To investigate the effect of Jianpiyiqi prescription on the expression of heat shock proteins and the changes of HSPs in gastric ulcerated rats, the rat model of chronic gastric ulcer was induced by acetic acid. The SABC immunohistochemical method was used to observe HSP70 of mucosa around the gastric ulcer. Imaging analysis was performed. Western dot blot was used to detect HSPs contents in the plasma and gastric mucosal homogenate in each group. The results showed that HSP70 contents of the mucosa around the gastric ulcer in the model group and ranitidine treated group were increased as compared with control group . Jianpiyiqi could increase the expression of HSP70 of the mucosa around the gastric ulcer further as compared with that in the model group and ranitidine treated group . The HSP70 contents in the serum and mucosa in the model group and ranitidine treated group were increased as compared with control group ( P <0.01, P <0 05 respectively). HSP70 of serum and mucosa in the Jianpiyiqi treated group was higher than in the model group and ranitidine treated group ( P <0.05, P <0.01 respectively).It was concluded that HSP might play a role in the process of pathophysiology of gastric ulcer. Jianpiyiqi could enhance gastric ulcer healing through the protective mechanism of HSPs.

Objective To investigate the manifestations that heat shock proteins(HSPs) possess in the pathogenesis of asthma and chronic bronchitis.Methods Using reverse transcription DNA polymerase chain reaction(RT PCR), we investigated the expression levels of HSP70, HSP90α and HSP90β genes in peripheral blood mononuclear cells(PBMC) at natural state and after heat shock in 14 healthy volunteers, 21 patients with asthma and 18 patients with chronic bronchitis.Results No HSP70 gene but HSP90α and HSP90β expressions were found in non heat shocked PBMC of normal control; HSP90α and HSP90β genes may be expressed in PBMC of patients no matter whether they were in acute episode or not. Expression of HSP70 was found in PBMC of patients in acute episodes and of three symptoms free patients with Aas 3, step 2 No expression of HSP70 gene was found in PBMC of patients in convalescent period but in PBMC of patients in acute episode. HSP90α and HSP90β genes were expressed in PBMC of the two patients groups; After heat shock, expressions of the three genes increased in amount significantly in PBMC of all normal controls and patients. Conclusion Expression of HSP70 gene in PBMC of asthmatic patients and chronic bronchitis was different, indicating that HSPs, especially HSP70 might be involved in the pathogenesis of asthma.

Bemisia tabaci (Gennadius) (Homoptera: Aleyrodidae) is the most destructive invasive pests in agricultural production and has a high tolerance to heat. heat shock proteins play an essential role in life activities such as growth and development, reproduction and diapause of B. tabaci. At the same time, they are also crucial in resisting adverse environments and in adaptive evolution. The expression of heat shock protein in B. tabaci is not only related to temperature, but also to the tolerance of the environment. After receiving external stimuli, the expression level can be increased or decreased to maintain the stability of cells in vivo. This paper reviews the classification, biological characteristics, biological functions, and research status of HSPs in recent years. This mini-review will provide helpful information related to the use of heat shock proteins to study the occurrence and damage of B. tabaci. This has important theoretical and practical significance for revealing Hsps in explaining the population expansion mechanism of B. tabaci invasion and predicting population dynamics.

