drug-resistant varieties of pathogens are now a recognized global threat. Insights into the routes for drug resistance in these pathogens are critical for developing more effective antibacterial drugs. A systems-level analysis of the genes, proteins, and interactions involved is an important step to gaining such insights. This paper discusses some of the computational challenges that must be surmounted to enable such an analysis; viz., unreliability of bacterial interactome maps, paucity of bacterial interactome maps, and identification of pathways to bacterial drug resistance.

Prostate cancer(PCa)is a leading cause of cancer-related morbidity and mortality in men *** improvements in the diagnosis and treatment of PCa,a significant proportion of patients with high-risk localized disease and all patients with advanced disease at diagnosis will experience progression to metastatic castration-resistant prostate cancer(mCRPC).Multiple drugs are now approved as the standard of care treatments for patients with mCRPC that have been shown to prolong *** the majority of patients will respond initially,primary and secondary resistance to these therapies make mCRPC an incurable *** molecular mechanisms underlie the development of mCRPC,with the androgen receptor(AR)axis being the main driver as well as the key drug *** resistance mechanisms is crucial for discovering novel therapeutic strategies to delay or reverse the progression of the *** this review,we address the diverse mechanisms of drug resistance in *** addition,we shed light on emerging targeted therapies currently being tested in clinical trials with promising potential to overcome mCRPC-drug resistance.

Hyperactive RAS/RAF/MEK/ERK signaling has a well-defined role in cancer *** this pathway results in complete or partial regression of most *** recent years,cancer genomic studies have revealed that genetic alterations that aberrantly activate the RAS/RAF/MEK/ERK signaling mainly occur on RAF or upstream,which motivated the extensive development of RAF inhibitors for cancer ***,the first-generation RAF inhibitors have been approved for treating late-stage cancers with BRAF(V600E)*** these inhibitors have achieved promising outcomes in clinical treatments,their efficacy is abolished by quick-rising drug ***,cancers with hyperactive RAS exhibit intrinsic resistance to these *** resolve these problems,the second-generation RAF inhibitors have been designed and are undergoing clinical ***,we summarize the recent findings from mechanistic studies on RAF inhibitor resistance and discuss the critical issues in the development of next-generation RAF inhibitors with better therapeutic index,which may provide insights for improving targeted cancer therapy with RAF inhibitors.

Glioblastoma(GBM)represents the main form of brain tumors in adults,and one of the most aggressive cancers *** treatment of GBM is a combination of surgery(when possible),chemotherapy(usually Temozolomide,TMZ)and radiotherapy(RT).However,despite this heavy treatment,GBM invariably recur and the median length of survival following diagnosis is 12 to 15 months,with less than 10%of people surviving longer than five *** is extremely resistant to most treatments because of its heterogeneous nature,which is associated with extreme clonal plasticity and the presence of cancer stem cells,refractory to TMZ-and RT-induced cell *** this review,we explore the mechanisms by which cancer cells,and especially GBM,can acquire resistance to *** describe and discuss the concept of persister/tolerant cells that precede and/or accompany the acquisition of ***/tolerant cells are cancer cells that are not eliminated by treatment(s)because of different mechanisms ranging from dormancy/quiescence to *** discuss the possibility of targeting these mechanisms in new therapeutic regimen.

Cancer immunotherapy,a therapeutic approach that inhibits tumors by activating or strengthening anti-tumor immunity,is currently an important clinical strategy for cancer treatment;however,tumors can develop drug resistance to immune surveillance,resulting in poor response rates and low therapeutic *** addition,changes in genes and signaling pathways in tumor cells prevent susceptibility to immunotherapeutic ***,tumors create an immunosuppressive microenvironment via immunosuppressive cells and secrete molecules that hinder immune cell and immune modulator infiltration or induce immune cell *** address these challenges,smart drug delivery systems(SDDSs)have been developed to overcome tumor cell resistance to immunomodulators,restore or boost immune cell activity,and magnify immune *** combat resistance to small molecules and monoclonal antibodies,SDDSs are used to co-deliver numerous therapeutic agents to tumor cells or immunosuppressive cells,thus increasing the drug concentration at the target site and improving ***,we discuss how SDDSs overcome drug resistance during cancer immunotherapy,with a focus on recent SDDS advances in thwarting drug resistance in immunotherapy by combining immunogenic cell death with immunotherapy and reversing the tumor immunosuppressive *** that modulate the interferon signaling pathway and improve the efficacy of cell therapies are also ***,we discuss potential future SDDS perspectives in overcoming drug resistance in cancer *** believe that this review will contribute to the rational design of SDDSs and development of novel techniques to overcome immunotherapy resistance.
