以hptlc分析平台,分别用像素分析法和光密度法,建立并对比两种快速检测茶饮料中EGCG含量的方法。以3种市售茶饮料为代表性样品,对比两种方法的应用性能,评估像素分析替代仪器依赖的光密度检测作为茶饮料中EGCG含量检测的新方法的先进性。结果显示,hptlc-像素分析方法、hptlc-光密度方法检测的定量限分别为6.8,5 mg/kg,均可以定量茶饮料中EGCG。两种方法目标物在200~1200 ng/斑点浓度范围内线性相关系数相差不大,均在0.999以上。两种方法的回收率均满足要求,hptlc-像素分析方法测定3种样品加标回收率分别在102.4%~110.8%,102.3%~118.8%,100.3%~113.5%之间,hptlc-光密度方法测定3种样品加标回收率分别在:103.5%~110.8%,95.3%~111.2%,99.3%~111.2%之间,表明两种方法具有良好的准确性。两种方法的精密度也相差不大,hptlc-像素分析方法RSDhptlc-光密度方法RSDhptlc检测技术中,简单易用的像素分析可以较好地代替昂贵的仪器化光密度扫描,提高方法的性价比和实用性。

Macrothelypteris torresiana is a fern species belonging to the family Thelypteridaceae. The present study was conducted to evaluate hepatoprotective potential of ethanol extract from M. torresiana aerial parts(EEMTAP)and detect the polyphenolic compounds present in the extract using high performance thin layer chromatography(hptlc). Hepatoprotective potential of EEMTAP were tested at doses of 300 and 600 mg/kg, per os(p.o.), on Wistar albino rats. The extract and silymarin treated animal groups showed significant decrease in activities of different biochemical parameters like serum glutamic oxaloacetic transaminase(SGOT), serum glutamate-pyruvate transaminase(SGPT), and alkaline phosphatase(ALP), which were elevated by carbon tetrachloride(CCl_4) intoxication. The levels of total bilirubin and total protein along with the liver weight were also restored to normalcy by EEMTAP and silymarin treatment. After CCl_4 administration, the levels of hepatic antioxidant enzymes such as glutathione(GSH) and catalase(CAT) were decreased whereas the level of hepatic lipid peroxidation(LPO) was elevated. The levels of these hepatic antioxidant enzymes were also brought to normalcy by EEMTAP and silymarin treatment. Histological studies supported the biochemical findings, and treatment with EEMTAP at doses of 300 and 600 mg/kg, p.o. was found to be effective in restoring CCl_4-induced hepatotoxicity in rats. A simple hptlc analysis was conducted for the detection of polyphenolic compounds in EEMTAP, and the result revealed the presence of caffeic acid as phenolic acid and quercetin as flavonoid. The proposed hptlc method is simple and concise and provides a good resolution of caffeic acid and quercetin from other constituents present in EEMTAP.

The objective of this study was to evaluate the free radical scavenging potential and high performance thin layer chromatography （hptlc） fingerprinting of Indigofem tinctoria （I. tinctoria）, Phytochemical analysis was carried out using standard methods, and free radical scavenging activity of the plant was determined using 2,2-diphenyl-l-picrylhydrazy （DPPH）, nitric oxide （NO） and superoxide anion （O2-） radical scavenging capacities. hptlc plate was kept in CAMAG TLC Scanner 3 and the Rf values at fin- gerprint data were recorded by WlNCATS software, Aqueous extract of I. tinctoria reliably showed the total phenolics （267.2 ± 2.42 mg/g）, flavonoids （75.43 ± 3.36 mg/g） and antioxidants （349.11 ±8.04 mg/g）. The extract was found to have DPPH （52.08%）, NO （23.12%） and 02 （26.79%） scavenging activities at the concentration of 250 pg/mL and the results were statistically significant compared with ascorbic acid standard （p 〈 0.05）. hptlc results confirmed that the extract contained several potential active com- ponents such as phenols, flavonoids, saponins and terpenoids as the slides revealed multi-colored bands of varying intensities. This study confirmed that the plant had multipotential antioxidant and free ra- dicals scavenging activities.

目前,酚类抗氧化剂TBHQ的常用检测方法存在前处理烦琐、耗时长、单次只能测定一个样品的问题,因此亟需建立一种高通量、快速准确的检测方法。为此,将高效薄层色谱(hptlc)分离与1,1-二苯基-2-三硝基苯肼(DPPH)还原变色反应、图像像素定量分析相结合,建立了一种快速筛检酚类抗氧化剂TBHQ的方法。在此基础上,以食用油、面包和蛋糕为代表性样品,验证所建方法的实际应用性能。结果显示:所建方法检测TBHQ的检测限为70 ng/zone (35 mg/kg),定量限为140 ng/zone (70 mg/kg),定量限低于GB 2760—2014规定的使用限量(200 mg/kg);通过对比分析,样品提取物中共存的背景基质对目标物定量的干扰可以忽略,证明了该方法具有良好的选择性;目标物在150~550 ng/zone范围内表现出良好的线性(R^(2)=0.989 2);多次平行(n=3)测试和国标规定使用限量水平附近的加标回收率测试结果证明了该方法具有较好的精密度和准确度。综上,该方法具有基质耐受性强、定量准确、简便可靠且分析通量大等优点,特别适合粮油食品中各种抗氧化剂的快速定量筛查,具有凝炼成为相关领域国家标准的潜力和价值。

Objective:The study was conducted to develop and validate a high performance thin-layer chromatography(hptlc)-densitometric method for the quantitative analysis of morin in Maclura cochinchinensis heartwood collected from different locations in Thailand.Methods:hptlc analysis was performed on an aluminium sheet of silica gel 60 F254 using toluene:ethyl acetate:formic acid(36:12:7,volume percent)as a mobile phase.The densitometric scanning was performed at the wavelength 410 nm.hptlc method was validated according to ICH guideline.Results:The proposed hptlc method showed acceptable validation parameters.The content of morin in M.cochinchinensis heartwood collected from eight different provinces in Thailand were in the ranges of1.53%-2.73%.Conclusion:The simple and sensitive hptlc method was successfully developed and validated for determination of morin in M.cochinchinensis heartwood.The proposed hptlc method was found to be simple,fast and inexpensive,and can be used for the routine quality control of raw materials.

