Histone H1 and its C-terminal lysine rich fragments were recently found to be potent inhibitorsof furin,a mammalian proprotein ***,its role in the regulation of furin-dependent proproteinprocessing remains *** we report that histone H1 efficiently blocks furin-dependent pro-yonWillebrand factor（pro-vWF）processing in a dose-dependent *** and immunof-luorescence studies confirmed that histone H1 could interact with furin,and the interaction mainly took placeon the cell *** noted that histone H1 was released from cells undergoing necrosis and apoptosisinduced by *** findings suggested that histone H1 might be involved in extracellular and/or intracellu-lar furin regulation.

AIM: To model the three-dimensional structure and investigatethe interaction mechanism ofthe proprotein convertase furin/kexin and their inhibitors (eglin c mutants). METHODS: The three-dimensional complex structures of furin/ kexin with its inhibitors, eglin c mutants, were generated by modeller program using the newly published X-ray crystallographical structures of mouse furin and yeast kexin as templates. The electrostatic interaction energy of each complex was calculated and the results were compared with the experimentally determined inhibition con- stants to find the correlation between them. RESULTS: High quality models of furin/kexin-eglin c mutants were obtained and used for calculation of the electrostatic interaction energies between the proteases and their inhibitors. The calculated electrostaticenergies of interaction showeda linear correlation to the experimental inhibition constants. CONCLUSION: The modeled structures give good explanations of the specificity of eglin c mutants to furin/ kexin. The electrostatic interactions play important roles in inhibitory activity of eglin c mutants to furin/kexin. The results presented here provided quantitative structural and functional information concerning the role of the charge- charge interactions in the binding of furin/kexin and their inhibitors.

furin is an important mammalian proprotein convertase that catalyzes the proteolytic maturation of a variety of prohormones and proproteins in the secretory pathway. In the brain, the substrates of furin include the proproteins of growth factors, receptors and enzymes. Emerging evidence, such as reduced furin mRNA expression in the brains of Alzheimer’s disease patients or schizophrenia patients, has implicated a crucial role of furin in the pathophysiology of neurodegenerative and neuropsychiatric diseases. Currently, compared to cancer and infectious diseases, the aberrant expression of furin and its pharmaceutical potentials in neurological diseases remain poorly understood. In this article, we provide an overview on the physiological roles of furin and its substrates in the brain, summarize the deregulation of furin expression and its effects in neurodegenerative and neuropsychiatric disorders, and discuss the implications and current approaches that target furin for therapeutic interventions. This review may expedite future studies to clarify the molecular mechanisms of furin deregulation and involvement in the pathogenesis of neurodegenerative and neuropsychiatric diseases, and to develop new diagnosis and treatment strategies for these diseases.

The current pandemic of COVID-19 caused by a novel coronavirus,severe acute respiratory syndrome coronavirus-2(SARS-CoV-2),threatens human health around the *** particular concern is that bats are recognized as one of the most potential natural hosts of SARS-CoV-2;however,coronavirus ecology in bats is still ***,we performed a degenerate primer screening and next-generation sequencing analysis of 112 bats,collected from Hainan Province,*** coronaviruses,namely bat betacoronavirus(Bat CoV)CD35,Bat CoV CD36 and bat alpha-coronavirus CD30 were *** CoV CD35 genome had 99.5%identity with Bat CoV CD36,both sharing the highest nucleotide identity with Bat Hp-betacoronavirus Zhejiang2013(71.4%),followed by SARS-CoV-2(54.0%).Phylogenetic analysis indicated that Bat CoV CD35 formed a distinct clade,and together with Bat Hp-betacoronavirus Zhejiang2013,was basal to the lineage of SARS-CoV-1 and ***,Bat CoV CD35 harbored a canonical furin-like S1/S2 cleavage site that resembles the corresponding sites of *** furin cleavage sites between CD35 and CD36 are *** addition,the receptor-binding domain of Bat CoV CD35 showed a highly similar structure to that of SARS-CoV-1 and SARS-CoV-2,especially in one binding *** conclusion,this study deepens our understanding of the diversity of coronaviruses and provides clues about the natural origin of the furin cleavage site of SARS-CoV-2.

Breast cancer bone metastasis poses significant challenge for therapeutic *** the metastatic environment,osteoclasts and tumor cells interact synergistically to promote cancer *** this study,the proprotein convertase furin is targeted due to its critical roles in both tumor cell invasion and osteoclast ***,the furin inhibitor is specifically delivered by bone targeting superparamagnetic iron oxide(SPIO)*** in vitro and in vivo data demonstrate that this system can effectively inhibit both osteoclastic bone resorption and breast cancer invastion,leading to alleviated ***,the bone targeting&furin inhibition nanoparticle system is a promising therapeutic and diagnostic strategy for breast cancer bone metastasis.

