Background:multiple myeloma(MM) is a malignancy of terminally differentiated plasma cells that are predominantly localized in bone *** still remains incurable in most of the *** hypothesis of tumor stem cells(TSCs) states that although chemotherapy kills the malignant plasma cells, it is believed to leave a reservoir of TSCs that continue *** to identify TSCs in MM cell lines, however, remains to further ***:The purpose of this study was to isolate and identify tumor stem-like cells from MM cell lines RPMI-8226 and NCI-H929 and provided an experiment data for targeting therapy of MM TSCs in ***:The mouse antihuman CD138 and CD34 antibodies coupled to magnetic microbeads were respectively used to isolate CD138-CD34-cells from the RPMI-8226 and NCI-H929 cell lines by magnetic associated cell *** characteristics of CD138- CD34- cells were analyzed by the colony formation assay in the soft agar media, proliferative assay, drug resistant assay, transwell assay and the tumorigenic assay in nonobese diabetic/severe combined immunodeficiency(NOD/SCID) mice, ***:The isolated CD138-CD34-cells were capable of forming colonies in the soft agar media, exhibited high proliferative and migratory potency, possesed a strong drug resistant to paclitaxel and had a powerful tumorigenicity in NOD/SCID mice compared with non-CD138-CD34-MM ***, CD138-CD34- cells were found to possess the characteristics of tumor stem-like cells, such as high proliferative potency, more self-renewal and stronger ***:The biological characteristics of CD138-CD34-cells suggested the cells might be tumor stemlike cells in MM cell lines RPMI-8226 and *** data contributes to the identification of molecular targets important for future MM TSC targeting therapy.

Triptolide-induced Apoptosis by Inactivating Nuclear Factor-kappa B Apoptotic Pathway in multiple myeloma in vitro

The effect of triptolide on proliferation and apoptosis of human multiple myeloma RPMI-8226 cells in vitro,as well as the roles of nuclear factor-kappa B（NF-κB） and IκBα was *** effect of tritptolide on the growth of RPMI-8226 cells was studied by MTT *** was detected by Hoechest 33258 staining and Annexin V/PI double staining *** expression of NF-κB and IκBα was observed by Western blot and confocal *** results showed that triptolide inactivated NF-κB apoptotic pathway in human multiple myeloma RPMI-8226 *** at nM range induced proliferation inhibition in a dose-and time-dependent manner and apoptosis in a dose-dependent fashion in RPMI-8226 ***,we observed the inhibition of NF-κB /p65 in the nuclear fraction was correlated with the increase in the protein expression of IκBα in the *** results suggested that triptolide might exhibit its strong anti-tumor effects via inactivation of NF-κB/p65 and IκBα.
