Wolbachia symbiosis and insect immune response

Bacterial intraceUular symbiosis is very common in insects, having significant consequences in promoting the evolution of life and biodiversity. The bacterial group that has recently attracted particular attention is Wolbachia pipientis which probably represents the most ubiquitous endosymbiont on the planet. W. pipientis is a Gram-negative obligatory intracellular and maternally transmitted a-proteobacterium, that is able to establish symbiotic associations with arthropods and nematodes. In arthropods, Wolbachia pipientis infections have been described in Arachnida, in Isopoda and mainly in Insecta. They have been reported in almost all major insect orders including Diptera, Coleoptera, Hemiptera, Hymenoptera, Orthoptera and Lepidoptera. To enhance its transmission, W. pipientis can manipulate host reproduction by inducing parthenogenesis, feminization, male killing and cytoplasmic incompatibility. Several polymerase chain reaction surveys have indicated that up to 70% of all insect species may be infected with W. pipientis. How does W. pipientis manage to get established in diverse insect host species？ How is this intracellular bacterial symbiont species so successful in escaping the host immune response？ The present review presents recent advances and ongoing scientific efforts in the field. The current body of knowledge in the field is summarized, revelations from the available genomic information are presented and as yet unanswered questions are discussed in an attempt to present a comprehensive picture of the unique ability of W. pipientis to establish symbiosis and to manipulate reproduction while evading the host＇s immune system.

A pattern recognition receptor C-type lectin TcCTL14 contributes to immune response and development in the red flour beetle, Tribolium castaneum

Evidence is accumulating that pattern recognition receptor(PRR)C-type lectins(CTL)play essential roles in recognition of ***14(accession ***00871)contains the most domains among all CTL of Tribolium *** the biological function of TcCTL14 remains *** this study,TcCTL14 exhibiting typical motif and domain of CTL was cloned from *** expression pattern analysis showed that TcCTL14 was highly expressed in late pupae and central nervous system,and was upregulated after treatment with Escherichia coli and Staphylococcus aureus,*** of binding affinity revealed that recombinant TcCTL14 not only could bind to lipopolysaccharide and peptidoglycan in a dose-dependent fashion,but possibly could bind to and agglutinate different bacteria in a Ca^(2+)-dependent *** of TcCTL14 before injection with bacteria led to the downregulation of nuclear factor-κB transcription factors of Toll/IMD and 4 antimicrobial *** of TcCTL14 also caused suppressed metamorphosis,reduced fecundity,and delayed embryogenesis of *** observation discovered that knockdown of TcCTL14 inhibited the development of ovaries and *** detection of signaling pathways revealed that TcCTL14 may be involved in metamorphosis and fecundity by impacting 20-hydroxyecdysone and vitellogenin,***,these results indicate that TcCTL14 may contribute to immune response by agglutination or regulating the expression of antimicrobial peptides by the Toll/IMD pathway,and is required for *** development including metamorphosis,fecundity,and *** findings will improve the functional cognition of PRR CTL in insects and provide the new strategy for pest control.

Tomato SlPti5 plays a regulative role in the plant immune response against Botrytis cinerea through modulation of ROS system and hormone pathways

While SlPti5 has been shown to play a crucial role in the regulation of antagonistic genes in Solanum lycopersicum and Arabidopsis against pathogen infection,there have been no comprehensive studies on the effects of SlPti5 on the regulatory response mechanism of reactive oxygen species(ROS) system and hormone pathways during growth and disease resistance of tomato ***,we investigated the function of SlPti5 in the defense response of tomato against Botrytis cinerea utilizing a virus-induced gene silencing(VIGS)-based *** profile analysis showed that SlPti5 was significantly induced upon *** infection,with high expression levels in the leaves and fruit of ***-based silencing of SlPti5 inhibited early vegetative growth,increased the plant’s susceptibility to infection,promoted the development of ROS,affected the expression of genes involved in the ROS scavenging system,and attenuated the expression of genes associated with pathogenesis and the ethylene/jasmonic acid signaling *** sum,our data demonstrated that SlPti5 stimulates the immune response of tomato plant to Botrytis cinerea infection by involving the ethylene(ET)-and jasmonic acid(JA)-mediated pathways and modulating the expression of some key pathogenesis-related(PR) genes.