Background:Ischemia-reperfusion injury(IRI)poses a significant challenge to liver transplantation(LT).The underlying mechanism primarily involves overactivation of the immune system.heat shock protein 110(HSP110)functions as a molecular chaperone that helps stabilize protein structures.Methods:An IRI model was established by performing LT on Sprague-Dawley rats,and HSP110 was silenced using siRNA.Hematoxylin-eosin staining,TUNEL,immunohistochemistry,ELISA and liver enzyme analysis were performed to assess IRI following LT.Western blotting and quantitative reverse transcription-polymerase chain reaction were conducted to investigate the pertinent molecular changes.Results:Our findings revealed a significant increase in the expression of HSP110 at both the mRNA and protein levels in the rat liver following LT(P<0.05).However,when rats were injected with siRNAHSP110,IRI subsequent to LT was notably reduced(P<0.05).Additionally,the levels of liver enzymes and inflammatory chemokines in rat serum were significantly reduced(P<0.05).Silencing HSP110 with siRNA resulted in a marked decrease in M1-type polarization of Kupffer cells in the liver and downregulated the NF-κB pathway in the liver(P<0.05).Conclusions:HSP110 in the liver promotes IRI after LT in rats by activating the NF-κB pathway and inducing M1-type polarization of Kupffer cells.Targeting HSP110 to prevent IRI after LT may represent a promising new approach for the treatment of LT-associated IRI.

The overexpression of heat shock proteins(HSPs)in tumor cells can activate inherent defense mechanisms during hyperthermia-based treatments,inducing thermoresistance and thus diminishing the treatment efficacy.Here,we report a distinct“non-inhibitor involvement”strategy to address this issue through engineering a calcium-based nanocatalyst(G/A@CaCO_(3)-PEG).The constructed nanocatalyst consists of calcium carbonate(CaCO_(3))-supported glucose oxidase(GOD)and 2D antimonene quantum dots(AQDs),with further surface modification by lipid bilayers and polyethylene glycol(PEG).The engineered G/A@CaCO_(3)-PEG nanocatalyst features prolonged blood circulation,which is stable at neutral pH but rapidly degrades under mildly acidic tumor microenvironment,resulting in rapid release of drug cargo in the tumor microenvironment.The integrated GOD effectively catalyzes the depletion of glucose for reducing the supplies of adenosine triphosphate(ATP)and subsequent down-regulation of HSP expression.This effect then augments the therapeutic efficacy of photothermal hyperthermia induced by 2D AQDs upon irradiation with near-infrared light as assisted by reversing the cancer cells’thermoresistance.Consequently,synergistic antineoplastic effects can be achieved via low-temperature photothermal therapy.Systematic in vitro and in vivo evaluations have demonstrated that G/A@CaCO_(3)-PEG nanocatalysts feature potent antitumor activity with a high tumor-inhibition rate(83.92%).This work thus paves an effective way for augmenting the hyperthermia-based tumor treatments via restriction of the ATP supply.

Despite immunotherapy involving immune checkpoint inhibitors(ICIs)have revolutionized cancer therapy,the clinical efficacy is limited due to ICI resistance.Pyroptosis is a gasdermin-mediated programmed cell death that enhances responses to ICIs.However,nontargeted elicitation of pyroptosis may induce systemic side effects and toxicity.Therefore,we reasonably design and construct a tumor-specific prodrug that combines the heat shock protein 90 inhibitor tanespimycin(17-AAG)with the photosensitizer chlorin e6(Ce6)to induce pyroptosis,by utilizing the high glutathione level in the tumor microenvironment.The released Ce6 and 17-AAG produce reactive oxygen species by laser triggering,which induces gasdermin E-mediated pyroptosis.Furthermore,17-AAG reduces myeloid-derived suppressor cells and sensitizes tumors to anti-programmed death^(-1)(PD-1)therapy.Thus,our prodrug strategy achieves tumor-targeted pyroptosis to suppress tumor growth,thereby improving the response to anti-PD-1 therapy and extending the survival of 4T1 breast tumor-bearing mice.Consequently,this pyroptosis-based prodrug represents a novel strategy for enforcing immunogenic photodynamic therapy.

