losartan improves erectile function through suppression of corporal apoptosis and oxidative stress in rats with cavernous nerve injury

This study aimed to investigate the functional and morphological changes in the corpus cavernosum after cavernous nerve(CN)injury or neurectomy and then reveal whether treatment with the angiotensin II Type 1 receptor antagonist losartan would improve erectile function as well as its potential mechanisms.A total of 48 10-week-old Sprague-Dawley male rats,weighing 300-350 g,were randomly divided into the following four groups(n=12 per group):sham operation(Sham)group,bilateral cavernous nerve injury(BOND group,losartaretreated BCNI(BCNI+losartan)group,and bilateral cavernous neurectomy(Neurectomy)group.losartan was administered once daily by oral gavage at a dose of 30 mg kg^-1 day^-1 for 4 weeks starting on the day of surgery.The BCNI and the Neurectomy groups exhibited decreases in erectile response and increases in apoptosis and oxidative stress,compared with the Sham group.Treatment with losartan could have a modest effect on erectile function and significantly prevent corporal apoptosis and oxidative stress?The phospho-B-cell lymphoma 2(Bcl-2)-associated death promoter(p-Bad)/Bad and phospho-the protein kinase B(p-AKT)/AKT ratios were substantially lower,while the Bcl-2-associated X protein(Bax)/Bcl-2 ratio,nuclear factor erythroid 2-related factor 2(Nrf2)/Kelch-like ECH-associated protein 1(Keap-1),transforming growth factor-p 1(TGF-pi)and heme oxygenase-1(HO-1)levels,and caspase-3 activity were higher in the BCNI and Neurectomy groups than in the Sham group.After 4 weeks of daily administration with losartan,these expression levels were remarkably attenuated compared with the BCNI group.Taken together,our results suggested that early administration of losartan after CN injury could slightly improve erectile function and significantly reduce corporal apoptosis and oxidative stress by inhibiting the Akt/Bad/Bax/caspase-3 and Nrf2/Keap-1 pathways.

losartan Protects Podocytes against High Glucose-induced Injury by Inhibiting B7-1 Expression

The role of B7-1 in podocyte injury has received increasing attention.The aim of this study was to investigate whether losartan protects podocytes of patients with diabetic kidney disease(DKD)by regulating B7-1 and the underlying mechanisms.Rats with streptozotocin-induced DKD were treated with losartan for 8 weeks.Biochemical changes in blood and urine were analyzed.Kidneys were isolated for electron microscopy,immunofluorescence,real-time quantitative PCR(RT-PCR),and Western blot analysis.Immortalized mouse podocyte cells were cultured in normal or high glucose medium in the presence or absence of losartan for 48 h,and then the cells were collected for immunofluorescence,PCR,Western blotting and monolayer permeability detection.The phosphatidylinositol 3-kinase(PI3K)110a subunit and angiotensin II type 1 receptor(AT1R)plasmids were transfected into podocytes,respectively,and then Western blotting was performed to assess the expression of B7-1 protein.The results showed that losartan ameliorated podocyte structure and function in the rat model of DKD,and reduced the expression of B7-1 protein.Overexpression of PI3K 110a subunit in podocytes attenuated the inhibitory effect of losartan on B7-1 expression in high glucose-stimulated podocytes.The expression of B7-1 was significantly increased by overexpression of ATI R and significantly reduced by blocking PI3K 110a subunit.We conclude that losartan protects podocytes against high glucose-induced injury by inhibiting AT1R-mediated B7-1 expression.This effect is dependent on the AT1R-PI3K 110a subunit pathway.

Effects of losartan on acute atrial electrical remodeling

Background Atrial electrical remodeling (AER) contributes to the maintainance of atrial fibrillation (AF). This study was to compare the effects of losartan with those of Diltiazem on tachycardia-induced acute AER in rabbits Methods Twenty-one rabbits paced with maximal atrial capture rate for 3 hours in the right atrium (RA) were randomly divided into saline group, Diltiazem group and losartan group After autonomic blockage, we measured atrial effective refractory period (AERP), AERP rate adapting feature, AERP dispersion and RA conduction time at basic cycle lengths (BCLs) of 200 ms and 150 ms at baseline, 0 5 hour, 1 hour, 2 and 3 hours after rapid atrial pacing Results In the saline group, there was a prompt decrease in AERP as a result of rapid atrial pacing, and AERP 200 and AERP 150 were shortened sharply within 0 5 hour of pacing (30 2±10 5 ms and 24 1±9 1 ms, respectively) The AERP did not change dramatically in the Diltiazem and losartan groups In the saline group, the value of (AERP 200 -AERP 150 )/50 ms in high RA was 0 17±0 08 at baseline and became significantly smaller at 0 5 hour (0 08±0 06), 1 hour (0 09±0 06), 2 hours (0 08±0 04) and 3 hours (0 09±0.05) (all P losartan groups In the saline group, AERP dispersion increased significantly at 2 and 3 hours ( P losartan infusion, the AERP dispersion was no longer increased after rapid atrial pacing There was no significant difference in RA conduction time among the three groups Conclusion Like calcium antagonist Diltiazem, losartan could prevent AERP shortening and preserve rate adaptation of AERP after rapid atrial pacing losartan is more effective than Diltiazem in inhibiting the increase of AERP d

