Establishment of chronic hepatitis c virus infection:Translational evasion of oxidative defence

hepatitis c virus(HcV)causes a clinically important disease affecting 3%of the world *** is a single-stranded,positive-sense RNA virus belonging to the genus Hepacivirus within the Flaviviridae *** virus establishes a chronic infection in the face of an active host oxidative defence,thus adaptation to oxidative stress is key to virus *** a small RNA virus with a limited genomic capacity,we speculate that HcV deploys a different strategy to evade host oxidative *** of counteracting oxidative stress,it utilizes oxidative stress to facilitate its own *** is the first step in the replication of a plus strand RNA virus so it would make sense if the virus can exploit the host oxidative defence in facilitating this very first *** is particularly true when HcV utilizes an internal ribosome entry site element in translation,which is distinctive from that of cap-dependent translation of the vast majority of cellular genes,thus allowing selective translation of genes under conditions when global protein synthesis is ***,we were the first to show that HcV translation was stimulated by an important prooxidant-hydrogen peroxide in hepatocytes,suggesting that HcV is able to adapt to and utilize the host antiviral response to facilitate its own translation thus allowing the virus to thrive under oxidative stress condition to establish *** how HcV translation is regulated under oxidative stress condition will advance our knowledge on how HcV establishes *** chronicity is the initiator step in disease progression this will eventually lead to a better understanding of pathogenicity,which is particularly relevant to the development of anti-virals and improved treatments of HcV patients using anti-oxidants.

Autophagy is a lysosome-associated,degradative process that catabolizes cytosolic components to recycle nutrients for further use and maintain cell *** c virus(HcV)is a major cause of chronic hepatitis,which often leads to end-stage liverassociated diseases and is a significant burden on worldwide public *** lines of evidence indicate that autophagy plays an important role in promoting the HcV life cycle in host ***,the diverse impacts of autophagy on a variety of signaling pathways in HcV-infected cells suggest that the autophagic process is required for balancing HcVhost cell interactions and involved in the pathogenesis of HcV-related liver ***,the detailed molecular mechanism underlying how HcV activates autophagy to benefit viral growth is still ***,how the autophagic response contributes to disease progression in HcV-infected cells remains largely ***,in this review,we overview the interplay between autophagy and the HcV life cycle and propose possible mechanisms by which autophagy may promote the pathogenesis of HcVassociated chronic liver ***,we outline the related studies on how autophagy interplays with HcV replication and discuss the possible implications of autophagy and viral replication in the progression of HcV-induced liver diseases,e.g.,steatosis and hepatocellular ***,we explore the potential therapeutics that target autophagy to cure HcV infection and its related liver diseases.

