Photodynamic therapy(PDT)has shown a promising capability for cancer treatment with minimal side *** green(ICG),the only clinically approved near-infrared(NIR)fluorophore,has been used as a photosensitizer for PDT in clinical ***,the main obstacle of directly utilizing ICG in the clinic lies in its low singlet oxygen(1O_(2))quantum yield(QY)and instability in aqueous *** improve the PDT efficacy of ICG,free ICG molecules were assembled with free oxygen nanobubbles(NBs-O_(2))to fabricate ICG-NBs-O_(2) by hydrophilic-hydrophobe interactions on the gas-liquid ***,1O2 QY of ICG-NBs-O_(2) solution was significantly increased to 1.6%,which was estimated to be 8 times as high as that of free ICG ***,ICG-NBs-O_(2) exhibited better aqueous solution stability compared with free ***,through establishing tumor models in nude mice,the therapeutic efficacy of ICG-NBs-O_(2) was also assessed in the PDT treatment of oral *** tumor volume in ICG-NBs-O_(2) treated group on day 14 decreased to 0.56 of the initial tumor size on day 1,while the tumor volume in free ICG treated group increased to 2.4 *** results demonstrated that ICG-NBs-O_(2) showed excellent tumor ablation in ***,this facile method provided an effective strategy for enhanced PDT treatment of ICG and showed great potential in clinical application.

indocyanine green-Conjugated Magnetic Prussian Blue Nanoparticles for Synchronous Photothermal/Photodynamic Tumor Therapy

indocyanine green(ICG) is capable of inducing a photothermal effect and the production of cytotoxic reactive oxygen species for cancer therapy. However, the major challenge in applying ICG molecules for antitumor therapy is associated with their instability in aqueous conditions and rapid clearance from blood circulation,which causes insufficient bioavailability at the tumor ***, we conjugated ICG molecules with Prussian blue nanoparticles enclosing a Fe_3O_4 nanocore, which was facilitated by cationic polyethyleneimine via electrostatic adsorption. The nanocarrier-loaded ICG formed stable aggregates that enhanced cellular uptake and prevented fluorescence quenching. Moreover, the strong superparamagnetism of the Fe_3O_4 core in the obtained nanocomposites further improved cellular internalization of the drugs guided by a localized magnetic field. The therapeutic efficacy of this nanoplatform was evaluated using tumor models established in nude mice, which demonstrated remarkable tumor ablation in vivo due to strong photothermal/photodynamic effects. This study provides promising evidence that this multifunctional nanoagent might function as an efficient mediator for combining photothermal and photodynamic cancer therapy.

Colorectal cancer(CRC)is a lethal malignancy with a high mortality rate due to its low immunogenicity,the strong immunosuppressive milieu and poor drug *** overcome these obstacles,a cascade synergistic nanosystem(denoted as R837/ICG@Lip)was developed via self-assembly of heater indocyanine green(ICG)and toll-like receptor-7 agonist imiquimod(R837)into thermosensitive liposome for simultaneous induction of immunogenic cell death(ICD)and reversing of suppressive tumor *** obtained nanoparticles exhibited NIR-triggered drug release,good photothermal conversion efficiency and phototoxicity towards CT26 colorectal cancer *** vivo results reveal that the R837/ICG@Lip could be effectively accumulated in CT26 subcutaneous tumors and the draining lymph *** importantly,R837/ICG@Lip-mediated low-temperature photothermal therapy triggers ICD,promotes the maturation of host dendritic cells(DCs),and subsequently amplifies adaptive antitumor T-cell responses,resulting in‘Cold to Hot'*** directly affecting immune cells,the secretion of some immune-related cytokines further indirectly boosted anti-cancer *** combining with the indoleamine 2,3-dioxygenase(IDO)inhibitor,the systemic antitumor immune response was further augmented,achieving best tumor inhibition ***,low-temperature mediated photoimmunotherapy targeting multiple antitumor immune pathways boost synergistic antitumor immunity of tolerance tumors.

indocyanine green clearance test and model for end-stage liver disease score of patients with liver cirrhosis

BACKGROUND: The indocyanine green (ICG) clearance test (clearance rate (K) and retention rate at 15 minutes (R(15))) is a sensitive indicator to evaluate liver function. The model for end-stage liver disease (MELD) score has emerged as a useful tool for estimating the mortality of patients awaiting liver transplantation and has recently been validated on patients with liver diseases of various etiologies and severity. In this study, we investigated the correlation between the ICG clearance test and MELD score of patients with liver cirrhosis. METHODS: From June 2007 to March 2008, 52 patients with liver cirrhosis admitted to our center were classified into Child-Pugh class A (8 patients), B (14) and C (30). The ICG clearance test (K value and R(15)) was performed by ICG pulse spectrophotometry (DDG-3300K), and the MELD scores of patients were calculated. RESULTS: As the Child-Pugh classification of liver function gradually deteriorated, the K value decreased, while R(15) and MELD score increased. There were significant statistical differences in K value, R(15) and MELD score in patients with different Child-Pugh classifications. Significant correlations were found between the parameters of the ICG clearance test (K value and R(15)) and MELD score. A negative correlation was observed between K value and MELD score (r=-0.892, P < 0.05), while a positive correlation was observed between R(15) and MELD score (r=0.804, P < 0.05). CONCLUSIONS: The ICG clearance test and MELD score are good parameters for evaluating liver function. Moreover, K value and R(15) have significant correlations with MELD score, especially the K value, which may be a convenient and appropriate indicator to evaluate liver function of patients with liver cirrhosis.

indocyanine green kinetics to assess liver function: Ready for a clinical dynamic assessment in major liver surgery?

indocyanine green(ICG) kinetics(PDR/R15) used to quantitatively assess hepatic function in the perioperative period of major resective surgery and liver transplantation have been the object of an extensive, updated and critical review. New, non invasive bedside monitors(pulse dye densitometry technology) make this opportunity widely available in clinical practice. After having reviewed basic concepts of hepatic clearance, we analysed the most common indications ICG kinetic parameters have nowadays in clinical practice, focusing in particular on the diagnostic and prognostic role of PDR and R15 in the perioperative period of major liver surgery and liver transplantation. As recently pointed out, even if of extreme interest, ICG clearance parameters have still some limitations, to be considered when using these tests.
