jasminum sambac(jasmine flower),a world-renowned plant appreciated for its exceptional flower fragrance,is of cultural and economic ***,the genetic basis of its fragrance is largely ***,we present the first de novo genome assembly of *** with 550.12 Mb(scaffold N50=40.10 Mb)assembled into 13 *** synthase(TPS)genes associated with flower fragrance are considerably amplified in the form of gene clusters through tandem duplications in the *** clusters within the salicylic acid/benzoic acid/theobromine(SABATH)and benzylalcohol O-acetyltransferase/anthocyanin O-hydroxycinnamoyltransferases/anthranilate N-hydroxycinnamoyl/benzoyltransferase/deacetylvindoline 4-O-acetyltransferase(BAHD)superfamilies were identified to be related to the biosynthesis of phenylpropanoid/benzenoid *** key genes involved in jasmonate biosynthesis were duplicated,causing an increase in copy *** addition,multi-omics analyses identified various aromatic compounds and many genes involved in fragrance biosynthesis ***,the roles of JsTPS3 in b-ocimene biosynthesis,as well as JsAOC1 and JsAOS in jasmonic acid biosynthesis,were functionally *** genome assembled in this study for *** offers a basic genetic resource for studying floral scent and jasmonate biosynthesis,and provides a foundation for functional genomic research and variety improvements in jasminum.

Being a valued aromatic and ornamental plant, jasminum sambac is widely cultivated in the Southeast Asia for utilization of its flowers. Floral aging in J. sambac starts about 24 hours post anthesis（HPA） and ends up in about 36 HPA, with a visual sign of petal-color-turning from white to violet. UPLC-TOF-MS analysis indicated the accumulation of cyanidin 3-O-rutinoside associated with the aging progression. Floral ethylene and salicylic acid release peaked at c.a. 12 HPA correlating with senescence development, 12 hours ahead of color change. To gain more inside into the gene regulation network in this particular flower aging process, we conduct a transcriptome analysis using total RNA samples from petals at 0, 12, 24 and 36 HPA. We identified several transcription factors which were co-expressed well with anthocyanin biosynthesis and ethylene and salicylic acid biosynthesis/responsive genes. These included one MYC gene, two bHLH genes and three WRKY genes among others. We isolated the full-length cDNA sequences of these genes and further characterized their functions in transgenic J. sambac, petunia and Arabidopsis that together provided primary evidences of a master effect on floral aging by the WRKY transcription factors. Interactions of the three WRKY proteins indicated that they co-operated in the regulations of the senescence processes.
