Interfamily pregnancy and expression of CD57, CD68 in deciduas between golden hamster and mouse

Pregnancy between different species is one of the key steps to interspecific somatic cell cloning. Although interspecific clone embryos have been constructed, they could not develop to birth after being transferred to recipi-ents. In order to clarify the mechanism of this phenomenon, interfamily pregnancy between golden hamste (Mesocricetus auratus) and mouse (Mus musculus) was studied. Co-culture results indicated that the adhesion ratios of golden hamster blastocysts on mouse uterine epithelia monolayer 12, 24, 48 and 72 h after co-culture were all significantly lower than those of mouse blastocysts. The outgrowth ratios of golden hamster blastocysts on mouse uterine epithelia monolayer 48, 72 h after co-culture were both significantly lower than those of mouse blastocysts (P hamster抯 blastula could be implanted and develop to D 11 of pregnancy after being transferred to mouse uterus (the 7th day after embryo transfer). Compared to the transfer of mouse embryo to mouse uterus, the successful ratio of interfamily embryo transfer was lower and the bulk of fetus was smaller than that of intraspecific fetus. Compared to intraspecific preg-nancy of mouse, the remote decidual tissue of interfamily pregnancy on D8 is looser. At the same time, expressions of CD57 and CD 68 in remote deciduas were both higher than those in the secondary deciduas in both intraspecific and interfamily pregnancy. However, expressions of the two molecules in interfamily pregnancy were lower than those in intraspecific pregnancy. These results showed that interfam-ily pregnancy could be established between golden hamster and mouse. But the development of fetus in interfamily pregnancy was slower than that in intraspecific pregnancy. The expression difference of CD57 and CD68 indicates the difference of immunoreaction between interfamily and in-traspecific pregnancy, which may be one of the reasons lead-ing to interfamily pregnancy termination.

TELOMERASE ACTIVITY DURING 7,12-DIMETHYLBENZ [a] ANTHRACENE-INDUCED hamster BUCCAL POUCH CARCINOGENESIS

Objective: To investigate the roles of telomerase activity (TA) in relation to hamster buccal pouch tumor progression. Methods: male hamster were treated three times weekly with 0.5% of 7, 12-dimethyl-benzanthracene (DMBA) over a IS weeks experimental period. hamsters were sacrificed at 3, 6, 9, 12 and 15 weeks after treatment. Telomerase activity of hamster buccal pouch tissue were measured along with the analyses of the formation of DMBA-induced hamster buccal pouch tumors. Results: DMBA-induced squamous cell carcinomas were found at the 6th week after dosing. Telomerase activity elevation began at the 3rd week and was increasing to a plateau at the 12th week. Conclusion: Our results show that telomerase activity in the target tissue may be detected at the early stage of the DMBA-induced hamster buccal pouch tumor formation and suggests that telomerase activity may be used as a biomarker for an early clinical detection of buccal pouch cancer.

Three transfection reagents, Lipofectamine® 2000, TransIT-PRO® and linear 25 kDa polyethylenimine were evaluated for transient expression of enhanced green fluorescent protein in Chinese hamster ovary cells. TransIT-PRO® was found to be more efficient under the examined conditions, but comes at an increased cost compared to the widely used PEI.

EXPERIMENTAL STUDY ON MICROWAVE RADIATION IN BLOCKING LEUKOPLAKIA FROM CANCERATION IN GOLDEN hamster CHEEK POUCH

Objective Lately, microwave radiation (MR) has been successfully used to cure a variety ofdiseases including oral mucosal diseases and oral tumors in stomatology. Preventive MR of precancerous lesionsfrom canceration is the goal of this study which had been rarely reported. Methods Between leukoplakia modelanimals and MR treated model animals, the cancer incidences were compared and the macroscopic and microscopicpathology were detailed. The functional parameters of the microwave apparatus were analysed to yield the optimumdata for future clinical use. Results The cancer blocking effect of MR was proved by the incidence of canceration3.5 times less frequent in MR treated animals. Conclusion Analysis of the macroscopic and microscopicpathology may disclose the mechanism of the therapeutic MR.

To elucidate the mechanism of sterility induced by gossypol,we studied the relationship between the activities of acrosomal enzymes and their fertilizing capacity in the hamster. The results showed that the ability of spermatozoa to penetrate into bovine cervical mucus, hyper-activated motility(HAM)and fertility in vivo were significantly inhibited when spermatozoa were exposed to gossypol (2.5μg0μg/mL)for 15 min in ***,following administration of gossypol (12.5mg/kg/day)for 6 weeks,sperm motility,HAM and rate of fertilization in vitro by the hamster cauda epididymal spermatozoa were significantly decreased and the extracts of testis delayed dispersion of the cumulus oophorus cells, suggesting that hyaluronidase and other acrosomal enzymes might be inhibited by *** addition, acrosin and arylsulfatase activities were also markedly *** data show that the inhibition of acrosin and arylsulfatase activities is the main cause of gossypol-induced infertility .The inhibition was dependent upon gossypol dose and the duration of ***, the assay of acrosin and arylsulfatase activities may provide a useful tool for monitoring sterility induced by gossypol.

