MicroRNA-99a mimics inhibit M1 macrophage phenotype and adipose tissue inflammation by targeting TNFα

In human adipose tissue and obesity,miR-99a expression is negatively correlated with ***,the present study investigated the role of miR-99a in macrophage phenotype activation and adipose tissue inflammation.M2 BMDMs showed a significant increase in miR-99a expression when compared to the M0 and M1 ***-switching experiments established an association between upregulated miR-99a expression and the M2 *** of miR-99a prevented M1 phenotype activation and attenuated bactericidal ***,knockdown of miR-99a abolished M2 phenotype *** means of in silico target prediction tools and a luciferase reporter assay,TNFαwas identified as a direct target of *** of TNFαrecapitulated the effect of miR-99a overexpression in M1 *** a db/db mice model,miR-99a expression was reduced in eWAT and F4/80+*** overexpression of miR-99a in db/db mice attenuated adipocyte hypertrophy with increased CD301 and reduced CD86 *** cytometry analysis also showed an increased M2 and a reduced M1 macrophage *** of miR-99a also improved the diabetic dyslipidemia and insulin signaling in eWAT and liver,with an attenuated expression of gluconeogenesis and cholesterol metabolism genes in the ***,adoptive transfer of miR-99a-overexpressing macrophages in the db/db mice recapitulated in vivo miR-99a mimic effects with increased M2 and reduced M1 macrophage populations and improved systemic glucose,insulin sensitivity,and insulin signaling in the eWAT and *** present study demonstrates that miR-99a mimics can regulate macrophage M1 phenotype activation by targeting TNFα.miR-99a therapeutics in diabetic mice reduces the adipose tissue inflammation and improves insulin sensitivity.

In the latest issue of Nature Immunology,Piccolo et al.1 elegantly explored the effect of coexisting interferon-γ(IFN-γ)and interleukin 4(IL-4)antagonistic signals on macrophage transcriptional and epigenetic profiles and,interestingly,identified a plastic cross-talk as opposed to mutually exclusive programs between M1 and M2 polarized *** results support the fascinating hypothesis that transcriptional and epigenetic reprogramming overcame genetics to drive the evolution of eukaryotic organisms.A rapid reshaping of chromatin acetylation redirected the expression of hundreds of genes under the control of a few transcription factors in response to two coexisting but opposing signals that indicated inflammation and its resolution simultaneously.

Prostate cancer(PCa)is considered as an age-related disease and accounts for the most prevalence of urinary malignancies in men.1 We previously analyzed the cellular landscape of tumor microenvironment in PCa patients,where we found that cancer-related fibroblasts might play an important role in tumorigenesis and the development of PCa and tumor-associated macrophage(TAM)could function synergistically with them.2 Thus,we further integrated single-cell and bulk RNA sequencing with meta-analysis to specify the prognostic effect of TAM on PCa and construct molecular subgroups and predictive index to guide clinical practice.

Establishment and characterization of murine bone marrow stromal macrophage line

The authors established a murine bone marrow stromal cell line QXMSC1 by discarding the suspensible hematopoietic cells and passaging the adherent stromal cells many times in long_term bone marrow culture. QXMSC1 cells have been passaged 65 times for 15 months and immortalized. The mean number of QXMSC1 chromosomes is 66±4. The appearance of the cells is elliptical. There are many pseudopods and mononuclei under the optical microscope. Many lisosomes and phagosomes in cytoplasm exist under the transmission electron microscope. There are no desmosome junction between the cells, no lipid drops, no intermediate filament. Vimentin is positive and keratin is negative by stain of immunocytochemistry. Nonspecific lipase staining is positive. These results indicate that QXMSC1 cells are murine bone marrow macrophages.

Cy5.5-MSA-G250 nanoparticles(CMGNPs)had been proved to have unique advantages for cancer treatment,including excellent photothermal performance,tumor cell-selective cytotoxicity,direct visualization,and good ***,to cellular systems,the CMGNPs are considered as fo reign invaders,and the effect of CMGNPs on immunity system is still ***,more efforts are needed to understand the role of CMGNPs on the immunity *** this study,we attempted to screen the pro-inflammatory responses on RAW264.7 macrophages after treated with the *** vitro experiments clearly showed that CMGNPs not only enhances phagocytosis capacity of RAW264.7 cells,but also promotes Ml polarization,associated with changes in cell morphology and increased expression of inflammatory *** ability to induce Ml polarization may be beneficial to CMGNPs to achieve better anticancer effects in clinical ***,the observed Ml macrophages’ polarization triggered by CMGNPs can be abolished after adding TLR4 inhibitor,CLI095,suggesting that TLR4 is involved in CMGNP-induced inflammation.
