Molecular regulatory mechanism of ferroptosis and its role in gastrointestinal oncology:Progress and updates

gastrointestinal(GI)tumors,including liver,pancreatic,gastric,and colorectal cancers,have a high incidence rate and low survival rate due to the lack of effective therapeutic methods and frequent *** and postoperative chemoradiotherapy have largely reduced the fatality rates for most GI tumors,but these therapeutic approaches result in poor prognoses due to severe adverse reactions and the development of drug *** studies have shown that ferroptosis plays an important role in the onset and progression of GI *** is a new non-apoptotic form of cell death,which is iron-dependent,non-apoptotic cell death characterized by the accumulation of lipid reactive oxygen species(ROS).The activation of ferroptosis can lead to tumor cell ***,regulating ferroptosis in tumor cells may become a new therapeutic approach for tumors,making it become a research *** studies suggest that ferroptosis is mainly triggered by the accumulation of lipid ***,several studies have indicated that ferroptosis may be a new approach for the treatment of GI ***,we review current research progress on the mechanism of ferroptosis,current inducers and inhibitors of ferroptosis,and the role of ferroptosis in GI tumors to propose new methods for the treatment of such tumors.

Primary intestinal non-Hodgkin's lymphoma:A clinicopathologic analysis of 81 patients

AIM:To analyze the clinicopathologic features and the prognosis of primary intestinal ***:Patients were included in the study based on standard diagnostic criteria for primary gastrointesti-nal lymphoma,and were treated at Sun Yat-sen Univer-sity Cancer Centre between 1993 and ***:The study comprised 81 *** most common site was the ileocaecal ***-two point two percent patients had low-grade B-cell ***-one point nine percent patients had high-grade B-cell lymphoma and 25.9% patients had T-cell *** patients had localized *** were more patients and more early stage diseases in the latter period,and the origin sites *** ma-jority of patients received the combined treatment,andabout 20% patients only received nonsurgical *** wverall survival and event-free survival rates after 5 years were 71.6% and 60.9% *** mul-tivariate analysis revealed that small intestine and ileo-caecal region localization,B-cell phenotype,and normal lactate dehydrogenase were independent prognostic factors for better patient *** based treat-ment did not improve the survival ***:Refined stratification of the patients according to the prognostic variables may allow indi-vidualized *** treatment may be an optimal therapeutic modality for selected patients.

MUC1 and colorectal cancer pathophysiology considerations

Several lines of evidence point towards a biological role of mucin and particularly MUC1 in colorectal cancer. A positive correlation was described between mucin secretion, proliferation, invasiveness, metastasis and bad prognosis. But, the role of MUC1 in cancer progression is still controversial and somewhat confusing. While Mukherjee and colleagues developed MUC1-specific immune therapy in a CRC model, Lillehoj and coinvestigators showed recently that MUC1 inhibits cell proliferation by a β-catenin-dependent mechanism. In carcinoma cells the polarization of MUC1 is lost and the protein is over expressed at high levels over the entire cell surface. A competitive interaction between MUC1 and E-cadherin, through β-catenin binding, disrupts E-cadherin-mediated cell-cell interactions at sites of MUC1 expression. In addition, the complex of MUC1-β-catenin enters the nucleus and activates T-cell factor/leukocyte enhancing factor 1 transcription factors and activates gene expression. This mechanism may be similar to that just described for DCC and UNC5H, which induced apoptosis when not engaged with their ligand netrin, but mediate signals for proliferation, differentiation or migration when ligand bound.
