Bushenhuoxue improves cognitive function and activates brain-derived neurotrophic factor-mediated signaling in a rat model of vascular dementia

OBJECTIVE:To explore the protective mechanisms of the Traditional Chinese Medicine Bushenhuoxue(BSHX)in a rat model of vascular dementia(VD).METHODS:A rat model of VD was developed using bilateral common carotid artery occlusion(BCCAO).Rats were administered BSHX(10.14 or 5.07 g/kg),nimodipine(11.06 mg/kg;positive control),or saline(control)by gavage daily for 30 d *** and memory abilities were assessed using the Morris water *** changes in the hippocampus were observed using light microscopy(hematoxylin and eosin staining)and transmission electron microscopy(TEM).The m RNA and protein expression levels of brain-derived neurotrophic factor(BDNF),tyrosine receptor kinase B(Trk B),phosphatidyl inositol 3-kinase(PI3 K),serine/threonine kinase(AKT),and c AMP response element binding protein(CREB)were measured by real-time polymerase chain reaction(RT-PCR)and Western blot,***:Compared with the sham group,rats with BCCAO exhibited impaired learning and memory abilities(Morris water maze)and showed abnormalities in neuronal morphology(light microscopy)and ultrastructure(TEM)in the *** also had decreased m RNA and protein expressions of BDNF,Trk B,PI3 K,AKT,and CREB in hippocampal tissue(all P<0.05).In rats with BCCAO,administration of BSHX attenuated deficits in learning and memory,improved the morphology and ultrastructure of hippocampal neurons,and enhanced m RNA and protein expression levels of BDNF,Trk B,PI3 K,AKT,and CREB(all P<0.05).CONCLUSION:BSHX may protect hippocampal neurons and improve learning and memory abilities,at least in part via the activation of BDNF/Trk B/PI3 K/AKT/CREB signaling.

Efficacy of tanshinone ⅡA and mesenchymal stem cell treatment of learning and memory impairment in a rat model of vascular dementia

OBJECTIVE: To investigate the efficacy of administration of tanshinone Ⅱ A(TSA) combined with mesenchymal stem cells(MSCs) for the treatment of learning and memory impairment caused by vascular dementia(Va D) and to determine the underlying ***: Modified four-vessel occlusion was used to establish a Va D model in rats, and their spatial learning and memory capacity was assessed by the Morris water maze. The rats were randomized into MSCs, TSA, MSCs combined with TSA, vehicle and sham groups. Histological changes were determined by hematoxylin and eosin staining, and the hippocampal neuron apoptosis ratio was assessed by flow cytometry. Western blotting was performed to detect Bcl-2 and Bax expression. The reactive oxidative species(ROS) levels and the activity of total superoxide dismutase(T-SOD), an antioxidant enzyme in the rat hippocampus, were ***: TSA combined with MSCs treatment administered by intravenous injection in the tail significantly attenuated cognitive deficits in the Va D model compared with the vehicle group(P < 0.01),and its protective effect on cognitive function was greater than that obtained by treatment with MSCs or TSA alone. Furthermore, TSA combined with MSCs treatment achieved synergistic effects in suppressing neuronal apoptosis in the rat hippocampus caused by global brain ischemia via up-regulating the expression of Bcl-2, an anti-apoptosis protein, and decreasing the expression of Bax, a pro-apoptotic protein. In addition, TSA combined with MSCs treatment attenuated ROS production and enhanced T-SOD activity in the rat hippocampus, and the antioxidant effect was greater than that of treatment with MSCs or TSA ***: TSA combined with MSCs treatment improved the spatial learning and memory capacity in a Va D model via suppressing neuronal apoptosis and antioxidant activity in the hippocampus, and this improvement was greater with combined treatment than with treatment with MSCs or TSA alone.

Tongluo Huatan capsule(通络化痰胶囊) improves cognitive function by regulating the endocytosis of N-methyl-D-aspartic acid receptors mediated by clathrin in a rat model of vascular dementia

OBJECTIVE:To explore the neuroprotective mechanisms of Tongluo Huatan capsule(THC)in a rat model of vascular dementia(VD).METHODS:A rat model of VD was established by repeated clamping of bilateral common carotid arteries with the intraperitoneal injection of sodium nitroprusside *** rats were administered THC,memantine hydrochloride,or distilled water daily for 14 d after *** and memory abilities were assessed using the step-down passive avoidance test,novel object recognition(NOR)test,and Morris water maze(MWM)*** changes in the hippocampus were observed through hematoxylin and eosin and Nissl *** expression levels of clathrin,RAB5 B,andN-methyl-D-aspartic acid receptor 1(NMDAR1)were measured by immunohistochemistry staining,real-time quantitative polymerase chain reaction and Western ***:Rats in VD group showed impaired learning and memory abilities(step-down passive avoidance,NOR,and MWM)and abnormalities in neuronal morphology(light microscopy)in the *** m RNA or protein expression levels of clathrin and RAB5 B were decreased,and NMDAR1 was increased in hippocampal tissues(P<0.05).Administration of THC promoted the learning and memory abilities and the morphological structure of hippocampal neurons in VD ***,THC enhanced m RNA or protein expression levels of clathrin and RAB5 B,and decreased NMDAR1(P<0.05).CONCLUSION:THC may improve cognitive functions by regulating the endocytosis of NMDA receptors mediated by clathrin.

