Subpopulation genetic structure of a plant panmictic population of Castanea sequinii as revealed by microsatellite markers

Castanea squinii Dode,an endemic tree widely distributed in China,plays an important role both in chestnut breeding and forest ecosystem *** spatial genetic structure within and among populations is an important part of the evolutionary and ecological genetic dynamics of natural populations,and can provide insights into effective conservation of genetic *** the present study,the spatial genetic structure of a panmictic natural population of *** in the Dabie Mountain region was investigated using microsatellite *** prescreened microsatellite loci generated 29-33 alleles each,and were used for spatial autocorrelation *** on Moran’s I coefficient,a panmictic population of *** in the Dabie Mountain region was found to be lacking a spatial genetic *** results suggest that a high pollen-mediated gene flow among subpopulations counteract genetic drift and/or genetic differentiation and plays an important role in maintaining a random and panmictic population structure in *** ***,a spatial genetic structure was detected in each subpopulation’s scale(0.228 km),with all three subpopulations showing significant fine-scale *** genetic variation was found to be nonrandomly distributed within 61 m in each subpopulation(Moran’s I positive values).Although Moran’s I values varied among the different subpopulations,Moran’s I in all the three subpopulations reached the expected values with an increase in distances,suggesting a generally patchy distribution in the *** fine-scale structure seems to reflect restricted seed dispersal and microenvironment selection in *** results have important implications for understanding the evolutionary history and ecological process of the natural population of *** and provide baseline data for formulating a conservation strategy of Castanea species.

A Modified Enrichment Method to Construct microsatellite Library from Plateau Pika Genome(Ochotona curzoniae)

A microsatellite-enriched library of plateau pika （Ochotona curzoniae） was constructed according to the strong affinity between biotin and streptavidin. Firstly, genomic DNA was fragmented by ultrasonication, which is a major improvement over traditional methods. Linker-ligated DNA fragments were hybridized with biotinylated microsatellite probes, and then were subjected to streptavidin-coated magnetic beads. PCR amplification was performed to obtain double-stranded DNA fragments containing microsatellites. Ligation and transformation were carried out by using the pGEM-T Vector System I and Escherichia coli DH10B competent cells. Sequencing results showed that 80.2% of clones contained microsatellite repeat motif. Several modifications make this protocol time-efficient and technically easier than the traditional ones; particularly, composition and relative abundance of microsatellite repeats in plateau pika genome were truly represented through the optimized PCR conditions. This method has also been successfully applied to construct microsatellite-enriched genomic libraries of Chinese hamster （Cricetulus griseus） and small abalone [Haliotis diversicolor （Reeve）] with high rates of positive clones, demonstrating its feasibility and stability.

Grass carp is one of the most important cultured fishes all over the *** genetic diversity of grass carp plays an important role whatever in selective breeding progress or ecological conservation ***,some genetic diversity researches were not accuracy and cannot be compared with each other due to different molecular markers,sample size and detection *** this study,we constructed five multiplex PCR assays contained 20 microsatellites with highly polymorphism and heterozygosity for evaluating genetic information of grass *** used nine cultured populations consisting of 507 individuals to detect stability of the five multiplex PCR *** results showed that the number of alleles(Na),effective number of alleles(Ne),observed heterozygosity(Ho),expected heterozygosity(He)and polymorphism information content(PIC)of the 20 microsatellite loci were relative high compared with the genetic diversity parameters of microsatellite loci developed by other *** loci were significantly deviated from Hardy-Weinberg equilibrium(P<0.01).And with exception of the Shaoguan,Indian and Nepal cultured population,all other cultured populations showed very high genetic *** the test of grass carp populations,we developed an effective and accurate multiplex SSR-PCR assays that can be as statistical powerful tool for detecting genetic information of grass carp.

The microsatellite-enriched library was constructed using magnetic bead hybridization selection method, and the mi-crosatellite DNA sequences were analyzed in Pacific abalone Haliotis discus hannai. Three hundred and fifty white colonies were screened using PCR-based technique, and 84 clones were identified to potentially contain microsatellite repeat motif. The 84 clones were sequenced, and 42 microsatellites and 4 minisatellites with a minimum of five repeats were found (13.1% of white colonies screened). Besides the motif of CA contained in the oligoprobe, we also found other 16 types of microsatellite repeats including a dinucleotide repeat, two tetranucleotide repeats, twelve pentanucleotide repeats and a hexanucleotide repeat. According to Weber (1990), the microsatellite sequences obtained could be categorized structurally into perfect repeats (73.3%), imperfect repeats (13.3%), and compound repeats (13.4%). Among the microsatellite repeats, relatively short arrays (microsatellites was 48 repeats, and the average number of repeats was 13.4. The data on the composition and length distribution of microsatellites obtained in the present study can be useful for choosing the repeat motifs for microsatellite isolation in other abalone species.

Novel Poncirus trifoliata simple sequence repeat(SSR) markers were developed to evaluate their utility for genetic diversity and breeding studies of P. trifoliata and related species. A total of 108 primer pairs were characterized by PCR amplification experiments. Among these, 61 were polymorphic and transferable to other citrus species. The number of alleles per locus ranged from 2 to 6, with an average of 2.37 alleles per locus. The expected heterozygosity and observed heterozygosity ranged from 0 to 0.83 and 0 to 1.00, *** novel polymorphic SSR markers will be useful for citrus cultivar identification and evaluation as well as breeding studies.

Five microsatellites, IDVGA-2, IDVGA-27, IDVGA-46, IDVGA-55 and TGLA-44,were analyzed for polymorphisms in beef cattle. The number of alleles and polymorphism information content (PIC) values were 12/0. 82, 5/0. 58, 8/0. 70, 6/0. 57 and 11/0. 86 respectively. Each microstellite was typed on a half-sib family in order to verify the segregation of the alleles.

With the development of satellite miniaturization and remote sensing,the establishment of microsatellite constellations is an inevitable *** to their limited size,weight,and power,spaceborne storage systems with excellent scalability,performance,and reliability are still one of the technical bottlenecks of remote sensing *** on the commercial off-the-shelf field-programmable gate array and memory devices,a spaceborne advanced storage system(SASS)is proposed in this *** work provides a dynamic programming,queue scheduling multiple-input multiple-output cache technique and a high-speed,high-reliability NAND flash controller for multiple microsatellite payload *** results show that SASS has outstanding scalability with a maximum write rate of 2429 Mb/s and preserves at least 78.53%of the performance when a single NAND flash *** scheduling technique effectively shortens the data scheduling time,and the data remapping method of the NAND flash controller can reduce the retention error by at least 50.73%and the program disturbance error by at least 37.80%.

microsatellite-Based Genetic Structure and Differentiation of Goldfish (Carassius auratus) with Sarcoma

Ten microsatellite loci were used for analyzing six populations of goldfish (Carassius auratus) with sarcoma. It showed that there was the highest genetic diversity among the white oranda with red cap (RC) population, and the lowest among the white tigerhead (WT) population. However, the outcross existed among every population. There was huge genetic differentiation between WT and the other four populations. The average observed heterozygosity (HO) among populations ranged from 0.3571 to 0.7381. And significant genetic difference (FCT = 0.1891, P = 0.0186) appeared among goldfish varieties which can be classified into three groups (RT, WT;RL, BL, YC;RC). The software Principal Coordinate Analysis (PCA) and STRUCTURE showed that significant genetic differences were revealed between RC population of goldfish and other five populations.
