Activity Determination of 8 Chinese Herbs against Hepatoma Cell SMMC-7721 in Vitro by mtt method

[Objective] The aim was to build up a set of efficient and rapid models for laboratory to screen anti-hepatocellular carcinoma active substance in vitro. [ method] By using mtt method, the activities of anti-hepatocellular carcinoma SMMC-7721 in vitro from Cymbopogon distans, Lobelia chinensis, Buddleja offlcinalis, Glycyrrhiza uralensis, Sanguisorba officinalis, Bupleurum chinense, Apium graveolen and Curuma zedoaria were tested. The growth curve of hepatoma cell was described, and the growth status in different periods were observed by inverted microscope. [ Result] Cells induced by active substance would be condensing, clear brim, which have significant differences from normal SMMC- 7721 cells. The results suggested that ESCG, ESCC, ESCB could inhibit proliferation of SMMC-7721 cells at the concentration of 1.0 -1.5 mg/ml, and the inhibition rate were 51.6%, 48.5%, 52.9% respectively. With the increasing of concentration, the inhibition strengthened. [ Conclusion] mtt method could be used as a basic model for screening important anti-hepatoma.

EFFECT OF BUNGARUS MULTICINCTUS CRUDE VENOM AND ITS COMPONENTS ON TUMOR CELLS

Objective: To confirm whetherBungarus multicinctus crude venom induces the apoptosis of K562 tumor cells and to find out the components inducing apoptosis of K562 cells from the crude venom. methods: the crude venom separated and purified by cation exchange chromatography, and the effect of venoms on K562 was studied by mtt method and flow cytometry. Results: The crude venom began to kill K562 cells at than 8×l03ng/ml (the survival rate was 82.5%) concentration and the effect was more significant in 24 h when administrating 8×l05ng/ml (the survival rate was 29.4%) crude venom. Apoptotic bodies were observed in the K562 tumor cells by fluorescent microscopy after administration of 5 μg/ml cycloheximide (CHX) or the peak VI solution at about 8×105 ng/ml. The same results were detected by the flow cytometry. A sub-Gi peak appeared after administration of CHX or the sixth peak solution. Conclusion: The authors found that the venom can kill K562 tumor cells in time- and dose-dependent manner. However, the killing effect of the venom is not apoptosis. What’s more, the peak VI solution, a component of the crude venom can induce the apoptosis of K562 tumor cells.

Studies on Antivirus Effects of Polysaccharides from Sargassum fusiforme in vitro

　　To assay antiviral effects of the polysaccharide and its separated products from Sargassumfusiforme on HSV-1 and CVB3 and their antiviral mechanism, the antiviral effects of all samples on HSV-1 and CVB3 and their cytotoxicity on vero cells were studied by CPE and mtt method. Antiviral mechanism of two samples was briefly studied by four different ways. The cytotoxicity of samples on vero cells was not detected (CC＞5000 μg/mL). All samples except PSJ had obviously HSV-1 inhibitory effect. Antiviral activities of them were increasingly strengthened with raising their *** activities of F1, F2 and F4 were better than that of ACV, and antiviral activities of all samples on CVB3 were better than ribavirin injection. Fi, F2 and F4 had remarkable anti-CVB3 activity. The experiment showed that polysaccharide was able to not only kill the above viruses directly but also restrain them by getting into cell or absorbing on cells.……

　　To assay the antiviral activities on HSV-1 and CVB3 in vitro of the polysaccharide from Eucheuma striatum, its antiviral mechanism was explored. Vero cells were infected by HSV-1 and CVB3, and they were cultured with serial dilutions of polysaccharide. The cells cytotoxicity of Polysaccharide was evaluated by the mtt method. The inhibitory effects were evaluated by the cytopathic effect (CPE). Its antiviral mechanism was researched by the method of giving samples in different time. The polysaccharide could inhibit the CPE of cells infected by HSV-1 and CVB3. It showed low cytotoxicity on vero cells. Its antiviral activities were better than those of acyclovir and ribavirin which were run in parallel as the positive control samples. The polysaccharide from Eucheuma striatum has potent antiviral activities. Its antiviral mechanism is that it can prevent the virus from absorbing to the cell surface.……

Synthesis and Anti-Cancer Activities of Resveratrol Derivatives

A novel series of resveratrol derivatives were synthesized according to Wittig-Horner reaction with 3,5-dihydroxybenzyl alcohol or 3,5-dimethoxybenzyl alcohol or 4-hydroxybenzyl alcohol as raw material and the inhibitory activities on breast carcinoma (MDA-MB-231) and gastric carcinoma cell lines (SGC-7901) in vitro were evaluated by the standard methyl thiazole tetrazolium (mtt) method. The result of biological test shows that some of resveratrol derivatives possess stronger anti-cancer activities than 5-FU. Compound 5c shows the strongest activity against breast carcinoma (MDA-MB-231) and gastric carcinoma cell lines (SGC-7901) with IC50 value of 50.19 ± 1.02 μM, 122.68.27 ± 2.04 μM, compared to that IC50 value of 5-FU is 98.59±3.61 μM,156.74±6.16 μM, respectively.
