Peroxidase from foxtail millet bran exerts anticolorectal cancer activity via targeting cellsurface GRP78 to inactivate STAT3 pathway

Molecular targeted therapy has become an emerging promising strategy in cancer treatment,and screening the agents targeting at cancer cell specific targets is very desirable for cancer *** previous study firstly found that a secretory peroxidase of class III derived from foxtail millet bran(FMBP)exhibited excellent targeting anti-colorectal cancer(CRC)activity in vivo and in vitro,whereas its underlying target remains *** highlight of present study focuses on the finding that cell surface glucose-regulated protein 78(cs GRP78)abnormally located on CRC is positively correlated with the anti-CRC effects of FMBP,indicating it serves as a potential target of FMBP against ***,we demonstrated that the combination of FMBP with the nucleotide binding domain(NBD)of cs GRP78interfered with the downstream activation of signal transducer and activator of transcription 3(STAT3)in CRC cells,thus promoting the intracellular accumulation of reactive oxygen species(ROS)and cell grown *** phenomena were further confirmed in nude mice tumor ***,our study highlights cs GRP78 acts as an underlying target of FMBP against CRC,uncovering the clinical potential of FMBP as a targeted agent for CRC in the future.

Inflammatory bowel disease(IBD)is a chronic relapsing-remitting systemic disease of the gastrointestinal tract,characterized by an inflammatory *** mycobiota community dysbiosis has been reported that is closely related to the development of *** previous findings indicated that polyphenol of the inner shell(BPIS)from foxtail millet bran could restore the gut microbiome and inhibit the progress of colorectal cancer(CRC).In the present study,we studied the anti-inflammatory potential of BPIS in the dextran sodium sulfate(DSS)-induced mouse colitis *** suggested that BPIS alleviated experimental colitis by restoring body weight,colonic length and protecting the epithelial architecture from damage by ***,we found that BPIS strengthened the gut barrier function and inhibited the activation of Wnt1/β-catenin *** sequence analysis indicated that BPIS remodeled the overall structure of the gut mycobiota from colitis mice toward that of the normal counterparts,including 1 phylum and 9 ***,BPIS significantly increased the abundance of Aspergillus *** further verified that BPIS significantly promoted the growth of *** in ***,BPIS has great potential to develop into an effective against IBD drug.

Clinically,the postoperative infections are a pressing public health ***,a major component of gram-negative bacterial cell wall plays a crucial role to provoke the immune response,which lead to chronic inflammation and cancer recurrence particularly colorectal cancer(CRC).In general,inflammation is a complex physiological response of body tissues to disease causing irritants,which aims to remove the harmful stimuli and promotes wound ***,long-term inflammatory reactions are considered as the new malignant phenotype of cancer cells and diffusely approved that chronic inflammation plays a pivotal role in the early stage of ***,controlling LPS-induced inflammatory responses might reduce CRC risk and improve the clinical *** millet(Setaria italica)contains an abundance of vitamins,minerals and proteins containing massive essential amino acids *** millet bran,the by-product of foxtail millet processing,is an excellent source of phytochemicals,among which polyphenols exhibit a wide range of pharmacological and medicinal ***,polyphenols may occur in both free and bound forms,however most of them are observed in bound *** lots of studies showed that bound polyphenols had multiple biological activities both in vivo and in vitro,such as anti-oxidant,anti-tumor,anti-inflammatory and immunomodulation et *** results also showed that BPISexhibited the anti-inflammatory effect in LPS-induced HT-29 cell through reducing the levels of pro-inflammatory cytokines(IL-lβ,IL-8 and IL-6)and promoting the expression of anti-inflammatory cytokine(IL-10)in vitro and in ***,our data showed that the active constituents of BPIS primarilycontained ferulic acid,p-coumaric ***,there is a growing evidence showed that bound polyphenols could also act as pro-oxidant chemical messenger in tumor *** previous studies also proved thatbound polyphenols of inner she
