was used to screen Cercospora leaf spot(CLS)resistant varieties of mungbean in genomic analysis that showed congruency with phenotypic *** of molecular marker linkage with CLS resistance was performed using rtPCR in transcriptomic *** sequenced PCR products showed 100%homology with MB-CLsRG sequence and putative disease resistance proteins that confirmed the linkage of molecular marker with CLS resistance in *** antifungal potential of MB-CLsRG gene encoding protein was *** MB-CLsRG gene sequence was cloned in the *** expression vector for recombinant protein *** recombinant protein was then investigated for its in vitro antifungal potential against Cercospora *** in vitro investigation showed strong antifungal activity of recombinant protein as it restricted the growth of fungal mycelial *** results validated the linkage of developed marker with CLS-resistant mungbean varieties;therefore,it can be used to screen resistant varieties from a large population in ***,the recombinant protein of the MB-CLsRG gene sequence revealed antifungal potential,which proved the gene sequence could be suitable to use in transgenic plants technology to develop fungal-resistant transgenic crops.

Mapping Quantitative Trait Loci Controlling Endosperm Traits with molecular marker

Based on the genetic models for triploid endosperm traits and on the methods for mapping diploid quantitative traits loci (QTLs), the genetic constitutions, components of means and genetic variances of QTL controlling endosperm traits under flanking marker genotypes of different generations were presented. From these results, a multiple linear regression method for mapping QTL underlying endosperm traits in cereals was proposed, which used the means of endosperm traits under flanking marker genotypes as a dependent variable, the coefficient of additive effect (d) and dominance effect (h1 and/or h2) of a putative QTL in a given interval as independent variables. This method can work at any position in a genome covered by markers and increase the estimation precision of QTL location and their effects by eliminating the interference of other relative QTLs. This method can also be easily used in other uneven data such as markers and quantitative traits detected or measured in plants and tissues different either in generations or at chromosomal ploidy levels, and in endosperm traits controlled by complicated genetic models considering the effects produced by genotypes of both maternal plants and seeds on them.

Identification of molecular marker Linked to Salt Tolerance Gene in Alfalfa

The study has established the F2 offspring obtained by crossing salt-tolerant with salt-sensitive alfalfa, and appraised the salt-tolerant F2 offspring seedling was evaluated in pot culture. With the F2 segregated population, the research has obtained a molecular marker linked with salt-tolerant genes of alfalfa using the improved BSA combined with RAPD. The RAPD PCR products were excised from the agarose gel and purified using a kit, then were mixed with pMD-18T vector and sequenced. Sequencing result indicated the RAPD marker was 1 438 bp in length. Similarity researches using blast in Genbank indicated that the nucleotide sequence of the RAPD marker showed 93% and 91% similarity with mth2-6el8 gene fragment （347 bp） and ruth2-33122 gene fragment （334 bp） of Medicago truncatula respectively. Medicago truncatula is a close relative of alfalfa and Mth2-6el8 is a molecular marker of the gene coding for a cysteine protease which was saltinducible in some plants. These results indicated the RAPD marker was possibly related to cysteine protease genes in alfalfa.

molecular structure of the marker of tumour is determined by magneto-optical analysis of blood serum. The marker is the laevorotatory enantiomer of alanine. The cancer status of a subject is described by the number of molecules of the laevorotatory alanine enantiomer (-)ρ and the effectiveness of therapy is measured by the number of molecules of the dextrorotatory alanine enantiomer (+)ρ. The values of (-)ρ and (+)ρ are determined separately for the patient before and after therapy.

Designing a SCAR molecular marker for monitoring Trichoderma cf. harzianum in experimental communities

Several species of the fungal genus Trichoderma establish biological interactions with various micro- and macro-organisms. Some of these interactions are relevant in ecological terms and in biotechnological applications, such as biocontrol, where Trichoderma could be considered as an invasive species that colonizes a recipient community. The success of this invasion depends on multiple factors, which can be assayed using experimental communities as study models. Therefore, the aim of this work is to develop a species-specific sequence-characterized amplified region （SCAR） marker to monitor the colonization and growth of T. cf. harzianum when it invades experi- mental communities. For this study, 16 randomly amplified polymorphic DNA （RAPD） primers of 10-mer were used to generate polymorphic patterns, one of which generated a band present only in strains of T. cf. harzianum. This band was cloned, sequenced, and five primers of 20-23 mer were designed. Primer pairs 2F2/2R2 and 2F2/2R3 successfully and specifically amplified fragments of 278 and 448 bp from the T. cf. harzianum BpT10a strain DNA, respectively. Both primer pairs were also tested against the DNA from 14 strains of T. cf. harzianum and several strains of different fungal genera as specificity controls. Only the DNA from the strains of T. cf. hat-zianum was successfully amplified. Moreover, primer pair 2F2/2R2 was assessed by quantitative real-time polymerase chain reaction （PCR） using fungal DNA mixtures and DNA extracted from fungal experimental communities as templates. T. cf. harzianum was detectable even when as few as 100 copies of the SCAR marker were available or even when its population represented only 0.1% of the whole community.

The spiralling whitefly,Aleurodicus disperses Russell,is a invasive pest worldwide. Morphological identification of the pest is very difficult due to the small size and high degree similarity with other *** this study,the method for rapid detection of *** was established based on CO I gene sequence,one pair of primers which are specific to spiralling whitefly was *** fragment amplified by the primers was 399 bp in *** tests performed with this pair of primers showed that all specimens of *** were *** this method was tested on a single male and female adult,a single nymph(1st to 4th instar),even a single egg,,and proved to be applicable for these stages of *** ***,When the dilution was 1/4000 of a whole female adult of ***,the 399 bp DNA fragment could be identified for all *** establishment of the method for rapid detection of *** provided a technical support in prevention spread of the spiralling whitefly in China and in reduction of its harmfulness.
