Retinoic acid enhances lactoferrin-induced iga responses by increasing betaglycan expression

Lactoferrin （LF） and retinoic acid （RA） are enriched in colostrum, milk, and mucosal tissues. We recently showed that LF-induced iga class switching through binding to betaglycan （transforming growth factor-beta receptor III, TpRIII） and activation of canonical TGF-p signaling. We investigated the combined effect of LF and RA on the overall iga response. An increase in iga production by LF was further augmented by RA. This combination effect was also evident in Ig germ-line α （GLa） transcription and GLa promoter activity, indicating that LF in cooperation with RA increased iga isotype switching. We subsequently found that RA enhanced TβRIII expression and that this increase contributed to LF-stimulated iga production. In addition to the iga response, LF and RA in combination also enhanced the expression of the gut-homing molecules C-C chemokine receptor 9 （CCR9） and a4β7 on B cells. Finally, peroral administration of LF and RA enhanced the frequency of CCR9＋ iga＋ plasma cells in the lamina propria. Taken together, these results suggest that LF in cooperation with RA can contribute to the establishment of gut iga responses.

Correlation analysis of celiac sprue tissue transglutaminase and deamidated gliadin IgG/iga

AIM:To indirectly determine if tissue transglutaminase(tTG)-specific T cells play a crucial role in the propagation of celiac ***:Anti-deamidated gliadin peptide(DGP) and anti-tTG iga and IgG were measured in the sera of celiac patients(both untreated and treated).The correlations were determined by Spearman's rank correlation ***:In celiac patients,we found a very significant correlation between the production of DGP iga and IgG(r = 0.75),indicating a simultaneous and ongoing production of these two isotypes reminiscent of oral vaccination ***,there was far less association between the production of tTG iga and tTG IgG in celiac patients(r = 0.52).While tTG iga was significantly correlated with DGP iga(r = 0.80) and DGP IgG(r = 0.67),there was a weak correlation between production of anti-tTG IgG and the production of anti-DGP iga(r = 0.38) and anti-DGP IgG(r = 0.43).CONCLUSION:These data demonstrate that the production of anti-tTG iga is directly correlated to the production of anti-DGP IgG and iga,whereas anti-tTG IgG is only weakly *** result therefore supports the hapten-carrier theory that in well-established celiac patients anti-tTG iga is produced by a set of B cells that are reacting against the complex of tTG-DGP in the absence of a tTG-specific T cell.

GENE ENGINEERING EB VIRUS MEMBRANE ANTIGEN IN DETECTION OF MA-iga ANTIBODY(COMPARISON WITH VCA-iga AND EA-iga ANTIBODIES)

MA-IgA antibody was positive in 96.8% of the pretreatment NPC patients with a GMT of 1:36.3. MA-IgA detection by this method was more sensitive than EA-IgA detection by IE. In contrast patients with tumors other than NPC were negative for MA-IgA antibody. 9.1% of VCA-IgA positive persons were MA-IgA positive with a GMT of less than 1:5. No MA-IgA positive was found in VCA-IgA negatives. The results indicated that this method was relatively specific. In the treatment group the positive rate and GMT of MA-IgA antibody declined with increase in survival time and the decline was faster than VCA-IgA. When recurrence or distant metastasis developed similar to VCA-IgA and EA-IgA antibodies the positive rate and GMT of MA-IgA antibody increased to its pretreatment level. Therefore MA-IgA detection might be valuable in the early diagnosis and monitor of NPC.

With gene engineering EB virus membrane antigen as the diagnostic antigen, indirect immunofluo-rescence (IF) assay was used to detect iga antibody against EB virus membrane antigen (MA-iga) in sera from 202 nasopharyngeal carcinoma (NPC) patients and 315 controls (normal and patients with other tumors). MA-iga antibody was positive in 96.8% of the pretreatment NPC patients with a GMT of 1:36.3. MA-iga detection by this method was more sensitive than EA-iga detection by IE. In contrast, patients with tumors other than NPC were negative for MA-iga antibody. 9.1% of VCA-iga positive persons were MA-iga positive with a GMT of less than 1:5. No MA-iga positive was found in VCA-iga negatives. The results indicated that this method was relatively specific. In the treatment group, the positive rate and GMT of MA-iga antibody declined with increase in survival time and the decline was faster than VCA-iga. 通过 1 ,4-丁二醇 -二缩水甘油醚法活化琼脂糖载体 ,既起到了活化载体的作用 ,同时又由于该活化剂分子是长链结构而给载体提供了一定长度的手臂 ,有利于人免疫球蛋白 G( Ig G)大分子配基与载体的连接 ,用该方法制成的免疫吸附剂对高 Ig A肾病病人血清中 Ig A进行了吸附研究 .通过体外实验条件的优化发现 ,最高可吸附 60 %的 Ig A,同 CDI法比较 (吸附率可达 3 5 %左右 ) 。

为了探讨胃肠局部对ＨＰ的体液免疫反应，作者采用间接ＥＬＩＳＡ法检测了８５例患者胃液中抗ＨＰＩｇＡ。结果表明：①ＨＰ培养阳性者胃液中抗ＨＰＩｇＡ含量及阳性率均显著高于ＨＰ阴性者（Ｐ＜０．００１），与细菌学检测结果比较，其敏感性、特异性及诊断符合率分别为９６．４％、９２．９％和９５．２％；②不同胃部疾病之间胃液抗ＨＰＩｇＡ含量及阳性率有显著差异（Ｐ＜０．０５），慢性活动性胃炎（ＡＣＧ）组显著高于消化性溃疡（ＰＵ）组和慢性非活动性胃炎（ＮＡＣＧ）组（Ｐ＜０．０５），ＰＵ组和胃癌（ＧＣａ）组显著高于ＮＡＣＧ组（Ｐ＜０．０５），中重度胃炎显著高于轻度胃炎（Ｐ＜０．０５）；③胃液中抗ＨＰＩｇＡ与血清中抗ＨＰＩｇＡ、ＩｇＧ含量呈正相关（ｒ＝０．９２８，Ｐ＜０．０５；ｒ＝０．９４８，Ｐ＜０．０２）。提示：胃液中抗ＨＰＩｇＡ测定能较好地反映胃粘膜局部对ＨＰ的体液免疫反应，它不但可用于探索ＨＰ的致病作用，而且也可用于ＨＰ感染的诊断。
