Inhalation of Cananga odorata Essential oil relieves anxiety behaviors in autism-like rats via regulation of serotonin and dopamine metabolism

Objective:Anxiety is one of the most common symptoms associated with autistic spectrum disorder.The Essential oil of Cananga odorata(Lam.)Hook.f.&Thomson,usually known as ylang-ylang oil(YYO),is often used in aromatherapy as a mood-regulating agent,sedative,or hypotensive agent.In the present study,the effects and mechanisms of YYO in alleviating anxiety,social and cognitive behaviors in autism-like rats were investigated.Methods:The prenatal valproic acid(VPA)model was used to induce autism-like behaviors in offspring rats.The effectiveness of prenatal sodium valproate treatment(600 mg/kg)on offspring was shown by postnatal growth observation,and negative geotaxis,olfactory discrimination and Morris water maze(MWM)tests.Then three treatment groups were formed with varying exposure to atomized YYO to explore the effects of YYO on the anxiety,social and cognitive behaviors of the autistic-like offspring through the elevated plus-maze test,three-chamber social test,and MWM test.Finally,the monoamine neurotransmitters,including serotonin,dopamine and their metabolites,in the hippocampus and prefrontal cortex(PFC)of the rats were measured using a high-performance liquid chromatography.Results:Offspring of VPA exposure rats showed autism-like behaviors.In the VPA offspring,mediumdose YYO exposure significantly elevated the time and entries into the open arms in the elevated plusmaze test,while low-dose YYO exposure significantly enhanced the social interaction time with the stranger rat in session 1 of the three-chamber social test.VPA offspring treated with YYO exposure used less time to reach the platform in the navigation test of the MWM test.YYO exposure significantly elevated the metabolism of serotonin and dopamine in the PFC of VPA offspring.Conclusion:YYO exposure showed the effects in alleviating anxiety and improving cognitive and social abilities in the offspring of VPA exposure rats.The role of YYO was related to the regulation of the metabolism of serotonin and dopamine.

Dietary oregano Essential oil supplementation improves intestinal functions and alters gut microbiota in latephase laying hens

Background:Dietary Essential oil(EO)supplementation can exert favorable effects on gut health in broilers.However,it is unknown whether EO could improve intestinal functions,consequently beneficial for egg performance and quality in late-phase laying hens.This study was aimed to investigate the potential effects of EO on production performance,egg quality,intestinal health and ileal microbiota of hens in the late phase of production.A total of 28860-week-old Hy-line Brown laying hens were randomly divided into 4 groups and fed a basal diet(control)or basal diets supplemented with oregano EO at 100,200 and 400 mg/kg(EO100,EO200 and EO400).Results:Dietary EO supplementation resulted in a quadratic decrease(P<0.05)in feed conversion ratio with lower(P<0.05)feed conversion ratio in EO200 group than the control during weeks 9–12 and 1–12 of the trial.Compared to the control,EO addition resulted in higher(P<0.05)eggshell thickness at the end of week.4,8 and 12 and higher(P<0.05)chymotrypsin activity.There was a quadratic elevation(P<0.05)in ileal chymotrypsin and lipase activity,along with a linear increase in villus height to crypt depth ratio.Quadratic declines(P<0.05)in mRNA expression of IL-1β,TNF-α,IFN-γ and TLR-4,concurrent with a linear and quadratic increase(P<0.05)in ZO-1 expression were identified in the ileum with EO addition.These favorable effects were maximized at medium dosage(200 mg/kg)of EO addition and intestinal microbial composition in the control and EO200 groups were assessed.Dietary EO addition increased(P<0.05)the abundances of Burkholderiales,Actinobacteria,Bifidobacteriales,Enterococcaceae and Bacillaceae,whereas decreased Shigella abundance in the ileum.Conclusions:Dietary EO addition could enhance digestive enzyme activity,improve gut morphology,epithelial barrier functions and modulate mucosal immune status by altering microbial composition,thus favoring feed efficiency and eggshell quality of late-phase laying hens.

