Pentoxifylline enhances the protective effects of hypertonic saline solution on liver ischemia reperfusion injury through inhibition of oxidative stress

BACKGROUND：Liver ischemia reperfusion（IR）injury triggers a systemic inflammatory response and is the main cause of organ dysfunction and adverse postoperative outcomes after liver ***（PTX）and hypertonic saline solution（HTS）have been identified to have beneficial effects against IR *** study aimed to investigate if the addition of PTX to HTS is superior to HTS alone for the prevention of liver IR ***： Male Wistar rats were allocated into three groups. Control rats underwent 60 minutes of partial liver ischemia, HTS rats were treated with 0.4 mL/kg of intravenous 7.5% NaCl 15 minutes before reperfusion, and HPTX group were treated with 7.5% NaC1 plus 25 mg/kg of PTX 15 minutes before reperfusion. Samples were collected after reperfusion for determination of ALT, AST, TNF-α, IL-6, IL-10, mitochondrial respiration, lipid peroxidation, pulmonary permeability and myeloperoxidase. RESULTS： HPTX significantly decreased TNF-α 30 minutes after reperfusion. HPTX and HTS significantly decreased ALT,AST, IL-6, mitochondrial dysfunction and pulmonary myelo- peroxidase 4 hours after reperfusion. Compared with HTS only, HPTX significantly decreased hepatic oxidative stress 4 hours after reperfusion and pulmonary permeability 4 and 12 hours after reperfusion. CONCLUSION： This study showed that PTX added the beneficial effects of HTS on liver IR injury through decreases of hepatic oxidative stress and pulmonary permeability.

Garcinia cambogia extract(GC)with its active component consisting of hydroxycitric acid(HCA)is widely utilized for weight *** HCA salts are available,including calcium,magnesium,potassium and mixtures of ***,these salts exhibit different properties with some,but not all,improving glucose tolerance and blood ***,obesity-prone C57BL/6J mice were fed a high-fat diet(HFD,45 kcal%fat)with or without GC(1%,w/w)for 16 *** active arm reduced visceral fat,adipocyte size and serum glucose,yet purportedly also exhibited hepatic collagen accumulation,lipid peroxidation and increased mRNA levels of genes related to oxidative *** latter findings are at odds with a large body of animal and human studies that have been conducted on the safety and efficacy of *** literature shows HCA to be protective against the liver toxicity associated with ethanol and dexamethasone administration,and to maintain serum aspartate aminotransferase,alanine aminotransferase and alkaline phosphatase at near normal *** both animal and clinical literature,elevated intakes of HCA per se have not led to signs of inflammation or *** compound has been found to reduce markers of inflammation in brain,intestines,kidney and serum.

Polysaccharide isolated from wax apple suppresses ethyl carbamate-induced oxidative damage in human hepatocytes

Wax apple(Syzygium samarangense)has received growing research interest for its high nutritional and medicinal value due to its constituents such as polysaccharide,organic acids,flavonoids,minerals,and other *** this study,wax apple polysaccharide(WAP)was isolated from this plant and its protective effect against ethyl carbamate(EC)-induced oxidative damage was evaluated in human hepatocytes(L02 cells).Firstly,a series of analyses such as high-performance liquid chromatography(HPLC),high-performance gel permeation chromatography(HPGPC),Fourier transform infrared spectroscopy(FT-IR),gas chromatography/mass spectrometry(GC/MS),and 1H and 13C nuclear magnetic resonance(NMR)were conducted to identify the structure of,in vitro cell experiments were performed to verify the protective effects of WAP against EC-induced cytotoxicity,genotoxicity,and oxidative damage in L02 *** results revealed that WAP is composed of mannose,rhamnose,glucuronic acid,galacturonic acid,glucose,galactose,arabinose,and fucose in a molar ratio of 2.20:3.94:4.45:8.56:8.86:30.82:39.78:*** a combination of methylation and NMR spectroscopic analysis,the primary structure of WAP was identified as Araf-(1→,Glcp-(1→,→2)-Araf-(1→,→3)-Galp-(1→,→3)-Araf-(1→,and→6)-Galp-(1→.Cell experiments indicated that WAP exhibited significant protective effects on EC-treated L02 cells via suppressing cytotoxicity and genotoxicity,reducing reactive oxygen species(ROS)and O2?-formation,as well as improving mitochondrial membrane potential(MMP)and glutathione(GSH).In a nutshell,WAP has the potential as an important therapeutic agent or supplement for hepatic oxidative ***,further studies are needed to prove the above effects in vivo at the biological and clinical levels.