目的研究α1-PDX对宫颈癌细胞株He La细胞生长、转移及成瘤的影响并探讨其机制。方法α1-PDX转染He La细胞,通过MTT细胞增殖实验,Boyden细胞迁移和侵袭实验观察He La细胞的生长,迁移和侵袭能力。采用蛋白印迹法检测细胞furin及产物膜性Ⅰ型金属蛋白酶(MT1-MMP)蛋白量的变化,荧光酶底物实验检测furin活性。制作He La细胞致瘤裸鼠模型研究肿瘤体内生长。结果细胞增殖实验结果显示,与对照组相比24 hα1-PDX组He La细胞体外生长抑制18.4%,差异有统计学意义(Pfurin的活性和MTI-MMP的蛋白水平。He La/PDX致瘤裸鼠的成瘤机率及皮下肿瘤体积明显小于对照组。结论α1-PDX可有效抑制人宫颈癌He La细胞的生长、转移及成瘤能力,机制可能与降低furin活性及产物MTI-MMP的表达有关。

Protease controlled assembly of nanostructures has remained challenging for supramolecular ***,we report a condensation system which could be controlled by furin,an important protease which is associated with early development of *** designed Acetyl-Arg-Val-Arg-Arg-Cys（StBu）-Tyr（I-125）-CBT（1） for self-assembling radioactive nanoparticles（I 125-NPs） in living *** scrambled control Acetyl-Arg-Tyr（I-125）-Arg-Cys（StBu）-Arg-Val-CBT（1-scr） which can not be cleaved by furin was also prepared for study in *** furin cleavage at 100 μM,the cold form of compound 1（i.e.,1-cold） can condense to form macrocyclized dimer which self-assembles to form I-NPs with a mean diameter of 150 *** uptake indicated that more of 1 than 1-scr was uptaken by breast cancer cell ***,cells incubated with 1,1-scr,or 1-scr with 100 μM 1-scr-cold showed similar efflux curves while that of cells incubated with 1 and 100 μM of 1-cold was *** 160 min after cell efflux,17.9% of the radioactivity was retained in the cells incubated with 1 and 100 μM 1-cold while the percentages of other three groups are 4.4%,3.7%,and 3.7% *** means of intracellular assembly of radioactive nanoparticles,this provides us with a new method of furin detection or cancer early detection.

Background: Reported are increased risks for malignant transformation in human oral keratinocytes (HOK) from ethyl alcohol (ETOH), tobacco products or human papilloma virus oncogenic subtype 16 (HPV 16) infections. We examined various HOK cell responses to these factors to show inhibitors of epidermal growth factor receptor (EGFR) also inhibits furin;proprotein convertase (FC) and HPV 16 entry in HOK. Methods: Immortalized HOK by HPV 16 (HPV 16B) or human telomerase (hTERT);primary foreskin keratinocytes (NHFK), primary HOK, buccal keratinocytes (NHBK) and oral SCC-25 were treated with dibenz[a,l]pyrene (DBP), anthraquinone;nitrosamine (NNAL) or ethyl alcohol (ETOH) and acetaldehyde (AA). ETOH was tested for synthesis of malondialdehyde (MDA) and alcohol dehydrogenase expression (ADH). ETOH, and PAH were evaluated by Western immunoblot for oncogene changes, and phosphorylated EGFR expression. Inhibition of EGFR by WZ4002 and Erlotinib and/or carcinogens effect on HPV 16 entry were studied. A green fluorescent pseudovirus (PsV);chloromethylketone (CMK) an inhibitor of furin activity and Western immunoblot of furin cell distribution further characterized HPV 16 entry. Results: ETOH (10 μM) increased expression of phosphorylated EGFR and HPV 16 entry through furin activity, and membrane, nuclear and cytoskeletal accumulations. CMK suppressed HPV 16 entry and blockage of ADH while aldehyde dehydrogenase (ALDH) enhanced HPV 16 entry. Similarly PAH, DBP (4-8 nM), anthraquinone (98 nM) and NNAL (6.9 μM) enhanced HPV 16 entry through furin activity and membrane, nuclear and cytoskeletal accumulations. Furthermore, WZ4002 and Erlotinib suppressed expressions of phosphorylated EGFR, FC activity, and HPV 16 entry. ETOH and DBP treatments also enhanced expressions of protease activated receptor-1 (PAR-1), and p21waf1 while depressed p16 and p27KIP1 expressions in HOK/HPV 16B cells. Conclusion: EGFR inhibitors are candidates for suppression of alcohol and tobacco effects on EGFR
