gene expression Patterns in Different Wool Densities of Rex Rabbit Using cDNA Microarray

This study was carried out to identify gene expression patterns in different wool densities of rex rabbit. The mid-dorsal skin samples from 8 rex rabbits were studied. They were divided into two groups according to different wool densities. Total RNA was isolated and labeled by reverse transcription reaction with Cy5-dCTP and Cy3-dCTP for cDNA probe. The cDNA probe was hybridized with cDNA microarrays containing 14 601 rabbit’s genes. The differently expressed genes were analysed with the gene Ontology （GO） classification and the pathway analysis. Hierarchical clustering was performed to clarify genes in association with different wool densities. The 2 657 differentially expressed genes were identified. Among them, 1 103 genes were functionally known genes, 687 genes were up-regulated and 419 were down-regulated. GO analysis indicated that these altered genes were associated with metabolism, signal transduction, cell cycle, cell adhesion, cell proliferation, cell division, apoptosis, and other processes. KEGG analysis showed that 95 signal pathways associated with up-regulated genes and 87 signal pathways associated with down-regulated genes had changed significantly （P0.05）. Some important differentially expressed genes in different wool densities of rex rabbit were identified, such as MMP2, TGF-β1, TGF-β2, IGF-1, ITGB1, RPS6KB1, BMP2, ActRIIB, CDK2, and CCNA2. Hierarchical clustering analysis separated the differentially expressed genes into 5 main characteristic groups. Data from cDNA microarray experiments clearly distinguished between group A and group B. Some important genes were identified, which might be useful in further study on wool density markers of rex rabbit.

Colorectal cancer(CRC)is one of the most common malignancies *** of predictivemolecular markers may help to achieve the best outcome in *** purpose of this study is to identify thedifferentially expressed genes and new potential predictive and prognostic molecular biomarkers for *** study,CRC and matched normal tissues acquired from the same patient were used to extract total *** NanoString nCounter assay was applied to determine the differentially expressed *** results werethen validated by using the Cancer Genome Atlas ***,we identified 27 genes that revealed significantcorrelation with CRC in the tumor *** genes in the pan-cancer panel showed significant differentialexpression,which were more universal than others in the CRC *** some of them have not been reportedas being directly related to CRC yet,future mechanism studies can be designed based on this *** studydemonstrated NanoString nCounter assay could serve as an alternative approach for gene expression analysis andidentified several unreported differently expressed genes in CRC patients,which may provide some importantclues for more in-depth study of CRC and serve as potential predictive molecular biomarkers for clinical diagnosisapplication.

Background： Vascular endothelial growth factor A （VEGFA） can induce endothelial cell proliferation, promote cell migration, and inhibit apoptosis. These processes play key roles in physiological blood vessel formation and pathological angiogenesis. Methods： In this study, we examined VEGFA gene expression in the heart, liver, and kidney of Tibetan pigs （-I-P）, Yorkshire pigs that migrated to high altitudes （YH）, and Yorkshire pigs that lived at low altitudes （YL）. We used PCR and Sanger sequencing to screen for single nucleotide polymorphisms （SNPs） in 5＇-flanking DNA and exons of the VEGFA gene. Quantitative real-time PCR and western blots were used to measure expression levels and PCR products were sequenced. Results： Results showed that the VEGFA mRNA and protein expression in heart, liver and kidney of TP was higher than that in YH and YL. In addition, the mRNA sequence of the pig VEGFA gene was conserved among pig breeds, and only five SNPs were found in the 5＇-flanking region of the VEGFA gene, the allele frequency distributions of the 5 SNPs were not significantly different between the TP, Yorkshire （YL）, and Diannan small-ear （DN） pig populations. Conclusion： In conclusion, the Tibetan pig showed high tissues, which suggests that the VEGFA gene may play a levels of VEGFA gene expression in several hypoxic major functional role in hypoxic adaptation.

AIM： To compare the effects of thiamine pyrophosphate（TPP） and thiamine（TM） in oxidative optic neuropathy in rats induced by ***： The animals were divided into four groups：a control group（CG）,an ethambutol control（ETC） group,TM plus ethambutol group（TMG）,and TPP plus ethambutol group（TPPG）.One hour after intraperitoneal administration of TM 20 mg/kg to the TMG group and TPP 20 mg/kg to TPPG group,30 mg/kg ethambutol was given via gavage to all the groups but the *** procedure was repeated once daily for 90 *** that period,all rats were exposed to high levels of anaesthesia in order to investigate the gene expression of malondialdehyde and glutathione in removed optic nerve tissue and histopathologically to examine these tissues. RESULTS： Malondialdehyde gene expression significantly increased,whereas glutathione gene expression significantly decreased in the ETC group compared to the *** could not prevent the increase of malondialdehyde gene expression and the decrease of glutathione,while TPP significantly could ***,significant vacuolization in the opticnerve,single-cell necrosis in the glial cells,and a decrease in oligodendrocytes were observed in the ETC *** in the optic nerve,a decrease in oligodendrocytes and single-cell necrosis were found in the TMG group,while no pathological finding was observed in the TPPG group except for mild ***： TPP protects the optic nerve against the ethambutol-induced toxicity but TM does *** can be beneficial in prophilaxis of optic neuropathy in ethambutol therapy.

