Developmental regulation of neuronal gene expression by elongator complex protein 1 dosage

Familial dysautonomia(FD), a hereditary sensory and autonomic neuropathy, is caused by a mutation in the elongator complex protein 1(ELP1) gene that leads to a tissue-specific reduction of ELP1 protein. Our work to generate a phenotypic mouse model for FD headed to the discovery that homozygous deletion of the mouse Elp1 gene leads to embryonic lethality prior to mid-gestation. Given that FD is caused by a reduction, not loss, of ELP1, we generated two new mouse models by introducing different copy numbers of the human FD ELP1 transgene into the Elp1 knockout mouse(Elp1) and observed that human ELP1 expression rescues embryonic development in a dose-dependent manner. We then conducted a comprehensive transcriptome analysis in mouse embryos to identify genes and pathways whose expression correlates with the amount of ELP1. We found that ELP1 is essential for the expression of genes responsible for nervous system development. Further, gene length analysis of the differentially expressed genes showed that the loss of Elp1 mainly impacts the expression of long genes and that by gradually restoring elongator, their expression is progressively rescued. Finally, through evaluation of co-expression modules, we identified gene sets with unique expression patterns that depended on ELP1 expression.

Autophagy is responsible for maintaining fundamental cellular homeostasis and is,therefore,essential for diverse development *** study reported that PoElp3,the putative catalytic subunit of elongator complex,is involved in the maintenance of autophagy homeostasis to facilitate asexual development and pathogenicity in the rice blast fungus Pyricularia *** was found that the ΔPoelp3 strains were defective in vegetative growth,conidiation,stress response,and *** mutants exhibited hyper-activated autophagy in the vegetative hyphae under both nutrient-rich and nutrient-deficient *** hyper-activation of autophagy possibly suppressed the production of vegetative hyphae in the ΔPoelp3 ***,the ΔPoelp3 strains were found to be more sensitive to rapamycin during vegetative-and invasive-hyphal growth but have no effect on Target-of-Rapamycin(TOR)signaling *** together,these results demonstrated that PoElp3 is involved in asexual development and pathogenicity by regulating autophagy in the rice blast fungus.

目的：研究elongator复合物对60Coγ射线辐照诱导的肝癌细胞DNA损伤修复的影响,探讨在肝癌发生和发展过程中elongator对肝癌细胞基因组发挥保护作用的分子机制。方法：***-PCR检测肿瘤细胞中内源性elp3和elp4 mRNA的表达量。2.以OTM值为指标,用彗星电泳检测辐照后各组细胞的DNA损伤程度。3.以γ-H2AX为指标,用免疫荧光法和Western blot进一步确认辐照后各组细胞在不同时间点的DNA损伤程度和蛋白表达变化情况。4.流式细胞仪检测辐照后各组细胞的凋亡率。5.实时荧光定量PCR检测各组细胞中RAD50、Mre11、Nbs1、ATM、ATR、 DNAPK-cs、Chk1、Chk2、XRCC6、 BRCA1、GADD45G和CDC25A等损伤修复相关基因的表达水平。6. Western blot检测各组细胞中RAD50、Mre11和XRCC6等损伤修复相关蛋白的表达水平。结果：1. RT-PCR检测结果发现,内源性elp3和elp4 mRNA在不同肿瘤细胞株中均有表达,但相对表达量并没有显著变化。2.彗星电泳检测结果显示,与对照组HepG2/SK-Hep-1细胞组相比较,elongator过表达组中OTM值明显减小,而elongator干扰组中OTM值明显增加,从而说明elongator复合物能够减轻肝癌细胞DNA损伤程度,对保护细胞DNA免受60Coγ射线辐照造成的损伤是非常关键的。3.免疫荧光分析和Western blot的实验结果显示,与对照组HepG2细胞组相比较,elongator过表达组中单个细胞内含γ-H2AX的荧光数随着辐照后时间的推移显著减少,而elongator干扰组中γ-H2AX的荧光数显著增多,这说明表达elongator能够降低DNA损伤程度,并且对DNA损伤具有一定的修复作用,从而验证了彗星电泳的结果。4.流式细胞仪检测结果发现,与对照组HepG2细胞组相比较,elongator过表达组的细胞凋亡率明显减少,而elongator干扰组的凋亡率有所增加,这说明elongator能够降低细胞凋亡率,从而维持基因组的稳定性。5.实时荧光定量PCR和Western blot检测各组细胞中DNA损伤修复相关基因和蛋白的表达情况,根据实验结果推测elongator对DNA损伤的修复作用可能是通过RAD50、 Mre11、Nbs1、DNAPK-cs、XRCC6等因子作用完成的。结论：1. elongator复合物能够减轻由于60Coγ射线辐照引起的肝癌细胞DNA损伤程度,并降低细胞凋亡率,表明elongator对DNA损伤具有一定的修复作用,从而维持基因组的稳定性。2. elongator对DNA损伤的修复作用可能是通过RAD50、Mre11、 Nbsl、 DNAPK-cs、XRCC6等修复相关基因作用完成的。

酵母elongator是一个与延伸中的磷酸化RNAPⅡ作用的六亚基复合物,其中Elp3亚基具有组蛋白乙酰基转移酶(HAT)活性,为复合物行使功能所必需。使Elp3的HAT活性丧失的点突变可引起酵母elp表型。elongator可修饰组蛋白H3和H4尾部,其优先乙酰化位点是H3 K14和H4 K8。近来人elongator复合体也被分离,它也是一个具有HAT活性的不稳定的六亚基复合物,其结构和组成与酵母的elongator相似。目前尚未见对人elongator复合物及其亚基功能的体内研究的报道。本文以酿酒酵母为实验材料,对人elongator的Elp3功能进行了体内实验鉴定。证明了人与酵母的Elp3在功能上是保守的,初步确定了hElp3及其HAT活性在复合物中的功能,了解了其HAT 活性在基因表达调控中的作用。主要结果和结论如下:

ELP3,the catalytic subunit of the elongator complex,is an acetyltransferase and associated with tumor ***,the detail of ELP3 oncogenic function remains largely ***,we found that ELP3 stabilizes c-Myc to promote tumorigenesis in an acetyltransferase-independent ***,ELP3 competes with the E3-ligase FBXW7βfor c-Myc binding,resulting in the inhibition of FBXW7β-mediated ubiquitination and proteasomal degradation of ***3 knockdown diminishes glycolysis and glutaminolysis and dramatically retards cell proliferation and xenograft growth by downregulating c-Myc,and such effects are rescued by the reconstitution of c-Myc ***,ELP3 and c-Myc were found overexpressed with a positive correlation in colorectal cancer and hepatocellular *** together,we elucidate a new function of ELP3 in promoting tumorigenesis by stabilizing c-Myc,suggesting that inhibition of ELP3 is a potential strategy for treating c-Myc-driven carcinomas.
