Scleral TGF-β1 and Smad3 expression is altered by TCM Bu Jing Yi Shi Tablets in guinea pigs with form-deprivation myopia

Objective:To explore whether the traditional Chinese medicine(TCM)Bu Jing Yi Shi Tablets alters the expression of scleral TGF-b1 and Smad3 in guinea pigs with formdeprivation myopia(FDM).Methods:Sixty-five guinea pigs were randomly divided into control,model,low-,medium-,and high-dose treatment *** for the control group,FDM was induced by covering the right eye of each animal with opaque *** treatment groups were gavaged with different suspension concentrations of Bu Jing Yi Shi *** and axial length were performed before and after myopia *** the end of the experiment,all right eyes were extracted,and scleral sections were prepared for staining and TGF-b1 and Smad3 *** thickness and area,the scleral fibroblast quantity,and scleral TGFb1 and Smad3 expressions were ***:The 5 FDM groups had the same initial axial length and diopter,the final diopter and axial length of the model group were significantly increased compared with the control group and treatment groups(P<.01).The axial length of each treatment group decreased as the dose decreased compared with the model group(P<.01);the total scleral area(P<.05 e.01)and the number of scleral fibroblasts(P<.01)in the model group were significantly lower than the treatment *** the TGF-b1 and Smad3 integral optical densities in the model group were significantly lower than the control and medium-and high-dose treatment groups(P<.01).TGF-b1 and Smad3 mRNAs in the model group were decreased compared with the control group,but increased in expression after treatment.

There is now mounting evidence implying the involvement of retinal dopamine（DA）in the signaling cascade that leads to myopic eye ***,in one of our recent work,retinal DA levels are found unaltered in myopic eyes of C57BL/6 mice,which is in sharp disagreement with the results obtained in most other species,and thus suggesting a lack of association between retinal DA and myopia development in this mouse ***,in an attempt to further characterize the role of DA in mouse eye growth,we investigated whether destroying retinal dopaminergic system,by intravitreal application of6-hydroxy dopamine（6-OHDA）,would influence refractive *** of 6-OHDA significantly reduced the retinal DA levels,assessed by HPLC analysis,and the effect lasted for at least31 *** assessment using an automated eccentric infrared photorefractor showed that the6-OHDA-injected eyes became markedly myopic relative to their fellow ***,in injected eyes,form-deprivation failed to induce further myopic shifts,nor did it cause further reduction in retinal DA *** results indicate that,in C57BL/6 mice,normal refractive development requires an intact retinal dopaminergic system,including healthy dopaminergic amacrine cells and complete retinal DA *** data further suggest that retinal dopaminergic system is indispensible for the development of deprivation myopia in C57BL/6 mice.

Objective Unlike observed in most other species, retinal dopamine（DA） levels are unaltered in C57 BL/6 mice with form-deprivation myopia（FDM）. Although implying a novel, DA-independent mechanism for FDM, this discrepancy might be attributed to the fact that the C57 BL/6 strain is deficient in melatonin, a hormone that interplays with DA. Here we explored whether and how form deprivation changed refractive development and retinal DA levels in a mouse strain proficient in melatonin. Methods CBA/Ca J mice were used in this study. Normal refractive development was measured every 5-10 days from P21 to P106. FDM was induced by wearing monocular occluder, starting at P21, for 5 weeks. Refractions were measured with an eccentric infrared photorefractor. Ocular biometry was performed with A-scan ultrasonography, optical coherence tomography and photokeratometry. Retinal DA, DOPAC and vitreal DOPAC（an indicator of DA release） levels were assessed by HPLC analysis. Retinal tyrosine hydroxylase（TH） levels were examined by quantitative Western blotting. Results Under normal visual conditions, the refraction at P21 was 4.544 ± 0.147 D. It increased continuously in hyperopic direction, reaching a peak（18.262 ± 0.378 D） at P86, and then leveled off. FDM was successfully induced with the refractive status of deprived eyes（8.489 ± 0.877 D） being significantly different（P = 0.00000003） from fellow eyes（13.048 ± 0.870 D）. As expected, axial length in deprived eyes（2.698 ± 0.007 mm） is significantly longer（P = 0.0011） than that in fellow eyes（2.675 ± 0.008 mm）;but unexpectedly, both values were significantly smaller（P = 0.0012, 0.000001） than that of normal eyes（2.736 ± 0.006 mm） in intact animals. Consistent with previous results in C57 BL/6 mice, either retinal DA（P = 0.234）, DOPAC（P = 0.643） or TH（P = 0.653） levels were similar between deprived and fellow eyes. But surprisingly, both deprived and fellow eyes exhibited significantly lower retinal DA（P = 0.006, 0.0004）, DOPAC（P = 0.000