Objective:To determine the chemical profile and steroids composition of the medicinally important plant Aerva lanata(A.lanata) L.Methods:Preliminary phytochemicul screening was done by the method as Harborne described.hptlc studies were canied out as Harborne and Wagner et al described.The Ethyl acetate-ethanol-water(8:2:1.2) was employed as mobile phase for glycosides.Results:The desired aim was achieved using Chloroform-acetone(8:2) as the mobile phase.The methanolic extract of stem,leaves,root,flower and seeds of A.lanata showed the presence of 30 different types of steroids with 30 different Rf values from 0.04 to 0.97. Maximum number(11) of steroids has been observed in leaves followed by root(10).Conclusions: hptlc profile of steroids has been chosen here to reveal the diversity existing in A.lanata.Such finger printing is useful in differentiating the species from the adulterant and act as biochemical markers for this medicinally important plant in the pharma industry and plant systematic studies.

Objective:To develop a simple,selective,sensitive and accurate high-performance thin layer chromatography(hptlc)method to determine the quantity of hesperidin in different varieties of citrus fruits.Methods:The method was carried out in aluminum-backed silica gel 60 F_(254)plates with ethyl acetate-methanol-water 15:3:2(%,v/v)as mobile phase.Results:A compact band was obtained for hesperidin at R_f value of(0.40±0.04).The calibration plot was linear in the range of 100-800 ng/spot of hesperidin and the correlation coefficient of 0.9986was indicative of good linear dependence of peak area on concentration.Limit of detection(8.87ng/spot),limit of quantification(23.21 ng/spot),accuracy(less than 2%)and recovery(ranging from98.55-99.38)were found satisfactory.Conclusions:The method developed can be used for routine analysis of hesperidin in crude drug as well as in herbal and pharmaceutical dosage form containing citrus fruits as an ingredient.

Objective:To develop hptlc fingerprint profile of anti-inflammatory active extract fractions of Tribulus terrestris(family Zygophyllaceae).Methods:The anti-inflammatory activity was tested for the methanol and its fractions(chloroform,ethyl acetate,n-butanol and aqueous)and chloroform extract of Tribulus terrestris(aerial parts)by injecting different groups of rats(6 each)with carrageenan in hind paw and measuring the edema volume before and 1,2 and 3 h after carrageenan injection.Control group received saline i.p.The extracts treatment was injected i.***.in doses of 200 mg/kg 1 h before carrageenan administration.Indomethacin(30 mg/kg)was used as standard.hptlc studies were carried out using CAMAG hptlc system equipped with Linomat IV applicator.TLC scanner 3.Reprostar 3,CAMAG ADC 2 and WIN CATS-4 software for the active fractions of chloroform fraction of methanol extract.Results:The methanol extract showed good antiedematous effect with percentage of inhibition more than 72%,indicating its ability to inhibit the inflammatory mediators.The methanol extract was re-dissolved in 100 mL of distilled water and fractionated with chloroform,ethyl acetate and n-butanol.The four fractions(chloroform,ethyl acetate,n-butanol and aqueous)were subjected to anti-inflammatory activity.Chloroform fraction showed good anti-inflammatory activity at dose of 200 mg/kg.Chloroform fraction was then subjected to normal phase silica gel column chromatography and eluted with petroleum ether-chloroform,chloroform-ethyl acetate mixtures of increasing polarity which produced 15 fractions(F1-F15).Only fractions F1,F2,F4,F5,F7,F9,F11 and F14 were found to be active,hence these were analyzed with hptlc to develop their finger print profile.These fractions showed different spots with different R_f values.Conclusions:The different chloroform fractions F1,F2,F4,F5,F7,F9,F11 and F14 revealed 4,7,7,8,9,7,7 and 6 major spots,respectively.The results obtained in this experiment strongly support and validate the traditional uses of this Sudanese medic

Objective:To develop and validate a simple,accurate hptlc method for the analysis of 8-gingerol and to determine the quantity of 8-gingerol inZingiber officinaleextract and gingercontaining dietary supplements,teas and commercial creams.Methods:The analysis was performed on 10×20 cm aluminium-backed plates coated with 0.2 mm layers of silica gel 60 F254(E-Merck,Germany)with n-hexane:ethyl acetate 60:40(v/v)as mobile phase.Camag TLC Scanner III was used for the UV densitometric scanning at 569.Results:This system was found to give a compact spot of 8-gingerol at retention factor(Rf) value of(0.39依0.04)and linearity was found in the ranges 50-500 ng/spot(r2=0.9987).Limit of detection(12.76 ng/spot),limit of quantification(26.32 ng/spot),accuracy(less than 2%)and recovery(ranging from 98.22-99.20)were found satisfactory.Conclusions:The hptlc method developed for quantification of 8-gingerol was found to be simple,accurate,reproducible,sensitive and is applicable to the analysis of 8-gingerol in Zingiber officinaleextract and ginger-containing dietary supplements,teas and commercial creams.