Double sex and mab-3-related transcription factor 1(Dmrt1),which is expressed in goat male germline stem cells(mGSCs)and Sertoli cells,is one of the most conserved transcription factors involved in sex *** this study,we highlighted the role of Dmrt1 in balancing the innate immune response in goat ***1 recruited promyelocytic leukemia zinc finger(Plzf),also known as zinc finger and BTB domain-containing protein 16(Zbtb16),to repress the Toll-like receptor 4(TLR4)-dependent inflammatory signaling pathway and nuclear factor(NF)-κ*** of Dmrt1 in seminiferous tubules resulted in widespread degeneration of germ and somatic cells,while the expression of proinflammatory factors were significantly *** also demonstrated that Dmrt1 stimulated proliferation of mGSCs,but repressed apoptosis caused by the immune ***,Dmrt1 is sufficient to reduce inflammation in the testes,thereby establishing the stability of spermatogenesis and the testicular microenvironment.

The immune-inflammatory response is associated with increased nitro-oxidative *** aim of this mechanistic review is to examine:(a)the role of redox-sensitive transcription factors and enzymes,ROS/RNS production,and the activity of cellular antioxidants in the activation and performance of macrophages,dendritic cells,neutrophils,T-cells,B-cells,and natural killer cells;(b)the involvement of high-density lipoprotein(HDL),apolipoprotein A1(ApoA1),paraoxonase-1(PON1),and oxidized phospholipids in regulating the immune response;and(c)the detrimental effects of hypernitrosylation and chronic nitro-oxidative stress on the immune *** redox changes during immune-inflammatory responses are orchestrated by the actions of nuclear factor-κB,HIF1α,the mechanistic target of rapamycin,the phosphatidylinositol 3-kinase/protein kinase B signaling pathway,mitogen-activated protein kinases,5'AMP-activated protein kinase,and peroxisome proliferator-activated *** performance and survival of individual immune cells is under redox control and depends on intracellular and extracellular levels of ROS/*** are heavily influenced by cellular antioxidants including the glutathione and thioredoxin systems,nuclear factor erythroid 2-related factor 2,and the HDL/ApoA1/PON1 *** nitro-oxidative stress and hypernitrosylation inhibit the activity of those antioxidant systems,the tricarboxylic acid cycle,mitochondrial functions,and the metabolism of immune *** conclusion,redox-associated mechanisms modulate metabolic reprogramming of immune cells,macrophage and T helper cell polarization,phagocytosis,production of pro-versus anti-inflammatory cytokines,immune training and tolerance,chemotaxis,pathogen sensing,antiviral and antibacterial effects,Toll-like receptor activity,and endotoxin tolerance.

Active dry yeast supplementation improves the growth performance,rumen fermentation, and immune response of weaned beef calves

The objective of this experiment was to investigate the potential benefits of active dry yeast(ADY)on the growth performance,rumen fermentation,nutrient digestibility,and serum parameters of weaned beef *** Simmental crossbred male calves(body weight=86.47±4.41 kg and 70±4 d of age)were randomly divided into 2 groups:control(CON)(fed basal ration)and ADY(fed basal ration and 5 g/d ADY per calf).The dietary concentrate-to-roughage ratio was 35:*** the calves were regularly provided rations 3 times a day at 07:00,13:00,and 19:00 and had free access to wate *** experiment lasted for 60 *** average daily gain of ADY group was higher(P=0.007)than that of the CON group,and the ratio of feed intake to average daily gain in the ADY group was reduced(P=0.022)as compared to the CON *** concentration of ruminal ammonia-N was higher(P=0.023)in the CON group than that in the ADY group,but an opposite trend of microbial protein was found between the 2 ***,the ruminal concentrations of propionate and butyrate were higher(Pimmune response of weaned beef calves.