Objective:Cymbopogon citratus(DC.)Stapf is a medicinal and edible herb that is widely used for the treatment of gastric,nervous and hypertensive disorders.In this study,we investigated the cardioprotective effects and mechanisms of the Essential oil,the main active ingredient of Cymbopogon citratus,on isoproterenol(ISO)-induced cardiomyocyte hypertrophy.Methods:The compositions of Cymbopogon citratus Essential oil(CCEO)were determined by gas chromatography-mass spectrometry.Cardiomyocytes were pretreated with 16.9 μg/L CCEO for 1 h followed by 10 μmol/L ISO for 24 h.Cardiac hypertrophy-related indicators and NLRP3 inflammasome expression were evaluated.Subsequently,transcriptome sequencing(RNA-seq)and target verification were used to further explore the underlying mechanism.Results:Our results showed that the CCEO mainly included citronellal(45.66％),geraniol(23.32％),and citronellol(10.37％).CCEO inhibited ISO-induced increases in cell surface area and protein content,as well as the upregulation of fetal gene expression.Moreover,CCEO inhibited ISO-induced NLRP3 inflammasome expression,as evidenced by decreased lactate dehydrogenase content and downregulated mRNA levels of NLRP3,ASC,CASP1,GSDMD,and IL-1β,as well as reduced protein levels of NLRP3,ASC,pro-caspase-1,caspase-1(p20),GSDMD-FL,GSDMD-N,and pro-IL-1β.The RNA-seq results showed that CCEO inhibited the increase in the mRNA levels of 26 oxidative phosphorylation complex subunits in ISO-treated cardiomyocytes.Our further experiments confirmed that CCEO suppressed ISO-induced upregulation of mf-Nd1,Sdhd,mt-Cytb,Uqcrq,and mt-Atp6 but had no obvious effects on mt-Co1 expression.Conclusion:CCEO inhibits ISO-induced cardiomyocyte hypertrophy through the suppression of NLRP3 inflammasome expression and the regulation of several oxidative phosphorylation complex subunits.

Dietary Litsea cubeba Essential oil supplementation improves growth performance and intestinal health of weaned piglets

This paper was to determine the effects of dietary Litsea cubeba Essential oil(LEO)supplementation on growth performance,immune function,antioxidant level,intestinal morphology and microbial composition in weaned piglets.One hundred and ninety-two piglets(Duroc×[Large White×Landrace])with 6.85±0.22 kg mean body weight weaned at 21 d of age were randomly assigned to 4 treatment groups with 8 replicates and were fed with a basal diet(CON)or CON diet containing 100(LLEO),200(MLEO)and 400(HLEO)mg/kg LEO.The results revealed that HLEO supplementation(P<0.05)increased the average daily gain on d 28 compared with CON.MLEO and HLEO supplementation decreased(P<0.05)feed conversion ratio.LEO-containing diets had a lower(P<0.05)diarrhea rate.Supple-mentation with HLEO increased(P<0.05)total antioxidant capacity(T-AOC)both in the serum and liver.Meanwhile,the supplementation of MLEO and HLEO resulted in higher(P<0.05)glutathione peroxidase(GPx)activities both in serum and liver.Supplementation of HLEO increased(P<0.05)serum immu-noglobulin A,immunoglobulin G and interleukin-10,whereas supplementation with MLEO and HLEO decreased(P<0.05)tumor necrosis factor-α.Villus height in the duodenum or jejunum was increased(P<0.05)in the HLEO group,and the villus height to crypt depth ratio in the jejunum was also improved(P<0.05)in the MLEO group.The addition of LEO increased(P<0.05)the richness and diversity of the microbial community in the cecum,which mainly increased the relative abundance of Oscillospir-aceae_UCG-005,Faecalibacterium,Blautia and Coprococcus.Piglets supplemented with HLEO increased(P<0.05)the concentration of short chain fatty acids(SCFA),including acetic acid in the cecum and propionic acid in the colon.In conclusion,these findings indicated that LEO supplementation improved growth performance and intestinal health in weaned piglets.

Composition analysis and antimicrobial activity of Essential oil from leaves of Laurus nobilis grown in Oman

