Effect of simulated gastrointestinal digestion on antioxidant,and anti-inflammatory activities of bioactive peptides generated in sausages fermented with Staphylococcus simulans QB7

Dry-fermented sausages are a good source of bioactive peptides,whose stability against gastrointestinal(GI)digestion determines their *** study focused on evaluating the effect of peptide extracts from sausages fermented with Staphylococcus simulans QB7 during in vitro simulated GI digestion,including peptide profiles and antioxidant and anti-inflammatory *** present in sausages were degraded during digestion,with molecular weight reduced from>12 kDa todigestion,however,the changes in antioxidant activities were the opposite.A total number of 255,252 and 386 peptide sequences were identified in undigested,peptic-digested and GI-digested samples,***,BIOPEP-UWM and admetSAR were used to further predict the functional properties and intestinal absorption of the identified peptide sequences from GI ***,18 peptides were discovered to possess either antioxidant or anti-inflammatory capacities.

Characterization of SARS-COV-2 main protease inhibitory peptides from Ulva prolifera proteins

The main protease(M^(pro))is essential for the replication of SARS-COV-2 and therefore represents a promising anti-viral *** this study,we screened M^(pro)inhibitory peptides from Ulva prolifera protein on in-silico *** analysis using the online toxic prediction tool ToxinPred revealed that all the peptides were *** hexapeptide(SSGFID)exhibited high M^(pro)inhibitory activity in molecular docking and its IC_(50)value was 139.40±0.82μmol/L in vitro according to fluorescence resonance energy transfer assay(FRET).Quantitative real-time(qRT-)PCR results show that SSGFID could stimulate the expression of mitosis-related factors,including nuclear factor-κB,cyclin D1,and cyclin-dependent kinase 4,to promote the proliferation of mice *** study revealed that SSGFID showed resistance against pepsin and trypsin but lost D(Asp)after pretreatment at121℃ for 15 ***,SSGFID was mainly transported through the Caco-2 cell monolayer by the peptide transporter PepT1 and passive-mediated transport during the transport ***,the peptide was also degraded by Caco-2 intracellular enzymes,and the transfer rate of intact peptide was4.2%.Furthermore,Lineweaver–Burk plots demonstrated that SSGFID possessed a mixed inhibitory characteristic with M^(pro).Our study indicated the potential of Ulva prolifera as antiviral and immuneenhancing functional food ingredients and nutraceuticals.

This study aimed to in vitro evaluate the impact of common gastrointestinal(GI)alterations appearing with aging on protein digestibility,and functional related properties,in four different meats(chicken,turkey,pork and beef).Thus,three elderly digestion models were stated as long as altered GI conditions affected at oral(E1),oral and gastric(E2)and oral,gastric and intestinal stages(E3).Healthy adult GI conditions were also mimicked as standard control model(C).A notable trichloroacetic acid(TCA)soluble protein and the free amino acids(FAA)release reduction(p<0.05)were found under intestinal suboptimal conditions(E3),being more accused in beef than in other ***,chicken intake would be more advisable,than other meats,in terms of protein digestibility;while beef would provide a greater net supply of FAA under E3 model *** altered conditions,seem to favor protein ***,while gastric and intestinal suboptimal conditions diminish the angiotensin converting enzyme(ACE)inhibition of meat digesta,their antioxidant activity was only negatively affected by intestinal altered conditions.

Production of short chain fatty acids and vitamin B_(12) during the in-vitro digestion and fermentation of probiotic chocolate

The prebiotic functions of chocolate to improve the function and health benefit of the chocolate when enriched with probiotics has not been fully *** study investigated the role of chocolates enriched with probiotics in the production of SCFAs and vitamin B_(12) during the in-vitro colonic *** probiotic bacteria were encapsulated using a mixture of cocoa powder(10%),FOS(2%)and Na-alginate(1%)before added to *** results revealed that encapsulated Lactobacillus plantarum and Bifidobacterium animalis *** BB12 produced significantly higher amounts(1876.5±105.16 and 1348.51±77.37 mmol,respectively)of acetic acid after 48h of colonic *** with encapsulated BB12,La5,Lactobacillus sanfranciscensis and Streptococcus thermophilus yielded higher amounts of propionic,isobutyric,butyric and isovaleric ***-chocolates with 70%cocoa produced significantly more acetic,propionic,and butyric acids than that of 45%***,Probiotic-chocolates with 45%cocoa produced greater amounts of vitamin B_(12) than 70%*** data demonstrate that probiotic-chocolate could be a functional snack with additional health benefits.
