珠江口3种鲸豚的MHC-I基因多态性研究

旨在了解珠江口栖息的中华白海豚(Sousa chinensis)、宽脊江豚(Neophocaena phocaenoides)和点斑原海豚(Stenella attenuate)免疫相关基因MHC-I(Major histocompatibility Complex)的多态性及其表达情况,以期为这3种鲸豚的保育工作提供基础资料。通过克隆测序的方法,首次证实MHC-I基因在这3种鲸豚体内表达。选择压力分析,表明MHC-I基因在这3种鲸豚中均受到强烈的正选择作用,提示其具有重要的免疫功能。3种鲸豚的MHC-I基因在系统发育树上相互混杂在一起,表明MHC-I基因存在跨物种多态性。结合选择压力分析和跨物种多态性,发现MHC-I基因受到平衡选择作用。珠江口3种鲸豚MHC-I基因多态性可能由平衡选择维持。

目的探讨血清CA125、IL-10和外周血淋巴细胞主要组织相容性复合体(MHC)Ⅱ联合检测在非霍奇金淋巴瘤诊断中的价值。方法试验组92例非霍奇金淋巴瘤患者,对照组50名正常健康人。全自动电化学发光分析仪检测血清CA125,ELISA方法检测IL-10,流氏细胞仪检测淋巴细胞MHCⅡ分子荧光强度。结果非霍奇金淋巴瘤患者CA125为(82±35)U/ml,IL-10为(286±83)pg/ml,淋巴细胞MHCⅡ分子平均荧光强度为168±27,它们与对照组比较有显著性差异(P<0.01)。联合检测血清CA125、IL-10和外周血淋巴细胞MHCⅡ,非霍奇金淋巴瘤的敏感性提高为92%,阴性预测值均提高为97%。结论联合检测血清CA125、IL-10和外周血淋巴细胞MHCⅡ能更好地诊断非霍奇金淋巴瘤。

The common goal within the overwhelming interests in stem cell research is to safely translate the science to patients. Although there are various methods by which this goal can be reached, this editorial emphasizes the safety of mesenchymal stem cell(MSC) transplant and possible confounds by the growing information on cancer stem cells(CSCs). There are several ongoing clinical trials with MSCs and their interactions with CSCs need to be examined. The rapid knowledge on MSCs and CSCs has now collided with regards to the safe treatment of MSCs. The information discussed on MSCs can be extrapolated to other stem cells with similar phenotype and functions such as placenta stem cells. MSCs are attractive for cell therapy, mainly due to reduced ethical concerns, ease in expansion and reduced ability to be transformed. Also, MSCs can exert both immune suppressor and tissue regeneration simultaneously. It is expected that any clinical trial with MSCs will take precaution to ensure that the cells are not transformed. However, going forward, the different centers should be aware that MSCs might undergo oncogenic events, especially as undifferentiated cells or early differentiated cells. Another major concern for MSC therapy is their ability to promote tumor growth and perhaps, to protect CSCs by altered immune responses. These issues are discussed in light of a large number of undiagnosed cancers.

Our understanding of transplant immunology has advanced from gross allograft rejection to cellular response and to current molecular level. More sensitive assays have been developed to characterize patient sensitization and to detect pre-existing donor-specific antibodies(DSA) in pre-transplant crossmatch. After a transplant, pre-existing or de novo DSA are increasingly monitored to guide clinical management. Therefore, it is important for clinicians to understand the basic concepts and key components of transplant immunology as well as be familiarized with the modern immunological techniques used in kidney transplantation.

Porcine reproductive and respiratory syndrome(PRRS) is one of the most important diseases of swine industry. The causal agent, PRRS-virus(PRRSV), is able to evade the host immune response and survive in the organism causing transient infections. Despite all scientific efforts, there are still some gaps in the knowledge of the pathogenesis of this disease. Antigen presenting cells(APCs), as initiators of the immune response, are located in the first line of defense against microorganisms, and are responsible for antigen recognition, processing and presentation. Dendritic cells(DCs) are the main type of APC involved in antigen presentation and they are susceptible to PRRSV infection. Thus, PRRSV replication in DCs may trigger off different mechanisms to impair the onset of a host effective immune response against the virus. On the one side, PRRSV may impair the basic functions of DCs by regulating the expression of major histocompatibility complex class Ⅱ and CD80/86. Other strategy followed by the virus is the induction of cell death of APCs by apoptosis, necrosis or both of them. The impairment and/or cell death ofAPCs could lead to a failure in the onset of an efficient immune response, as long as cells could not properly activate T cells. Future aspects to take into account are also discussed in this review.
