Exosomes are membrane bound extracellular vesicles that play an important role in many biological *** they have great application value,exosome isolation is still considered a major scientific *** the present study,a novel separation strategy for exosomes is proposed based on the specific interaction between immobilized peptide ligands and phosphatidylserine moieties which are highly abundant on the surface of *** the new affinity method,intact model exosomes can be recovered with a high yield in a short processing *** purity of exosome samples enriched from serum by the affinity method is far higher than that isolated by ultrafiltration,and similar to that obtained by density gradient centrifugation and ***,the variety of contaminants co-isolated by the affinity method is relatively low due to its specific separation *** analysis of exosomes isolated by the affinity method from the serum of healthy,hepatocellular carcinoma patients,and intrahepatic cholangiocarcinoma patients was performed to prove the applicability of this *** conclusion,our novel strategy shows characteristics of easy preparation,high specificity,and cost-effectiveness,and provides a promising approach for exosome isolation which should have wide applications.

Objective: To evaluate the hemodynamic changes of hepatic artery (HA), portal vein (PV) and tumors in hepatic cancer patients treated by cluster electrode radio-frequency ablation with the aid of color Doppler flow imaging (CDFI). Methods: The hemodynamic changes of HA, PV and 42 tumors in 30 cases of hepatic cancer were investi- gated by CDFI one week before and after cluster e- lectrode radio-frequency ablation. Results: One week after radio-frequency ablation, the velocity of HA decreased (Pcancer nodes (83.0 % of all 42 nodes). After radio- frequency ablation, blood signals were reduced in 15 nodes and disappeared in 14 nodes. Early investiga- tion implied that the decrease of blood supply was parallel with the reduction of node size. However, the outcome in case of huge nodes with double blood supply was not as promising as those small nodes. Conclusion: CDFI is useful to assess blood supply in ablation of hepatic cancer by using cluster electrode radio-frequency therapy.

Objective： The aim of the study was to observe the transfection efficacy of hepatitis B virus envelope （HBVE） and evaluate its ability as a gene transfer vector for liver cancer cells. Methods： To obtain HBVE, the supematant fluid of HepG 2.2.15 cells was mixed with a PEG8000 solution for concentration and was inactivated by β-propiolactone. The acquired HBVE was used to pack plRES2-EGFP to test its package ability. Then, we examined its quantity and quality with ELISA, PCR, SDS-PAGE and electron microscopy. The plRES2-EGFP was packed with HBVE and obtained the product HBVE-GFP. The plRES2-EGFP was packed with liposome and obtained the product liposome-GFP. HBVE-GFP and liposome-GFP were used to transfer HepG 2 cells to study the transfection efficiency. HBVE-GFP was used to transfer HepG 2, A549, HeLa and FB cells to study the targeting ability. The green fluorescent protein （GFP） expression was observed under a fluorescent microscope. The rate of GFP positive cells was determined by flow cytometry. Results： 1. The acquired HBVE could retain the surface protein HBsAg ＋ pre S1 ＋ pre S2 and had no virus DNA. It had good package ability for plRES2-EGFP. 2. Transfection efficiency： The GFP could be observed in both the liposome group and HBVE group under the fluorescent microscope. But the HBVE group had a higher fluorescent intensity than liposome group. The transfection rate of liposome group was 49.97% ＋ 2.37% while the HBVE group was 70.65% ＋ 3.15% and the fluorescent intensity of the HBVE group was 3-4 times （P = 0.000） for liposome group with the determination of flow cytometry. 3. Targeting ability： The GFP could be observed in the four groups under the fluorescent microscope. The HepG 2 group had the highest fluorescent intensity among the four groups. The transfection rate of HepG 2 group was 71.35% ＋ 0.03% which was highly expressed than other groups （P = 0.000） and the fluorescent intensity of the HepG 2 group was 2-3 times （P = 0.000） for the other 3 groups

Background and Objective:Geometric uncertainties resulted from treatment positioning variation and respiratory motion limit the precision of intensity-modulated radiotherapy(IMRT) for hepatic cancer,which could substantially lead to an increased volume of irradiated normal *** breathing control(ABC) is a promising and effective method to immobilize the liver by minimizing the breathing motion during a course of *** with the kilovoltage(KV) cone-beam computed tomography(CBCT) technology,high precision hepatic cancer IMRT could be realized using the ABC *** aim of this study is to evaluate the inter-fractional reproducibility of the liver position using an active breathing coordinator in patients with unresectable hepatic carcinoma treated with IMRT under the guidance of CBCT technology. Objective：To observe the efficacy of Chinese medicine comprehensive therapeutic project in treating the middle/late stage primary hepatic carcinoma（PHC）.Methods：With prospective randomized controlled design, 97 patients with PHC were assigned to the test group（49 cases） treated with Chinese medicine comprehensive therapy using Oleum fructus bruceas intervention combining oral intake of Ganji Decoction（肝积方） and external application of Ailitong（癌理通）,and the control group（48 cases） treated with chemotherapeutic agents combining iodized oil chemo-embolization and *** immediate and long-term efficacy,adverse reaction,pain-relieving initial time（PRIT） and pain-relieving sustained time（PRST） of the treatment,as well as the change in patients＇ quality of life（QOL） were ***：The difference between the two groups in illness control rate was statistically insignificant（P〉0.05）,but the adverse reaction occurence rate in the test group was lesser than that in the control group（P〈0.05）.PRIT was insignificantly different in the two groups（P〉0.05）,but the PRST was significantly superior in the test group than that in the control group（10.37±2.18 h vs 7.78±1.95 h,P〈0.01）.After treatment,the increased Karnofsky scores in the test group indicated that the patients＇ somatic activity,symptoms and QOL were improved significantly,which were significantly superior to those in the control group（P〈0.05）.The survival rate in the two groups was similar at the 3rd month after treatment,but the test group did show superiority in terms of half- and 1-year survival rate（65.9%vs 42.5%and 38.6%vs 18.1%,respectively,P〈0.05）.The median survival time in the test group was 8.9 months and that in the control group was 5.3 ***：Chinese medicine comprehensive therapy is an effective treatment for the middle/late stage patients of PHC,and it could extend the PRST,improve the patients＇ QOL and long-term survival with less adverse reaction.