Efficacy of cefepime,ertapenem and norfloxacin against leptospirosis and for the clearance of pathogens in a hamster model

Animals and humans with severe leptospirosis may require empirical *** many antibiotics are active against multiple leptospira serovars in vitro,their efficacy in vivo is *** evaluated the efficacy of cefepime（daily dose:2,5,10,and 20 mg/kg）,ertapenem（daily dose:2.5,5,and 10 mg/kg） and norfloxacin（daily dose:20,40,and 80 mg/kg） for the treatment of leptospirosis and the ability to clear leptospira in target organs（liver,kidney,lung,heart,and spleen） in a lethal hamster model using Leptospira interrogans serovar *** histopathology of infected kidney,lung and liver was also evaluated using hematoxylin and eosin stain（H & E stain）.All untreated animals,serving as a negative control,died with leptospira existing in the target organs between the 5th and 7th day after *** of the treated groups displayed improved survival compared to the untreated group and demonstrated a dose-dependent decrease in the presence of leptospira in the target *** showed survival benefit comparable to the standard treatment,*** conclude that all of the antibiotics tested in vivo produce a statistically significant survival advantage,alleviate tissue injury and decrease the abundance of leptospira in target organs.

Dose-dependent and combined effects of N-methyl-D-aspartate receptor antagonist MK-801 and nitric oxide synthase inhibitor nitro-L-arginine on the survival of retinal ganglion cells in adult hamsters

This study investigated the effects of daily intraperitoneal injections of N-methyl-D-aspartate receptor antagonist MK-801 and nitric oxide synthase inhibitor nitro-L-arginine （L-NA） on the survival of retinal ganglion cells （RGCs） at 1 and 2 weeks after unilateral optic nerve transection in adult hamsters. The left optic nerves of all animals were transected intraorbitally 1 mm from the optic disc and RGCs were retrogradely labeled with Fluorogold before they received different daily dosages of single MK-801 or L-NA as well as daily combinational treatments of these two chemicals. All experimental and control animals survived for 1 or 2 weeks after optic nerve transection. Our results revealed that the mean numbers of surviving RGCs increased and then decreased when the dosage of MK-801 （1.0, 3.0 and 4.5 mg/kg） and L-NA （1.5, 3.0, 4.5 and 6.0 mg/kg） increased at both 1 and 2 weeks survival time points. Daily combinational use of 1.0 mg/kg MK-801 and 1.5 mg/kg L-NA lead to a highest RGC number that was even higher than the sum of the RGC numbers in 1.0 mg/kg MK-801 and 1.5 mg/kg L-NA subgroups at 2 weeks. These findings indicated that both MK-801 and L-NA can protect axotomized RGCs in a dose-dependent manner and combinational treatment of these chemicals possesses a potentiative and protective effect.

Pravastatin activates PPARα/PPARγexpression in the liver and gallbladder epithelium of hamsters

BACKGROUND:Our earlier study with cultured gallbladder epithelial cells demonstrated that statins(HMG-CoA reductase inhibitors)activate the expression of PPARαand PPARγ, consequently blocking the production of pro-inflmmatory *** present study used hamsters to investigate the effects of pavastatin on PPARα/PPARγexpression in the liver and gallbladder epithelium,and to determine whether pravastatin suppresses cholesterol crystal formation in the gallbladder. METHODS:A total of 40 Golden Syrian male hamsters(4 weeks old)were randomly assigned to four groups(basal diet control; basal diet+pavastatin;high cholesterol diet;high cholesterol diet+pravastatin).All hamsters were 11 weeks old at the end of the *** liver,gallbladder and bile were harvested. Immunohistochemical staining and Western blotting for PPARαand PPARγwere performed in the liver and gallbladder. A drop of fresh bile was examined for cholesterol crystals under a microscope. RESULTS:In the gallbladder and liver of the hamsters, pravastatin activated the PPARαand PPARγexpression of gallbladder epithelial cells and hepatocytes,and particularly the response of PPARγwas much stronger than that of PPARα. Pravastatin suppressed the formation of cholesterol gallstones or crystals in the gallbladder. CONCLUSION:Pravastatin is an effective medication to activate PPARs(especially PPARγ)in the liver and the gallbladder epithelium of hamsters,and contributes to the prevention of gallstone formation.