Near-infrared(NIR)-light-triggered photothermal therapy(PTT)is a promising treatment for breast cancer.However,its therapeutic efficiency is often compromised due to the heatinduced up-regulation of heat shock proteins,which confer photothermal resistance.To solve this urgent problem,PEGylated two-dimensional boron nanosheets(B-PEG)-which allow both multimodal imaging and photothermal conversion-were loaded with gambogic acid(GA),which can inhibit heat shock protein 90(Hsp90).Experimental findings indicated that this combination of B-PEG and GA could serve as an integrated drug delivery system for cancer diagnosis and treatment.It could be used to administer mild PTT as well as chemotherapy for breast cancer,provide improved anti-tumor effects,and reduce the toxicity of PTT,all while inhibiting breast cancer growth.This drug delivery system could offer a novel tool for administering chemotherapy combined with PTT while avoiding the adverse effects of traditional PTT.

Temperature is an important factor affecting the growth,development,and survival of organisms.The effects of temperature stress on aquatic organisms have received increasing attention,as these organisms are mostly poikilotherms and their body temperature are directly corresponding changes with ambient temperature,resulting in they are easily exposed in temperature stress.However,little is known about the effects of high temperature on Sipuncula.In this study,we investigated the effects of acute heat stress on malondialdehyde(MDA)concentration,the activities of antioxidant(superoxide dismutase and glutathione peroxidase)and immunity-related(acidic and alkaline phosphatase)enzymes,heat shock protein 70(hsp70)and hsp90 gene expression,and the histological structure of the sipunculid Phascolosoma esculenta.Within the coelom fl uid,the MDA concentration and all detected antioxidant enzyme activities increased during high temperature stress;signifi cant increases were also observed here and in the intestine in the hsp70 and hsp90 mRNA expression levels.These results indicated that acute heat stress caused oxidative stress;antioxidants and heat shock proteins probably act to protect P.esculenta against oxidative damage,constituting part of its physiological mechanism for adaptation to high temperatures.In addition,the increased activity of the acidic and alkaline phosphatases indicated eff ects on its nonspecifi c immune system.Furthermore,damaged tissue structures were observed in the body wall,retractor muscle,intestine,and nephridium after 96 h of 40-℃ stress.The damaged cells of these tissues showed obvious condensed chromatin around the nuclear membrane.This histological damage suggests that heat stress could affect movement,food absorption,digestion,and excretion in P.esculenta.These results elucidate the effects of temperature stress on P.esculenta and its physiological response mechanisms and provide practical indicators for assessing heat stress status and determining suitable culture temperatures for P.esculen

Plant Dna JA proteins act as molecular chaperones in response to environmental stressors.The purpose of this study was to characterize the function and regulatory mechanisms of Dna JA genes in soybean.Gene expression profiles in various soybean tissues at various stages of development indicated that Gm Dna JAs function in the coordination of stress and plant hormone responses.Gm Dna JA6 was identified as a candidate regulator of saline and alkaline stress resistance and Gm Dna JA6 overexpression lines showed increased soybean saline and alkaline tolerance.Dna J interacted with Hsp70,and Gm Hsp70 increased the saline and alkaline tolerance of plants with chimeric soybean hairy roots.

Photothermal therapy(PTT)induces thermoresistance through cellular heat shock response,which impairs the therapeutic efficacy of the PTT.To resolve this problem,we developed a photothermal theranostics(denoted as PMH),which integrated the photothermal conversion agent of PdMo bimetallene with histone deacetylase 6(HDAC6)selected inhibitor(ACY-1215),showing the synergistic antitumor effect both in vitro and in vivo.Mechanistically,under the photoacoustic imaging(PA)navigation,the released ACY-1215 triggered by NIR laser irradiation decrease the heat shock proteins(HSPs)expression and weaken the HDAC6-regulated HSP90 deacetylation,thus hindering the degradation of PTT-induced misfolded or unfold proteins through proteasome dependent pathway.Moreover,mild photothermal therapy(mPTT)treatment compromised the autophagy,which induced by HDAC6 inhibition,leading to mPTTinduced misfolded or unfold proteins further accumulation.Given that inhibition of HDAC6 plus m PTT contribute to tumor eradication.This study develops a promising combination strategy based on m PTT for future cancer treatment.
