eimeria intestinalis is one of the most pathogenic species of coccidia which causes coccidiosis in rabbits.Live oocyst vaccines with attenuated E.intestinalis are attractive for the control of this wide-spread disease.In this study,we developed a precocious line(EiP132) of E.intestinalis after eight successively passaging and collecting of the first appeared oocysts of parental strain,ZJK strain,in the feces of infected rabbits.The prepatent time of EiP132 was reduced from 204 h to 132 h.For the pathogenicity and immunogenicity tests,results showed that inoculation with 1×10 and 1×10 oocysts did not cause loss of body weight gain or clinical syndrome.On the contrary,infection with 1×10 original strain caused rabbits body weight gain loss(P<0.05) and slight diarrhea The oocyst output of animals inoculated with EiP132 oocysts was decreased by about 99.9%(P<0.01).After challenge with 1×10 original strain 14 dpi,the body weight gain of rabbits immunized with original or precocious strain previously was similar with those of the control,and was higher than those of the unimmunized and challenged rabbits(P<0.01);while the oocyst output was lower than those of the unimmunized and challenged rabbits(P<0.01).These results demonstrated that the precocious line EiP132 is low pathogenic but with excellent immunogenicity.

Objective: eimeria intestinalis is one of the most pathogenic species of rabbit coccidia. For the development of an anticoccidial vaccine, a precocious line of E. intestinalis(PEI) was selected from an original E. intestinalis(OEI). To study the endogenous development of the precocious line, whose prepatent period shortened from 204 h to 132 h, the intestine tissue of rabbits infected was sampled and studied. Methods: Twenty-three rabbits were inoculated with 3 million oocysts per animal, 10 rabbits with the precocious line(group A) and 13 with the parental strain(group B). Five rabbits in each group were sacrificed at 24 h, 48 h, 84 h, 96 h, 120 h post inoculation(PI), respectively;while another 2 rabbits in group B were sacrificed at 168 h, 192 h PI, respectively. To detect the accurate peak time of the second generation schizonts, five rabbits in each group were sacrificed 90h-105 h PI, respectively. Intestine tissue(duodenum, jejunum, ileum) was sampled, processed by routine methods of histology, stained by HE, and observed under light microscope. Results: First generation schizonts were present in the epithelial cells of villi 24 h, 48 h and 84 h PI. Many mature schizonts were observed 84 h PI for the First generation schizonts and about 96 h PI for the second generation schizonts. The peak of the second generation schizonts was detected at 93 h PI for the precocious line and 96 h PI for the parental strain. Macrogamonts were detected 120 h post PEI infection, and they were easily identified by the appearance of wall-forming bodies around the gamonts. Some developing oocysts were noted as well. On the contrary, the third generation schizonts were observed 120 h and 144 h post OEI infection. And a lot of mature schizonts appeared 168 h post OEI infection. Macrogamonts and microgamonts were detected 192 h post OEI infection, 3 days later than that of PEI infection. Recent studies showed that trained immunity,or innate immune memory,demonstrates adaptive characteristics after challenge with pathogens or their products.This new concept leads us to hypothesis that trained immunity plays important role in the protection of rabbits against re-infection with eimeria,which are the causative agents of rabbit coccidiosis threating the rabbit industry.To test whether eimeria could stimulate trained immunity in rabbits against homologous and heterologous challenge,two groups of rabbits were individually immunized with 104 E.intestinalis oocysts,then challenged either with 106 E.intestinalis or 106 E.magna oocysts 14 days post immunization.Heparinized blood was sampled each day from day 0 to 9 post immunization(pi) and post challenge(pc).Numbers of white blood cells(WBC),polymorphonuclear neutrophils(PMN) and lymphocytes(LYM) were analyzed.For immunized groups,the numbers of WBC,PMN and LYM increased,and the peak numbers occurred on day 7 to 8 pi with a 2-fold and 1.8-fold higher than those on day 0 pi,respectively.After homologous or heterologous challenge,both WBC and PMN numbers in immunized groups increased with the peak numbers occurred on day 2 and 3 post challenge,respectively;while the LYM numbers increased from day 6 post challenge.Together,the results suggest that immunization with E.intestinalis could induce trained immunity against homologous and heterologous challenge.