Study on the Effects of losartan on Cardiomyocyte Apoptosis and Gene Expression After Ischemia and Reperfusion in vivo in Rats

Summary: In order to study the effects of losartan on cardiomyocyte apoptosis following ischemia (0. 5 h) and reperfusion (48 h) in vivo and bcl-2 and bax gene expression, TUNEL staining method, immunohistochemistry and in situ hybridization histochemistry (ISHH) were used to monitor the apoptotic cells, mRNA and protein of gene expression, respectively. Image processing system was used to quantitively dispose the positive metric substance of both immunohistochemistry and ISHH through the average optical density (OD) value. The number of the apop- totic cells were 38±9 (control group), 0-1 (sham operation group) and 9±4 (losartan-treated group) in each visual field respectively with the difference among the groups being significant (Plosartan-treated group ) respectively, while OD values of bcl-2 (immunohistochemistry) were 0. 1374±0. 01367 (control group ), 0. 08510±0. 01862 (sham operation group) and 0. 1252±0. 02064 (losartan-treated group). hcl-2 gene expression was increased significantly in the control group and losartan-treated group as com- pared with sham operation group (P losartan-treated group). bax gene expression was decreased very significantly in losartan-treated group and sham operation group as compared with control group (Plosartan-treated group) respectively. The results indicated that losartan might inhibit cardiomyocyte apoptosis following ischemia and reperfusion. The mechanism might be that bax gene expression was inhibited to increase bcl-2/bax ratio.

Beneficial effects of losartan or telmisartan on the local hepatic renin-angiotensin system to counter obesity in an experimental model

BACKGROUND Obesity has been associated with hepatic overexpression of the renin-angiotensin system(RAS).AIM To evaluate the action of two angiotensin II(ANGII) receptor blockers(losartan or telmisartan) on the modulation of local hepatic RAS and the resulting metabolic effects in a diet-induced obesity murine model.METHODS Twenty C57 BL/6 mice were randomly divided into two nutritional groups for 10 wk: control group(C, n = 5, 10% of energy as fat) or high-fat group(HF, n = 15,50% of energy as fat). After treatment started, the HF group was randomly divided into three groups: untreated HF group(n = 5), HF treated with losartan(HFL, n = 5) and HF treated with telmisartan(HFT, n = 5). The treatments lasted for 5 wk, and the dose was 10 mg/kg body mass.RESULTS HF diet induced body mass gain(+28%, P losartan or telmisartan treatments, caused hepatic and metabolic beneficial effects as demonstrated by reduced hepatic triacylglycerol levels coupled with reduced PLIN 2 expression and improved glycemic control.

Investigating the potential of essential oils as penetration enhancer for transdermal losartan delivery: Effectiveness and mechanism of action

The effect of tea tree oil(TTO),cumin oil(CO),rose oil(RO)and aloe vera oil(AVO)on the skin permeation of losartan potassium(LP)was investigated.In vitro skin permeation studies were carried out using rat skin.The mechanism of skin permeation enhancement of LP by essential oils treatment was evaluated by FTIR,DSC,activation energy measurement and histopathological examination.Both concurrent ethanol/enhancer treatment and neat enhancer pretreatment of rat SC with all the oils produced significance increase in the LP flux over the control.The effectiveness of the oils as the penetration enhancers was found to be in the following descending order:AVO>RO>CO>TTO.However,only AVO was the only enhancer to provide target flux required to deliver the therapeutic transdermal dose of LP.FTIR and DSC spectra of the enhancer treated SC indicated that TTO,CO,RO and AVO increased the LP permeation by extraction of SC lipids.The results of thermodynamic studies and histopathological examination of AVO treated SC suggested additional mechanisms for AVO facilitated permeation i.***.transient reduction in barrier resistance of SC and intracellular transport by dekeratinization of corneocytes which may be attributed to the presence of triglycerides as constituents of AVO.It is feasible to deliver therapeutically effective dose of LP via transdermal route using AVO as penetration enhancer.

