Effects of herba epimedii and Fructus Ligustri lucidi on the Transcription Factors in Hypothalamus of Aged Rats

Objective： To comparatively analyze the difference in the expression of 54 transcription factors in the hypothalamus using protein chips following the medication of Chinese drugs for Shen （肾）-tonification, herba epimedii, and Fructus Ligustri lucidi （FL） to aged rats. Methods： Wistar rats, aged 15 months of SPF grade, were randomized into three groups, three males and three females in each group. They were medicated with herba epimedii decoction （HED, 0.14 g/kg）, Fructus Ligustri lucidi decoction （FLD, 0.12 g/kg）, and distilled water, respectively, twice a day for 15 days. The rats were sacrificed at the morning of the 16th day 1 h after medication, and their hypothalamus was taken and made into homogenate under an ice-bath for detecting the expression of transcription factors with chip technique. Results： The expressions of signal transduction and transcription activation factor-6 （Stat-6） and androgen receptor （AR） were up-regulated, and those of pre-B transcription factor1 （Pbx-1）, stat-1 and AP-2 were down-regulated in both HED and FLD treated groups, but these changes occurred mainly in female rats in the former while mainly in males in the latter. Conclusions： Chinese drugs for Shen-tonification could impact the expression of transcription factors in the hypothalamus of aged rats, dominantly on the neuro-endocrine factors responsible for the growth and development. The effects of drugs for tonifying Shen-yang and for tonifying Shen-yin are different, which is probably one of the pharmacological mechanisms of the Shen-tonifying drugs.

The Neuropharmacological Effects of herba epimedii and its Main Bioactive Components:Icariin,Icariside Ⅱ and Icartin

herba epimedii,a traditional & Chinese medicine(T&CM),is used as a tonic,remedy for calming the nerves and anti-rheumatic agent in some Chinese proprietary medicine. Icariin,icarisideⅡ and icartin,are main bioactive products present in this herb. Previous studies have demonstrated that these compounds possess broad therapeutic capabilities,especially anti-osteoporosis,anti-oxidative stress and anti-cancer effects. In recent years,growing numbers of researches have focused on pharmacological actions of herba epimedii in central nervous system. Numerous preclinical studies have demonstrated that herba epimedii as agent for neurological disease,and our reports showed powerful neruoprotective effects of icariin and icarisideⅡ in Alzheimer’s disease(AD) and ischemic brain injury. Mechanistically,icariin increased cerebral blood flow and amyloid β-protein(Aβ) clearance,enhanced neurogenesis and memory,promoted synaptic plasticity may through the PDE5/NO/cGMP signaling pathway. Moreover,icariin is also effi cient at anti-depression,and possible molecular mechanisms involve in decrease of the hyper-responsiveness of corticotropin-releasing factor(CRF),regulation of 5-hydroxytryptamine system and anti-infl ammation effect. Although some Chinese proprietary medicine which are contain icariin have been used for osteoporosis in clinic,but no agents for neurological diseases. To further promote the researches on prevention and treatment of herba epimedii on central nervous system diseases,this review aim to summarize the anti-AD,anti-aging,and anti-ischemic brain injury effects of compounds of herba epimedii with reference to published data. Base on the literature compiled,the compounds of herba epimedii have tremendous potential to be developed therapeutic drugs for AD,ischemic stroke and depression. It is our hope that traditional Chinese medicine health industry could promote the development of health and contribute to the health welfare for all.

Obesity is associated with a number of metabolic abnormalities such as type 2 diabetes and has become a major health problem worldwide. In the present study, we investigated the effects of Epimedium koreanum Nakai(herba epimedii, HE) and its main constituent icariin on the adipocyte differentiation in 3T3-L1 preadipocytes. HE extract and icariin significantly reduced lipid accumulation and suppressed the expressions of PPARγ, C/EBPα, and SREBP-1c in 3T3-L1 adipocytes. They also inhibited fatty acid synthase(FAS), acyl-Co A synthase(ACS1), and perilipin. Moreover, HE extract and icariin markedly increased the phosphorylation of AMPK. These results indicated that HE extract and icariin can inhibit the adipocyte differentiation through downregulation of the adipogenic transcription factors, suggesting that HE containing icariin may be used as a potential therapeutic agent in the treatment and prevention of obesity.

Protective effects of kidney-tonifying Chinese herbal preparation on substantia nigra neurons in a mouse model of Parkinson's disease

The Chinese herbs herba epimedii, Fructus Ligustri Lucidi and Rhizoma Polygonati were injected into Parkinson＇s disease mice established via intraperitoneal injection of 1-methyl-4-phenyl-1, 2, 3, 6-tetrahydropyridine hydrochloride. The selective monoamine oxidase B inhibitor selegiline was used as a positive control drug. After successive administration for 4 weeks, herba epimedii could downregulate the expression of caspase-3 and increase the brain-derived neurotrophic factor level, as well as increase tyrosine hydroxylase activity in the substantia nigra of Parkinson＇s disease mouse models. Rhizoma Polygonaticould downregulate the expression of caspase-3 and FasL, and increase neural growth factor and brain-derived neurotrophic factor levels. Fructus Ligustn Lucidi could downregulate caspase-3 expression. Rhizoma Polygonati and Fructus Ligustn Lucidi did not produce obvious effects on tyrosine hydroxylase activity. herba epimedii and Fructus Ligustri Lucidi yielded similar effects on apoptosis-promoting factors to those elicited by selegiline. herba epimedii and Rhizoma Polygonati significantly increased the levels of neurotrophic factors compared with selegiline. herba epimedii significantly increased tyrosine hydroxylase activity compared with selegiline. It is indicated that the kidney-tonifying Chinese herbal preparation can downregulate the expression of apoptosis-promoting factors, increase neurotrophic factors levels in the substantia nigra and striatum, as well as increase tyrosine hydroxylase activity in the substantia nigra of Parkinson＇s disease mouse models, thereby exerting a stronger or similar neuroprotective effects compared with selegiline.

Optimization for supercritical CO2 extraction of icariin by response surface method

The aim of the present study was to optimize the supercritical CO 2 extraction conditions of icariin from herba epimedii by response surface method (RSM) and central composite design (CCD). A 3-factor, 5-level CCD was used for the optimization. Independent variables were extraction temperature, extraction pressure and entrainer flow rate. Dependent variable was yield ratio of icariin from herba epimedii. A two-order polynomial equation was fitted to the data. The results showed that the optimum extraction conditions were as follows: extraction temperature 46.5 °C, extraction pressure 30.6 MPa, entrainer flow rate 3.3 mL/min. CCD/ RSM is convenient and highly predictive for optimizing the extraction process of icariin from herba epimedii. © 2011 Journal of Chinese Pharmaceutical Sciences, School of Pharmaceutical Sciences, Peking University.