Objective: To explore inhibitory effects of genome-specific, chemically synthesized siRNAs(small interference RNA) against NS3 gene of hepatitis c virus(HcV) 1a genotype in stable Huh-7(human hepatoma) cells as well as against viral replication in serum-inoculated Huh-7 cells. Methods: Stable Huh-7 cells persistently expressing NS3 gene were produced under antibiotic gentamycin(G418) selection. The cell clones resistant to 1 000 μg antibiotic concentration(G418) were picked as stable cell clones. The NS3 gene expression in stable cell clone was confirmed by RT-PcR and Western blotting. siRNA cell cytotoxicity was determined by MTT cell proliferation assay. Stable cell lines were transfected with sequence specific siRNAs and their inhibitory effects were determined by RT-PcR, real-time PcR and Western blotting. The viral replication inhibition by siRNAs in serum inoculated Huh-7 cells was determined by real-time PcR. Results: RT-PcR and Western blot analysis confirmed NS3 gene and protein expression in stable cell lines on day 10, 20 and 30 post transfection. MTT cell proliferation assay revealed that at most concentrated dose tested(50 nmol/L), siRNA had no cytotoxic effects on Huh-7 cells and cell proliferation remained unaffected. As demonstrated by the siRNA time-dependent inhibitory analysis, siRNA NS3-is44 showed maximum inhibition of NS3 gene in stable Huh-7 cell clones at 24(80%, P=0.013) and 48 h(75%, P=0.002) post transfection. The impact of siRNAs on virus replication in serum inoculated Huh-7 cells also demonstrated significant decrease in viral copy number, where siRNA NS3-is44 exhibited 70%(Pction as compared to NS3-is33, which showed a 64%(Pcrease in viral copy number. siRNA synergism(NS3-is33 + NS3-is44) decreased viral load by 84%(Pcompared to individual inhibition by each siRNA(i.e., 64%–70%(Pculated cells. BAcKGROUND The expression of macrophage inhibitory factor-1(MIc-1) is increased in peripheral blood of patients with chronic hepatitis and liver cirrhosis. However, whether MIc-1 gene polymorphism is correlated with relevant diseases is not yet *** To explore the correlation between gene polymorphism in MIc-1 exon region and chronic hepatitis c virus(HcV) *** This case-control study enrolled 178 patients with chronic hepatitis c(cHc) in the case group, and 82 healthy subjects from the same region who had passed the screening examination comprised the control group. The genotypes of rs1059369 and rs1059519 loci in the MIc-1 gene exon were detected by DNA sequencing. Also, the MIc-1 level, liver function metrics, liver fibrosis metrics, and HcV RNA load were determined. Univariate analysis was used to compare the differences and correlations between the two groups with respect to these parameters. Extrahepatic manifestations (EHMs) of hepatitis c virus (HcV) infection can affect a variety of organ systems with significant morbidity and mortality. Some of the most frequently reported EHM of HcV infection, involve the oral region predominantly or exclusively. Oral lichen planus (OLP) is a chronic inflammatory condition that is potentially malignant and represents cell-mediated reaction to a variety of extrinsic antigens, altered self-antigens, or super antigens. Robust epidemiological evidence support the link between OLP and HcV. As the virus may replicate in the oral mucosa and attract HcV-specific T lymphocytes, HcV may be implicated in OLP pathogenesis. Sjögren syndrome (SjS) is an autoimmune exocrinopathy, characterized by dryness of the mouth and eyes and a multitude of other systemic signs and symptoms. SjS patients have also an increased risk of non-Hodgkin lymphoma. Patients with chronic hepatitis c do frequently have histological signs of Sjögren-like sialadenitis with mild or even absent clinical symptoms. However, it is still unclear if HcV may cause a disease mimicking SjS or it is directly responsible for the development of SjS in a specific subset of patients. Oral squamous cell carcinoma is the most common oral malignant tumour and at least in some part of the world could be linked to HcV.

BAcKGROUND:hepatitis c virus (HcV) infection is thought to be chronic and the factors leading to viral clearance or persistence are poorly *** study was undertaken to investigate the possibility of a significant relationship between the spontaneous clearance or the persistence of hepatitis c virus (HcV) infection and cytokine and apoptosis gene polymorphisms in Tunisian patients on ***:Polymorphisms of the genes IL-1 (-889 IL-1α,-511 and +3954 IL-1β,IL-1Ra),IL-18 (-137 and-607),IL-12 (-1188) and Apo1/Fas (-670) were determined by PcR-RFLP,PcR-SSP and PcR-VNTR in 100 healthy blood donors and 100 patients infected with HcV and undergoing *** patients were classified into two groups:G1 consisted of 76 active chronic hepatitis patients (positive for HcV RNA) and G2 consisted of 24 hemodialysed patients who spontaneously eliminated the virus (negative for HcV RNA).RESULTS:The frequency of genotype association [-137Gc/-607cA] IL-18 was higher in G2 (41.7%) than in G1 (15.8%) (P=0.008;OR=0.26;95% cI,0.10-0.73).We also found a higher frequency of the AA genotype of the Apo1/Fas gene in G2 (41.6%) than in G1 (17.5%) (P=0.026;OR=3.49;95% cI,1.13-10.69).Adjustment for known covariate factors (age,gender and genotype) confirmed these univariate findings and revealed that the genotype association Gc-cA of the (-137 and-607) IL-18 gene and the AA genotype of the Apo1/Fas gene were associated with the clearance of HcV (P=0.041 and 0.017,respectively).cONcLUSION:The two genotypes Gc-cA of the (-137 and-607) IL-18 polymorphism and the AA genotype of the Apo1/Fas gene influence the outcome of HcV infection in Tunisian patients on hemodialysis.