Effects of N-Linked Glycan on Lassa Virus Envelope Glycoprotein Cleavage,Infectivity,and immune response

Lassa virus(LASV)belongs to the Mammarenavirus genus(family Arenaviridae)and causes severe hemorrhagic fever in *** glycoprotein complex(GPC)contains eleven N-linked glycans that play essential roles in GPC functionalities such as cleavage,transport,receptor recognition,epitope shielding,and immune *** used three mutagenesis strategies(asparagine to glutamine,asparagine to alanine,and serine/tyrosine to alanine mutants)to abolish individual glycan chain on GPC and found that all the three strategies led to cleavage inefficiency on the 2nd(N89),5th(N119),or 8th(N365)glycosylation *** evaluate N to Q mutagenesis for further research,it was found that deletion of the 2nd(N89Q)or 8th(N365Q)glycan completely inhibited the transduction efficiency of pseudotyped *** further investigated the role of individual glycan on GPC-mediated immune response by DNA immunization of *** of the individual 1st(N79Q),3rd(N99Q),5th(N119Q),or 6th(N167Q)glycan significantly enhanced the proportion of effector CD4+cells,whereas deletion of the 1st(N79Q),2nd(N89Q),3rd(N99Q),4th(N109Q),5th(N119Q),6th(N167Q),or 9th(N373Q)glycan enhanced the proportion of CD8+effector T *** of specific glycan improves the Th1-type immune response,and abolishment of glycan on GPC generally increases the antibody titer to the glycan-deficient ***,the antibodies from either the mutant or WT GPC-immunized mice show little neutralization effect on wild-type *** glycan residues on GPC provide an immune shield for the virus,and thus represent a target for the design and development of a vaccine.

Schaftoside inhibits 3CL^(pro)and PL^(pro)of SARS-CoV-2 virus and regulates immune response and inflammation of host cells for the treatment of COVID-19

It is an urgent demand worldwide to control the coronavirus disease 2019(COVID-19)pandemic caused by the severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)*** 3-chymotrypsin-like protease(3CL^(pro))and papain-like protease(PL^(pro))are key targets to discover SARS-CoV-2 *** screening 12 Chinese herbal medicines and 125 compounds from licorice,we found that a popular natural product schaftoside inhibited 3CL^(pro)and PL^(pro)with IC_(50)values of 1.73±0.22 and 3.91±0.19μmol/L,respectively,and inhibited SARS CoV-2 virus in Vero E6 cells with EC_(50)of 11.83±3. 23μmol/L. Hydrogen-deuterium exchange mass spectrometry analysis, quantum mechanics/molecular mechanics calculations, together with site-directed mutagenesis indicated the antiviral activities of schaftoside were related with non-covalent interactions with H41, G143 and R188 of3CLpro, and K157, E167 and A246 of PLpro. Moreover, proteomics analysis and cytokine assay revealed that schaftoside also regulated immune response and inflammation of the host cells. The antiinflammatory activities of schaftoside were confirmed on lipopolysaccharide-induced acute lung injury mice. Schaftoside showed good safety and pharmacokinetic property, and could be a promising drug candidate for the prevention and treatment of COVID-19.

Matrix metalloproteinase 2 contributes to adult eclosion and immune response in the small hive beetle,Aethina tumida

During the pupal-adult eclosion process of holometabolous insects,the old cuticle is shed and replaced by a completely different new cuticle that requires tanning and expansion,along with extensive extracellular matrix(ECM)*** vertebrates,matrix metalloproteinases(MMPs),a class of zinc-dependent endopeptidases,play key roles in regulating the ECM that surrounds ***,little is known about these extracellular proteinases available in *** small hive beetle(SHB),Aethina tumida,is a widespread invasive parasite of honey *** this study,6 MMP homologs were identified in the SHB *** interference experiments showed that all 6 AtMmps are not required for the larval-pupal transition,only AtMmp2 was essential for pupal-adult eclosion in *** of AtMmp2 resulted in eclosion defects and wing expansion failure,as well as mortality within 3 d of adult *** analysis revealed that knockdown of AtMmp2 significantly increased expression of the Toll and Imd pathways,chitin metabolism,and cross-linking(such as the pro-phenoloxidase activating cascade pathway and the tyrosine-mediated cuticle sclerotization and pigmentation pathway).These data revealed evolutionarily conserved functions of Mmp2 in controlling adult eclosion and wing expansion,also provided a preliminary exploration of the novel function of regulating Toll and Imd pathways,as well as new insights into how MMPs regulate insect development and defense barriers.
