Rationale: The known prognosis factors for rhabdomyosarcoma (RMS) in children do not always explain the unsatisfactory outcome of treatment. Changes in the subpopulation composition of Bone Marrow (BM) effector cells during the development of RMS may indicate new directions for the search for prognostic factors and points for the impact of targeted therapy. Purpose: To identify correlations between quantitative changes in the levels of subpopulations of T, B and NK-lymphocytes of BM and known risk factors for RMS in children. Objects: The study included 31 patients. The main group included 16 patients with RMS, average age—6.8 ± 1.0 years, while children 1 - 10 years old—13 (81.3%), over 10 years old—3 (18.8%) people, girls and boys were 8 people each. Background:To investigate the performance of a domestic TB-IGRA kit with IFN-y release assays in detecting Mycobacterium tuberculosis(MTB)infection and determine the role of lymphocyte subpopulations in peripheral blood in patients with MTB ***:A retrospective study was performed with 166 TB patients(68 pulmonary tuberculosis TB and 98 extra-pulmonary TB)and 377 Non-TB patients diagnosed in our hospital from January 2016 and May *** routine blood test,biochemical parameters,immunoglobulins and lymphocyte subpopulations data were collected from the medical records for statistical ***,specificity and Youden index was used to evaluate the performance of ***:Data showed that the IFN-y levels in patients with PTB and EPTB were 203.58±18.00 and 201.83±14.56,which were dramatically higher than that in patients with Non-TB(32.12±4.36,plymphocyte counts and the lymphocytes percentage in patients with PTB and EPTB other than Non-TB *** were no any differences in the absolute counts and proportions of lymphocyte subpopulations among PTB,EPTB and *** AUC area of IFN-y for TB diagnosis was 0.884,with sensitivity of 88.6%and specificity of 77.5%,when the recommended cut-off value was 14 pg/ml and the Youden index was ***,if the cut-off value was supposed to be 28 pg/ml,the sensitivity was 84.9% and the specificity would go up to 83.6% for TB diagnosis,with the utmost Youden index ***:The results showed that the domestic TB-IGRA kit had a high sensitivity and specificity and presented a good performance in detecting MTB infection.

Decrease of Peripheral Blood CD8+/CD28- Suppressor T Cell Followed by Dentritic Cells Immunomodulation among Metastatic Breast Cancer Patients

Objective： To explore the effects of dentritic cells on the peripheral blood lymphocyte subpopulations of metastatic breast cancer patients treated with chemotherapy. Methods： The current study involved 44 metastatic breast cancer patients treated with docetaxel-based chemotherapy. Among them, 25 cases were treated with dendritic cells derived from CD34＋ hematopoietic stem cells enriched autologous peripheral mononuclear cells after chemotherapy, and 19 cases received chemotherapy alone. Peripheral blood samples were collected from each patient before and after treatment, and lymphocyte subpopulations including CD3＋, CD3＋/CD4＋, CD3＋/CD8＋, CD3-/CD16＋56＋, CD3＋/CD16＋56＋, CD4＋/CD25＋, CD8＋/CD28-, CD8＋/CD28＋, CD4/CD8, DC1, DC2 and DC1/DC2 were analysed by a 3-color flow cytometric analysis. Results： The two treatment groups were well matched with regard to demographic and baseline disease characteristics. Comparing the changes of lymphocyte subpopulations between the two groups, it showed that the difference of the change of CD8＋/CD28-lymphocyte had statistic significance. The percentage of CD8＋/CD28-lymphocyte was lower in the chemotherapy＋DC group, but higher in the chemotherapy-alone group. Conclusion： As CD8＋/CD28-lymphocyte represent a kind of suppressive T lymphocyte, we conclude that dentritic cell therapy can relieve immunosuppression to some extent.