Influence of Tuina plus oxiracetam on serum inflammatory factors and oxidative stress in mild vascular dementia patients

Objective To observe the clinical efficacy of Tuina(Chinese therapeutic massage)plus oxiracetam in treating mild vascular dementia(VD)and seek its underlying *** Ninety-six patients with mild VD were randomized into an observation group and a control group,with 47 cases in the observation group and 49 cases in the control *** control group received oral oxiracetam capsules for treatment,and the observation group was given additional Tuina *** and after treatment,the mini-mental state examination(MMSE)was adopted to assess the patient’s cognitive function;the activities of daily living(ADL)scale was used to evaluate their ability to conduct daily activities;changes in the serum inflammatory factors and oxidative stress indicators were also *** After treatment,the serum content of malondialdehyde(MDA)decreased in both groups(P<0.05)and was lower in the observation group than in the control group(P<0.05);the serum contents of superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)increased in both groups(P<0.05)and were higher in the observation group than in the control group(P<0.05);the serum contents of interleukin(IL)-1,tumor necrosis factor(TNF)-α,IL-6,and IL-8 declined in both groups(P<0.05)and were lower in the observation group than in the control group(P<0.05).After the intervention,the levels of systolic velocity(Vs)and mean velocity(Vm)of the middle cerebral artery elevated,and the pulsatility index(PI)dropped in patients in the two groups,showing significant intra-group differences(P<0.05);the levels of Vs and Vm in the observation group were higher than those in the control group,and the PI was lower in the observation group than in the control group,showing significant between-group differences(P<0.05).The MMSE and ADL scores increased in both groups after the intervention(P<0.05)and were higher in the observation group than in the control group(P<0.05).Conclusion In the treatment of mild VD,Tuina plus oxi

Effects of electroacupuncture of different frequencies on free radicals in hippocampus of mice with vascular dementia

Objective:To observe the effects of electroacupuncture (EA) of three different frequencies (2 Hz,80 Hz and 2 Hz/80 Hz) on the free radicals in hippocampus of vascular dementia (VD) model ***:A total of 100 Kunming mice were randomly divided into a sham operation group,a model group,a 2 Hz EA group,an 80 Hz EA group and a 2 Hz/80 Hz EA group,with 20 mice in each *** ischemia-reperfusion VD model was established by repeated blockade of bilateral common carotid *** in EA groups began EA treatment on the 4th day after the *** (GV 20),Dazhui (GV 14),Geshu (BL 17) and Zusanli (ST 36) were punctured and then connected to EA instrument,with different waves of 2 Hz,80 Hz or 2 Hz/80 Hz (10 min/time) applied accordingly,once a *** the jumping stand experiment,the learning performance,memory performance and hippocampal calcitonin gene-related peptide (CGRP),nitric oxide synthase (NOS),malondialdehyde (MDA),changes in superoxide dismutase (SOD) and true choline esterase (TChE) were *** hippocampus,the CGRP level was determined by radioimmunoassay;the MDA level was determined by thiobarbituric acid colorimetric method;the activities of NOS and TChE were determined by spectrophotometry;the activity of SOD was determined by xanthine oxidase ***:Compared with the sham operation group,the performances of learning and memory decreased significantly in the model group (P＜0.01);in hippocampus,the CGRP level decreased,the MDA level increased,the activities of NOS and TChE increased,and the activity of SOD decreased in the model *** with the model group,the learning and memory performances of the EA groups were significantly improved (P＜0.05 or P＜0.01);in hippocampus,the CGRP level increased,the MDA level decreased,the NOS and TChE activities decreased,and the SOD activity increased (P＜0.05 or P＜0.01).Among EA groups,the 2 Hz/80 Hz EA group was superior to the 2 Hz EA group and the 80 Hz EA group (P＜0.05 or P＜0.01)

Objective：To observe the effect of electroacupuncture on learning and memory in rats with vascular ***：Seventy-two SD rats were randomly divided into 4 groups,a normal group,a sham operation group,a model group and an electroacupuncture （EA） pretreatment *** intraluminal embolism method with nylon thread was applied to make rat model of vascular dementia,and the ability of learning and memory was observed in Morris water ***：The average time of escape and latency in the rats of model group in the water maze test was prolonged,with significant difference compared with that in the sham group （P〈0.01）.The average time of escape and latency and navigation test results of rats pretreated by EA had statistically significant difference compared with those of the model group （P〈0.01）.Conclusion：EA pretreatment can significantly improve the ability of learning and memory of the rats with vascular dementia.
