Objective: To study the protective effect of ascorbic acid and curcumin against the abnormal Estrous cycle and morphological changes in cells induced by repeated ultraviolet B(UVB)radiation in female Wistar rats.Methods: Sixteen female sexually mature Wistar rats weighing 130-150 g and aged 12-16 weeks were randomly divided into four groups.The control group received normal food and water ad libitum.The UVB group was exposed to a dose of 280 nm of UVB radiation for 2 h daily.The UVB+curcumin group received a dose of 280 nm of UVB radiation for 2 h daily and also an oral dose of curcumin(25 mg/kg body weight)daily.The UVB+ascorbic acid group received a dose of 280 nm of UVB radiation for 2 h daily and also an oral dose of ascorbic acid(250 mg/kg body weight)daily.All the treatments last for 15 consecutive days.Body and ovary weight and gonadosomatic index were measured.The stages(proestrus,estrus,metaestrus and diestrus)of the Estrous cycle were determined by the cell types observed in the vaginal smear.Results: UVB radiation caused irregular alterations on the Estrous cycle and morphological changes of the female Wistar rat as compared with the control group.Ascorbic acid and curcumin protected UVB-induced Estrous phases and their cells.But curcumin showed greater protection than ascorbic acid.Conclusions: Ascorbic acid and curcumin at low doses can alleviate abnormal Estrous cycle and morphological changes in cells induced by UVB radiations in female Wistar rats.

Objective:To evaluate the effect of Salacia lehmbachii (S. lehmbachii) ethanol root bark extract on Estrous cycle and sex hormones in female rats. Methods: Forty-eight virgin rats with regular 4-day cycle were grouped into four and each group was further subdivided into 'a' and 'b' (n=6). Each group was orally treated for 28 days with 2 mL of distilled water (control), ethanol root bark extract ofS. lehmbachii in doses of 250, 500 and 750 mg/kg/body weight (groups 2, 3, 4, respectively). Estrous cycle was determined daily using the vaginal smear method. Rats in'a' subgroups were weighed and sacrificed on the 29th day, and blood was collected for serum generation which was used for hormonal assay and sex organs were removed and weighed. Rats in 'b' subgroups were discontinued from treatment for 2 weeks and the parameters above were reassessed.Results:The mean length of Estrous cycle and duration of diEstrous of treated rats were prolonged dose dependently compared to control. The increase was significant (PEstrous phases were shortened in the same pattern. Relative weights of sex organs were reduced significantly (PEstrous and diEstrous cycle, and reduces serum sex hormones levels. The observed alterations were reversible.

Endoscopic monitoring the Estrous phase of Arabian mares with assaying its biopsy

Objective:To evaluate the changes occur in the vagina, cervix and endometrium during the pre- ovulatory period in Arabian mares by using endoscope as a diagnostic tool as well as the availability of taking an endoscopic biopsy during this stage.Methods:Ten cyclic and healthy arabian mares were subjected to ultrasound scanning of their ovaries to estimate the diameter of the preovulatory follicles, collection of blood samples for estimating the hormonal levels (Estradiol-17β and progestrone) for confirming the Estrous phase, endoscopic monitoring their vagina, cervix and endometrium, the availability of taking biopsical samples and histological examination of the obtained samples.Results:The obtained data revealed that, the mean diameter of the preovulatory follicles was (3.95±0.34) cm, the plasma estradiol and progestrone levels were (85.98±5.26) pg/mL and (0.73±0.23) ng/mL, respectively, endoscopic monitoring the vagina, cervix and endometrium revealed hyperemic vaginal mucous membrane, soft and opened cervical ostium with abundant Estrous mucous and folding, hyperemic and edematous appearance of the endometrium, respectively and the histological features of the biopsical endometrial samples revealed that, The lamina propria-sub mucosa was packed with simple or branched tubular glands lined with simple columnar epithelium showing oval or rounded dark nuclei with different degrees of secretory activities of the glands.Conclusions: The endoscope proved to be a reliable tool for confirming the Estrous phase in mares and monitoring the gross features of the vagina, the cervix and the endometrium, in addition to the availability of taking biopsical smears.

Prefrontal and Accumbal Electric Activity during Auditory Stimulation in Virgin Female Rats: Changes Related to the Estrous Cycle

The Estrous cycle is a physiological process modulated by hormonal changes associated, in turn, with alterations of the attraction or sensitivity with which females perceive different sensory stimuli. Auditory stimuli play an important role in the social interaction of several mammals, including their sexual behavior and the mother-young relationship. Since the prefrontal cortex (PFC) and nucleus accumbens (Acc) had been associated with the processing and assignation of the incentive value of stimuli, this study was designed to analyze whether electroencephalographic (EEG) activity in the PFC and Acc was modified during the two antagonist phases of the Estrous cycle (proestrus-estrus and diestrus) when female rats heard sound recordings with different types of cries emitted by other rats. EEGs were recorded in eight female virgin Wistar rats, 80-100 days old, bilaterally implanted in the PFC and Acc during an awake-quiet state, in three conditions: when hearing 1) cries of pups;2) cries of adult male rats;and, 3) cries of adult female rats. The cries were recorded from pups (2-10 days old) and male and female adult rats after gentle prodding by a human hand. Each auditory stimulus (which included vocalizations up to 22 kHz) was randomly played back through a speaker placed near the female rats. Only upon perceiving the cries of male rats did the females in the proestrus-Estrous phase show a lower relative power in the 14-30 Hz band in the Acc and an increased inter-prefrontal correlation in the 8-13 Hz band. It is probable that these EEG changes are associated with the lower activation or non-anxious state that the female rat presents specifically during the proestrus-Estrous phase in response to the alarm cries of adult male rats.