Vasoactive intestinal peptide,a promising agent for myopia?

AIM： To investigate the role of vasoactive intestinal peptide（VIP） in form-deprivation myopia（FDM）.METHODS： FDM was created in three groups of eight chicks by placing a translucent diffuser on their right *** injections of saline and VIP were applied once a day into the occluded eyes of groups 2 and 3,*** and axial length（AL） measurements were performed on the first and 8^th days of diffuser *** retina mR NA levels of the VIP receptors and the ZENK protein in right eyes of the three groups and left eyes of the first group on day 8 were determined using real time polymerase chain reaction（PCR）.RESULTS： The median final refraction（D） in right eyes were-13.75（-16.00,-12.00）,-11.50（-12.50,-7.50）,and-1.50（-4.75,-0.75） in groups 1,2,and 3,respectively（P〈0.001）.The median AL（mm） in right eyes were 10.65（10.00,11.10）,9.90（9.70,10.00）,and 9.20（9.15,9.25） in groups 1,2,and 3,respectively（P〈0.001）.The median delta-delta cycle threshold（CT） values for the VIP2 receptors were 1.07（0.82,1.43）,1.22（0.98,1.65）,0.29（0.22,0.45） in right eyes of groups 1,2,and 3,and 1.18（0.90,1.37） in left eyes of group 1,respectively（P=0.001）.The median delta-delta CT values for the ZENK protein were 1.07（0.63,5.03）,3.55（2.20,5.55）,undetectable in right eyes of groups 1,2,and 3 and 1.89（0.21,4.73） in left eyes of group 1,respectively（P=0.001）.CONCLUSION： VIP has potential inhibitory effects in the development of FDM.

Expression and role of specificity protein 1 in the sclera remodeling of experimental myopia in guinea pigs

Mechanism of the DL-alpha-aminoadipic acid inhibitory effect on form-deprived myopia in guinea pig

AIM:To investigate the effect of intravitreal injection of DL-alpha-aminoadipic acid (DL-α-AAA) on ocular refractive state and retinal dopamine, transforming growth factor-β2 (TGFβ2 ), vasoactive intestinal polypeptide (VIP) in guinea pig form-deprived myopia. METHODS:Four-week-old pigmented guinea pigs were randomly assigned to 4 groups:normal control, deprivation, deprivation plus DL-α-AAA, deprivation plus saline. form deprivation was induced with the self-made translucent eye shields, and lasted for 14 days. 8μg DL-α-AAA was injected into the vitreous chamber of deprived eyes. The corneal radius of curvature, refraction and axial length were measured. Retinal dopamine content was evaluated by the high-performance liquid chromatography with electrochemical detection, and TGFβ2 and VIP protein were detected by Western blotting. RESULTS:Fourteen days of eye occlusion caused the axial length to elongate and become myopic in the form-deprived eyes, with the decrease of retinal dopamine and the increase of TGFβ 2 and vasoactive intestinal polypeptide (VIP) protein. Intravitreal injection of DL-α-AAA could inhibit the myopic shift from (-3.65±1.06)D to (-1.48 ±0.63)D, P form deprivation myopia, which may be associated with retinalTGFβ2 protein in guinea pigs.