Traditionally,Laurus nobilis is used for the treatment of earaches and skin rashes as well as rheuma-tism.The most common technique to isolate the oil is hydro-distillation and steam-distillation.The investigation presented in this paper was aimed to unravel the antimicrobial activity and chemical composition of Essential oil in the leaves of L.nobilis grown in Oman.The Essential oil was extracted from the leave of the selected plant species by hydro distillation using Clevenger type apparatus.The antimicrobial assay was done by disc diffusion methods against three bac-terial strains,Staphylococcus aureus,Pseudomonas aeruginosa and Escherichia coli,and one fungal strain:Candida albicans.The isolated Essential oil was analyzed by gas chromatography-mass spectrometry(GC-MS).The percentage yield of the Essential oil was 0.064%(w/w).The essen-tial oil was found actively against the applied bacterial and fungal strains.The highest activity was found against S.aureus and the lowest was against E.coli,and the order was S.aureus>P.aeruginosa>E.coli.The Essential oil was also active against the fungus C.albicans.The range of inhibition zones was 6-14 mm.The results of GC-MS analysis showed(E)-𝛽-caryophyllene to be the major component about 59.62%.Other major components included𝛼-selinene(14.03%),𝛼-humulene(8.65%),𝛽-selinene(4.99%),and𝛼-pinene(4.98%).In addition,several minor com-ponents were found in the Essential oil of L.nobilis.Based on the experimental results,it showed that the plant species that grown in Oman were a potential source of therapeutic agents.In con-clusion,the Essential oil and the plant could be used as natural and potential therapeutic agents to treat earaches,skin rashes,and rheumatism.

Salvia microphylla and Teucrium polium are medicinal and aromatic plants with ethnobotanical uses.The present study was conducted to investigate the chemical composition of Salvia microphylla and Teucrium polium Essential oils,the secondary metabolites,and the biological activities of their infusion and methanolic(MeOH)extracts.Essential oils were extracted by hydrodistillation from shoots Salvia microphylla and Teucrium polium.Phenolic content,antioxidant and antimicrobial activities were determined.Gas chromatography-mass spectrometry(GCMS)results showed the presence of significant qualitative and quantitative variations for the composition of the both Essential oils(EO).Salvia microphylla EO were most complex and present 70 compounds with the major components wereβ-caryophyllene(13.32%),1,8 cineole(11.25%),Cis p-Menthane-3-one(10.74%),andβ-Selinenol(9.71%).Where in the Teucrium polium EO,45 compounds were identified with the important components areβ-cadinene(10%),β-citronellol(8.5%),Carvacrol(7.63%),and Eugenol(7.15%).Obtained results showed that both plant are very rich in secondary metabolites.Extracts isolated from Salvia microphylla presented the highest contents in the phenolic compound than extracted from Teucrium polium.The antioxidant activity data demonstrated that all extracts showed strong antioxidant and radical scavenging activities.Essential oil and methanol extracts presented a potential for antimicrobial activities against all tested microorganisms.The obtained results highlight the potential use of Salvia microphylla and Teucrium polium as possible natural antioxidant substances and sources of bioactive molecules.

Essential oil from Citrus aurantium Alleviates Anxiety of Patients Undergoing Coronary Angiography: A Single-Blind, Randomized Controlled Trial

Objective To determine the effectiveness and safety of Essential oil from Citrus aurantium on anxiety in patients undergoing coronary angiography.Methods A single-blind,randomized controlled trial was conducted in 80 patients experiencing coronary angiography in Imam Ali Hospital in Kermanshah,Iran from April to November in 2016.All patients were randomly divided into intervention and control groups by a random number table,40 cases in each group.The patients in the intervention group inhaled Citrus aurantium Essential oil for 15–20 min about 60 min before angiography.Following the same procedure,distilled water was used instead of Citrus aurantium in the control group.Spielbergers State-Trait Anxiety Inventory(STAI)was filled in and vital signs including systolic blood pressure(SBP),diastolic blood pressure(DBP),respiratory and pulse rate were recorded before and 20 min after the intervention.Adverse reactions after intervention were observed.Results In the intervention group,the mean scores of STAI,SBP,DBP,respiratory and pulse rate were 53.30±10.13,134.82±11.75 mm Hg,84.49±6.99 mm Hg,17.87±1.73 times/min,and 76.48±12.55 beats/min at baseline and significantly decreased to 42.37±10.15,124.49±10.48 mm Hg,79.23±6.62 mm Hg,14.54±1.43 times/min,and 70.03±13.66 beats/min respectively 20 min after intervention(all P0.05).All subjects reported no adverse reactions.Conclusion Inhalation of the Essential oil from Citrus aurantium was effective in reducing anxiety and stress levels in patients undergoing coronary angiography.

