Chemotherapy remains the main method for advanced pancreatic cancer.Chemotherapy resistance is an important for fact which influences the the therapeutic effect of pancreatic cancer.The resistance mechanism of chemotherapeutic drug such as gemcitabine which treats pancreatic canceris complicated and affected by genes and environment.Oridonin which could reverses drug resistance for pancreatic cancer has not been fully understood yet now.This study aims to assess the impact of oridonin on the resistance-related protein,apoptosis-related and energy metabolism in gemcitabine-resistant PANC-1/Gem cells through the experiments in vitro and in vivo.Gemcitabine-resistant PANC-1/Gem cells was induced by gradient concentration of gemcitabine.CCK-8 assay was used to detect the effect of Gemcitabine and oridonin on the proliferation of PANG-1 and PANC-1/Gem cells to determine the combination index of Gemcitabine and oridonin in PANC-1/Gem cells.Western blot analysis and immunofluorescence were used to detect the expression of Gemcitabine and oridonin in pancreatic cancer PANC-1/Gem cells resistance-related protein expression.The effects of Gemcitabine and oridonin on the apoptosis of PANC-1/Gem cellsweredetected by flow cytometry and Western blot analysis.Animal xenograft tumor assay was used to detect the effect of Gemcitabine and oridonin on pancreatic cancer in vivo.The effects of Gemcitabine and oridonin on ATP in pancreatic cancer PANC-1/Gem cells were detected by ATP Assay Kit.Western blot and immunofluorescence assay were used to detect the effects of Gemcitabine and oridonin on PANC-1/Gem cells density lipoprotein receptor protein(LRP1)expression.PANC-1/Gem cells were transfected with the lentiviral LV-siAFAPl-As1 and LV-NC.Clone formation assay and Western blot analysis were used to detect the effect of knocking down LRP1 on the proliferation of PANC-1/Gem cells.These results demonstrate that oridonin could reverse gemcitabine resistance inPANC-1/Gem cell by regulating the level of glutathione s-transferase pi(GsT

Background： several studies concerning the association between glutathione s-transferase P1 （GsTP1） Ilel05Val polymorphism and male infertility risk have reported controversial findings. The present study was aimed to explore this association using a recta-analysis. Methods： The PubMed, EMBAsE, China National Knowledge Infrastructure （CNKI）, and Wanfang databases were searched. Odds ratios （ORs） with 95% confidence intervals （C/s） were calculated to estimate the strength of the association. Results： A total of 3282 cases and 3268 controls in nine case-control studies were included. There was no significant association between GsTP1 llel05Val polymorphism and male infertility in the overall population, but significant associations were lbund under the dominant （OR = 1.23, 95% CI = 1.04-1.46, F = 32.2%） and heterozygote （OR = 1.29, 95% C1 - 1.08-1.53, F = 26.8%） models after excluding studies for which the data did not satisfy Hardy-Weinberg equilibrium （HWE）. similarly, subgroup analyses revealed no significant association in Asians or Chinese population although a significant association was apparent among Chinese population in studies with HWE under the heterozygote model （OR = 1.25, 95% CI = 1.03-1.52, F = 44.1%）. significant heterogeneity could be observed in some genetic models, but this heterogeneity was not significant when stratified by HWE. No evidence for publication bias was found. Conclusions： The Gs-FP1 lle105Val polymorphism might not be associated with male infertility risk, and thus additional well-designed studies with larger sample size are warranted.
