hla class i Expressions on Peripheral Blood Mononuclear Cells in Colorectal Cancer Patients

Objective： To investigate the expression change of human leukocyte antigen （hla） class i on human peripheral blood mononuclear ceils （PBMCs） at both mRNA and protein levels, and to evaluate its roles in the development of colorectal cancer （CRC）. Methods： in the present study, 50 patients with CRC, 35 patients with benign colorectal lesion and 42 healthy volunteers were enrolled. Expression levels of hla class i mRNA and protein were determined using real-time quantitative reverse transcription PCR （RT-PCR） and flow cytometry analysis, respectively. Results： The expression levels of hla class i mRNA and proteins were not influenced by age and gender. The relative ratios of hla class i mRNA were 0.99±0.27 in healthy controls, 0.76±0.19 in benign patients, and 0.48±0.21 in CRC patients. Mean fluorescence intensities of hla class i were 145.58±38.14 in healthy controls, 102.05±35.98 in benign patients and 87.44±34.01 in CRC patients. hla class i on PBMCs was significantly down-regulated at both mRNA and protein levels in patients with stage iii and iV CRC. CRC patients with lymph node metastasis also showed a decreased hla class i expression at protein level. Conclusion： hla class i expressions on PBMCs are associated with staging of CRC and lymph node metastasis. Monitoring the expression of hla class i on PBMCs may provide useful information for diagnosis and metastasis judgement of CRC.

Despite the growing knowledge of T cell responses in COViD-19 patients,there is a lack of detailed characterizations for T cell-antigen interactions and T cell ***,with a predicted peptide library from SARS-CoV-2 S and N proteins,we found that specific CD8+T cell responses were identified in over 75%of COViD-19 convalescent patients(15/20)and an epitope from the N protein,N361-369(KTFPPTEPK),was the most dominant epitope from our selected peptide ***,we discovered 2 N361-369-specific T cell receptors(TCRs)with high functional avidity that were independent of the CD8 *** TCRs exhibited complementary cross-reactivity to several presently reported N361-369 mutant variants,as to the wild-type ***,the natural functions of these TCRs in the cytotoxic immunity against SARS-CoV-2 were determined with dendritic cells(DCs)and the lung organoid *** found that the N361-369 epitope could be normally processed and endogenously presented by these different types of antigen presenting cells,to elicit successful activation and effective cytotoxicity of CD8+T cells ex *** study evidenced potential mechanisms of cellular immunity to SARS-CoV-2,and illuminated potential ways of viral clearance in COViD-19 *** results indicate that utilizing CD8-independent TCRs against SARS-CoV-2-associated antigens may provide functional superiority that is beneficial for the adoptive cell immunotherapies based on natural or genetically engineered T ***,this information is highly relevant for the development of the next-generation vaccines with protections against continuously emerged SARS-CoV-2 mutant strains.

Untreated human immunodeficiency virus(HiV)infections usually lead to death from AiDS,although the rate of the disease progression varies widely among *** cytotoxic T lymphocyte(CTL)response,which is restricted by highly polymorphic MHC class i alleles,plays a central role in controlling HiV *** is now recognized that the antiviral efficacy of CTLs at the single cell level is dependent on their antigen specificity and is important in determining the quality of host response to viruses so that the individual will remain ***,because of the extreme mutational plasticity of HiV,HiV-specific CTL responses are continuously and dynamically *** order to rationally design an effective vaccine,the questions as to what constitutes an effective antiviral CTL response and what characterizes a potent antigenic peptide to induce such responses are becoming highlighted as needing to be answered.