Repellent,larvicidal and adulticidal activities of Essential oil from Dai medicinal plant Zingiber cassumunar against Aedes albopictus

Zingiber cassumunar is an important plant used in traditional medicine and as a natural mosquito re-pellent.However,the compounds responsible for the repellent activity of the plant are still unknown.The aim of the study is to identify the components of Z.cassumunar Essential oil that show repellent activity against Aedes albopictus.We also evaluated the larvicidal and adulticidal activities of Z.cassumunar Essential oil against Ae.albopictus.In-cage mosquito repellent experiments showed that Z.cassumunar Essential oil possessed moderate repellent activity with a minimum effective dose(MED)of 0.16±0.01 mg/cm^(20,compared to reference standard N,N-diethyl-3-methylbenzamide(DEET,0.03±0.01 mg/cm^(20).Bioassay-guided fractionation identified the major active compound of Z.cassumunar Essential oil as(-)-terpinen-4-ol(1)(MED:0.19±0 mg/cm^(20).We also found that Z.cassumunar Essential oil showed moderate larvicidal activity against first instar larvae of Ae.albopictus with a LC50(50%lethal concentration)of 44.9 mg/L after 24 h.Fumigation bioassays showed that Z.cassumunar Essential oil exhibits moderate adulticidal activity against Ae.albopictus with a LC50 of 5.44%,while(-)-terpinen-4-ol showed significant adulticidal activity with a LC50 of 2.10%after 24 h.This study verifies that the Z.cassumunar Essential oil has mosquito repellent activity,and that(-)-terpinen-4-ol is mainly responsible for this activity.Furthermore,this study provides scientific support for the folk usage of Z.cassumunar Essential oil as mosquito repellent and indicates that Z.cassumunar Essential oil and(-)-terpinen-4-ol can be used as plant-derived repellents and insecticides for mosquito control.

A further study of the kinetics and optimization of the Essential oil hydrodistillation from lavender flowers

Hydrodistillation has commonly been used to recover Essential oil from various plant materials,including lavender(Lavandula officinalis) flowers.The main objectives of the present study were to model the kinetics of the lavender Essential oil(LEO) hydrodistillation using a phenomenological model,to evaluate the statistical significance of the hydromodule and hydrodistillation time on LEO yield combining a 4~2 full factorial design with the response surface methodology,to model statistically LEO yield by multiple non-linear regression and to determine the optimal process conditions that provided the maximum LEO yield.The fast-Essential oil distillation(washing stage) in the initial period and the slow diffusion stage until the saturation occurring simultaneously were observed,justifying the use of the phenomenological model.With increasing the hydromodule,the saturation LEO yield and the washable fraction of the LEO decreased while the washing and diffusion rate constants increased.Knowledge of the LEO oil yield and the hydrodistillation kinetics is important from the techno-economical point of view.

Effects of lanthanum on the growth and Essential oil components of lavender under osmotic stress

The effects of lanthanum on growth, soluble sugar content, Essential oil contents and quality were studied in lavender plant（Lavandula Angustifolia Mill, Variety 701） under osmotic stress by means of PEG-6000 solution. The results show that osmotic stress could reduce the growth rate of lavender, and increase the content of SSC（soluble sugar concentration） to some extent. Compared with control group,the contents of Essential oil in flowers and leaves respectively increase by 45.6% and 48.3% in the osmotic stress group induced by 15% PEG-6000. However, the presence of lanthanum can make the contents of Essential oil in the flowers and leaves of stressed lavender plants enhance by 19.4% and 18.6%, respectively. Under osmotic stress, the relative contents of four kinds of lavender Essential oil compositions of camphor, linalool, linalyl acetate and lavandulol acetate in the lavender flowers were successively 22.63%,0.61%,25.46%,7.03%,and 20.17%,0.62%,20.72%,10.80%,respectively in the presence and absence of lanthanum. The contents of all four main components of lavender Essential oils meet the requirements of the national standard under osmotic stress in the presence of lanthanum. However, in the absence of lanthanum, the contents of linalyl acetate and lavandulol acetate does not meet the requirements of the national standard under osmotic stress. Moreover, the contents of five components of borneol, camphor,eudesmol, caryophyllene oxide and bicyclic sesquiphellandrene from the Essential oil of lavender leaves are 12.89%, 3.84%, 8.76%, 11.30% and 8.10%, respectively. The total content of above five components accounts for 44.89% of the Essential oil of the lavender leaves. Particularly, the borneol content in leaf Essential oil is up to 12.89%. It is 125.2 times the amount of borneol in the flower Essential oil.