COMPARATIVE EFFECTS OF losartan AND CAPTOPRIL ON VENTRICULAR REMODELING AND FUNCTION AFTER MYOCARDIAL INFARCTION IN THE RAT

Objective. To determine the effects of losartan and captopril treatment on ventricular remodeling and function after myocardial infarction in rats. Methods. Thirty-two rats with MI induced by coronary ligation after seven days were divided into four groups randomly and treated with captopril(2 g. liter-1, group A), losartan(10 mg. kg-1. d-1, group B), losartan(30 mg. kg-1. d-1,group C) and placebo (no drug, group D) for six weeks, respectively. Shamoperated rats(group E)served as controls. Echocardiography was performed at 1 and 7 weeks after MI, re- spectively. Results. Compared with the results before treatment,both LV end-diastolic internal diameter and volume decreased significantly and the thickened Posterior wall was reversed in group A, B and C; the peak early filling velocity decreased whereas the peak velocity was increased in these three groups. There are no significant difference among the three treated groups. However,LV end-diastolic internal diameter and the E/A were still increased,whereas the thickness of anterior wall and the peak velocity of LV outflow were decreased in group A,B,and C after treatment comparing with group E. Conclusion. Both angiotensin converting enzyme inhibitor and angiotensin II receptor antagonist can prevent the ventricular remodeling and improve the ventricular function.

losartan activates sirtuin 1 in rat reduced-size orthotopic liver transplantation

AIM: To investigate a possible association between losartan and sirtuin 1(SIRT1) in reduced-size orthotopic liver transplantation(ROLT) in rats.METHODS: Livers of male Sprague-Dawley rats(200-250 g) were preserved in University of Wisconsin preservation solution for 1 h at 4 ℃ prior to ROLT.In an additional group,an antagonist of angiotensin Ⅱ type 1 receptor(AT1R),losartan,was orally administered(5 mg/kg) 24 h and 1 h before the surgical procedure to both the donors and the recipients.Transaminase(as an indicator of liver injury),SIRT1 activity,and nicotinamide adenine dinucleotide(NAD+,a co-factor necessary for SIRT1 activity) levels were determined by biochemical methods.Protein expression of SIRT1,acetylated Fox O1(ac-Fox O1),NAMPT(the precursor of NAD+),heat shock proteins(HSP70,HO-1) expression,endoplasmic reticulum stress(GRP78,IRE1 a,p-e IF2) and apoptosis(caspase 12 and caspase 3) parameters were determined by Western blot.Possible alterations in protein expression of mitogen activated protein kinases(MAPK),such as p-p38 and p-ERK,were also evaluated.Furthermore,the SIRT3 protein expression and m RNA levels were examined.RESULTS: The present study demonstrated that losartan administration led to diminished liver injury when compared to ROLT group,as evidenced by the significant decreases in alanine aminotransferase(358.3 ± 133.44 vs 206 ± 33.61,P losartan was associated with enhanced SIRT1 protein expression and activity(5.27 ± 0.32 vs 6.08 ± 0.30,P losartan treatment also provoked significant attenuation of endoplasmic reticulum stress parameters(GRP78,IRE1 a,p-e IF2) which was consistent with reduced levels of both caspase 12 and caspase 3.Furthermore,losartan administration stimulated H

Non-invasive assessment for intratumoural distribution of interstitial fluid flow

Interstitial fluid plays a vital role in drug delivery and tumour treatment.However,few non-invasive measurement methods are available for measuring low-velocity biological fluid flow.Therefore,this study aimed to develop a novel technology called interstitial flow velocity-MRI.The interstitial flow velocity-MRI sequence consists of a dual inversion recovery preparation and an improved stimulated echo sequence(ISTE)combined with phase-contrast MRI.A homemade flow phantom was used to assess the feasibility and sensitivity of interstitial flow velocity-MRI.In addition,xenografts of female BALB/c mouse models of 4T1 breast cancer administered losartan(40 mg/kg)or saline(n?6)were subjected to imaging on a 7.0 T scanner to assess the in vivo interstitial fluid flow velocity.The results showed a significant correlation(Plosartan group had a lower mean interstitial fluid velocity than the control group(85±16 vs 113±24 mm/s).In addition,compared to the saline treatment,losartan treatment reduced the proportion of collagen fibres by 10%and 12%in the Masson and Sirius red staining groups,respectively.Interstitial flow velocity-MRI has the potential to determine interstitial fluid flow velocity non-invasively and exhibits an intuitive velocity map.
