Schisandra chinensis Turcz.(Baill.) is a plant species with fruits that have been well known in Far Eastern medicine for a long time. It has traditionally been used as a stimulating and fortifying agent in cases of physical exhaustion and to inhibit *** major bioactive compounds found in S. chinensis are lignans with a dibenzocyclooctadiene skeleton, but little is known about their biosynthesis in plants. S. chinensis is the ideal medicinal plant for studying the biosynthesis of lignans, especially the dibenzocyclooctadiene skeleton. Genomic information for this important herbal plant is unavailable. To better understand the lignan biosynthesis pathway, we generated transcriptome sequences from the fruit during ripening and performed de novo sequence assembly, yielding136 843 unique transcripts with N50 of 1778 bp. Putative functions could be assigned to 41 824 transcripts(51.57%) based on BLAST searches against annotation databases including GO(Gene ontology) and KEGG(Kyoto Encyclopedia of Genes and Genomes). Furthermore, 22 candidate cytochrome P450 genes and 15 candidate dirigent proteins genes that were most likely involved in the lignan biosynthesis pathway were discovered based on transcriptome sequencing of S. chinensis. The genomic data obtained from S. chinensis, especially the identification of putative genes involved in the lignan biosynthesis pathway, will facilitate our understanding of lignan biosynthesis at the molecular level. The lignan metabolite profiles were analyzed by metabolomes, the accumulation patterns of 30 metabolites involved in the lignan pathway were studied. Co-expression network of lignan contents and transcriptional changes showed355 strong correlations(correlation coefficient, R^2 > 0.9) between 21 compounds and 153 transcripts. Phytochemical investigation on Forsythia suspensa(Thunb.) Vahl afforded ten compounds, including five lignan glycosides and five phenylethanoid glycosides. The compounds were isolated by using HP-20 macroporous resin, silica gel, octadecyl silica gel(ODS), size exclusion chromatography resin HW-40 chromatography, and preparative HPLC. The structures were established through application of extensive spectroscopic methods, including ESI-MS, 1D- and 2D-NMR spectroscopy. They were identified as forsythialanside E(1), 8′-hydroxypinoresinol-4′-O-β-D-glucoside(2), 8′-hydroxypinoresinol(3), lariciresinol-4′-O-β- D-glucoside(4), lariciresinol-4-O-β-D-glucoside(5), forsythoside H(6), forsythoside I(7), forsythoside F(8), plantainoside B(9), and plantainoside A(10). Compound 1 was a new lignan glycoside.

From the branches of Tabebuia chrysotricha (Bignoniaceae), two lignan glycosides (1, 2) and two phenolic compound glycosides (3, 4) were isolated, along with 15 known compounds (5 - 19). The structures of the new compounds were elucidated on the basis of spectroscopic evidence and those of the known compounds were identified by comparison of spectroscopic data with those reported in the literature. The DPPH radical-scavenging activity was assayed for the compounds isolated, the aryltetralin-type lignans showing moderate activity.

Objective:Gut-derived serotonin strongly inhibits bone formation by inhibiting osteoblast *** previous study demonstrated that the lignan-rich fraction prepared from Sambucus willimasii Hance,a folk herbal medicine used to treat bone fractures and joint diseases in China,exerted bone-protective effects,and its actions were modulated by suppressing the synthesis of gut-derived serotonin via the inhibition of intestinal tryptophan hydroxylase 1(TPH-1).However,there is no direct evidence for the action of lignans on *** study aimed to verify the direct action of lignans on the TPH-1 and its influence on serotonin synthesis and bone ***:Molecular docking and surface plasmon resonance were performed to determine the affinities of lignans to *** cell viability and the protein activity and expression of TPH-1 were measured in RBL2H3 *** serum serotonin level and bone mineral density upon lignan treatment in ovariectomized mice were ***:The lignans showed high binding scores and binding affinities to TPH-1,inhibited the activity and protein expression of TPH-1,suppressed the serum serotonin levels in ovariectomized mice as well as promoted bone mineral ***:This is the first study to report that lignans are novel TPH-1 inhibitors and that these lignans could be potential agents for the management of serotonin-related diseases,including osteoporosis.