This study aimed to determine the effects of black tea polyphenols on gene expression in hepatocellular cancer *** total RNA from HepG_(2) hepatocellular cancer cells treated with black tea polyphenols was subjected to Human 14K cDNA microarray ***-time PCR and Western blot analysis were conducted to verify microarray *** tea polyphenols treatment at the dose of 20 mg/L,40 mg/L or 80 mg/L for one to three days inhibited the growth of HepG_(2) cells in a dose and time dependent manner.A total of 48 genes showed more than two-fold change after black tea polyphenols treatment,including 17 upregulated genes and 31 downregulated genes,and they were involved in the regulation of cell growth,cell cycle,apoptosis,signaling,angiogenesis,tumor invasion and ***-time PCR analysis of the selected genes showed that their mRNA expression changes were consistent with the microarray *** addition,Western blot analysis of the selected genes showed that their protein expression changes were consistent with mRNA *** conclusion,gene expression profiles provide comprehensive molecular mechanisms by which black tea polyphenols exerts growth inhibition effects on cancer *** novel molecular targets identified in this study may be further exploited as therapeutic strategies for hepatocellular cancer.

Objective: To analyze the changes of gene expression in phenylbutyrate induced differentiation of glioma cells. Methods: The expression levels of 14000 genes in glioma cells before and after inducement with sodium phenyl- butyrate for 2 h or 6 days were evaluated by cDNA array technique and proved by multi-dot blotting. Results: expression of 98 genes in glioma cells showed changes after the inducement. Some genes involved in transcription and translation and some oncogenes are down-regulated, while some gene involved in differentiation or apoptosis are up-regulated. 18 unknown expression sequencing tag (EST) changed too. Conclusion: A gene expression profile associated with differentiation of glioma cells was established.

Objective: To explore the potentiality of retroviral etiology in human acute myeloid leukemia(AML). Methods: The expression of clone 6#11 in leukemic cell samples from 19 AML cases and peripheral blood mononuclear cells (PBMNCs) from 20 controls was studied by means of Northern blot and reversal transcription polymerase chain reaction (RT-PCR). Results: Northern blot and RT-PCR analyses showed that the expression level of clone 6#11 was significantly higher in AML patients than that in control. Conclusion: Northern blot and RT-PCR analyses revealed that the expression of novel retrovirus were associated with acute myeloid leukemia.

Objective: Transforming Growth Factor-β1 (TGF-β1)plays a central role in the process of . growth suppressionof the hepatocytes, and its type II receptor (TGF-β1R II)transfers the signal of growth suppression. In this study,the gene expression of TGF-β1R II in HCC and itsclinical significance was investigated. Methods: Theexpression of TGF-β1R II mRNA in 30 cases Of HCCtissue and the surrounding liver tissue was separatelydetected using reverse transcription-PCR. Results:The positive expression rate of TGF-β1R II mRNA wassignificantly lower in HCC tissue (11/30) than that in thesurrounding liver tissue (23/30) (Pexpression of TGF-β1 R II mRNA by tumoral hepatocyteresults in the defect of its negative growth regulation,and this may be one of the most important reasons forits carcinogenesis and uncontrolled growth.

Multiplicative Lévy noise-induced transitions in gene expression

gene expression is intrinsically noisy. Experimental studies have shown that random fluctuations are large bursts and heavytailed distributions. Therefore, this study aims to consider transition dynamics in a gene transcriptional regulatory system via the mean first exit time(MFET) and first escape probability(FEP), when the degradation rate is under multiplicative non-Gaussian Lévy fluctuations in the sense of It? and Marcus forms. We find that, in the Marcus form case, the FEP corresponding to different stability index and noise intensity has an intersection point, whereas in the It? form case, the turning point only occurs at stability index. Increasing the initial CI concentration is helpful for improving the likelihood of transcription in both cases. Our results also imply that larger jumps of Lévy noise and smaller noise intensity can shorten the time of state transition to boost protein production.
